Project description:BackgroundThe deep-sea biome (> 200 m depth) is the world's last great wilderness, covering more than 65% of the earth's surface. Due to rapid technological advances, deep-sea environments are becoming more accessible to scientific research and ocean exploration around the world and, in recent years, this is also true for the Galapagos Islands. Deep-sea habitats cover the largest proportion of Galapagos Marine Reserve (GMR), yet to date, no comprehensive baseline exists on the biodiversity of the benthic fauna associated with volcanic seafloor formations within this region. Closing this knowledge gap is essential to provide information for decision-making for the management of marine resources within the GMR and assessing any potential changes in biodiversity resulting from climate-driven alterations that deep-sea environments are expected to experience. In 2015, the Charles Darwin Foundation's Seamounts of the GMR Research Project, together with the Galapagos National Park Directorate (GNPD) and Ocean Exploration Trust (OET), conducted a joint expedition on board the EV Nautilus. Using Remotely operated vehicles (ROVs), the aim of the expedition was to characterise the geological formations and biological communities present on seamounts, lava flows and other deep-sea habitats (> 200 m) within the GMR.New informationWe provide the first comprehensive image inventory for the phylum Arthropoda from 260 to 3400 m of depth within the GMR. Past studies on deep-sea macroinvertebrates in the GMR have been limited to voucher samples collected from dredging (restricted to soft bottom environments) or by submersibles (only allowing limited biological sampling). The image inventory, presented here, is based on high-definition video transects conducted by the Hercules ROV on board the EV Nautilus. Images of macroinvertebrate morphospecies were captured, catalogued and identified, thus providing the first known image inventory of in-situ macroinvertebrate species from the deep-sea region of the GMR.We present 32 distinct morphospecies occurrences within the class Malacostraca and order Decapoda. We also report 17 different families, three species that are new records to the GMR, in-situ images of two new species to science recently described and one possible new squat lobster, as well as interesting behavioural observations.

Project description:Long-distance dispersal is believed to strongly influence coral reef population dynamics across the Tropical Pacific. However, the spatial scale and strength at which populations are potentially connected by dispersal remains uncertain. To determine the patterns in connectivity between the Eastern (ETP) and Central Tropical Pacific (CTP) ecoregions, we used a biophysical model incorporating ocean currents and larval biology to quantify the seascape-wide dispersal potential among all population. We quantified the likelihood and determined the oceanographic conditions that enable the dispersal of coral larvae across the Eastern Pacific Barrier (EP-Barrier) and identified the main connectivity pathways and their conservation value for dominant reef-building corals. Overall, we found that coral assemblages within the CTP and ETP are weakly connected through dispersal. Although the EP-Barrier isolates the ETP from the CTP ecoregion, we found evidence that the EP-Barrier may be breached, in both directions, by rare dispersal events. These rare events could explain the evolutionary genetic similarity among populations of pocilloporids in the ecoregions. Moreover, the ETP may function as a stronger source rather than a destination, providing potential recruits to CTP populations. We also show evidence for a connectivity loop in the ETP, which may positively influence long-term population persistence in the region. Coral conservation and management communities should consider eight-key stepping stone ecoregions when developing strategies to preserve the long-distance connectivity potential across the ETP and CTP.

Project description:Triplets of Lactobacullus plantarum strains were isolated from nine contrasting habitats. Without any passage through other culture media, isolation and cultivation were on model media that strictly reproduced the chemical and physical conditions and stressors of the habitats of origin. Here, we demonstrated how L. plantarum regulates and shapes its transcriptome in response to contrasting habitats. Firstly, multivariate clustering analysis of transcriptional data (RNA-Seq), complemented with metabolomics and phenomics, grouped the strains according to the habitats of origin. Subsequently, selected strains from each habitat switched to repeated cultivation on MRS medium and transcriptomes homogenized into a unique cluster. Adaptation to this common medium mainly relied on activation of genes for phage- and prophage-related proteins and transposases. Finally, the comparison of growth across model media and with respect to MRS medium showed that 44% of the overall 3,112 gene transcripts changed depending on the specific habitat. Regulation and shaping of transcriptomes mainly concerned carbohydrate acquisition, pyruvate catabolism, proteolytic system and amino acid, lipid and inorganic ion transport and metabolism, with contrasting responses for contrasting habitats. Pathways reconstruction demonstrated how the large genome size of L. plantarum imparts transcriptome and metabolic flexibility as the basic mechanism for a nomadic lifestyle.

Project description:Despite a large occurrence, especially over the Pacific Ocean, the chemical diversity of marine invertebrates belonging to the order Zoantharia is largely underexplored. For the two species of the genus Antipathozoanthus no chemical study has been reported so far. The first chemical investigation of Antipathozoanthus hickmani collected at the Marine Protected Area "El Pelado", Santa Elena, Ecuador, led to the isolation of four new ecdysteroid derivatives named ecdysonelactones. The structures of ecdysonelactones A-D (1-4) were determined based on their spectroscopy data, including 1D and 2D NMR and HRMS. The four compounds of this family of ecdysteroids feature an unprecedented ?-lactone fused at the C-2/C-3 position of ring A. These derivatives exhibited neither antimicrobial nor cytotoxic activities.

Project description:We examined rates of N2 fixation from the surface to 2000 m depth in the Eastern Tropical South Pacific (ETSP) during El Niño (2010) and La Niña (2011). Replicated vertical profiles performed under oxygen-free conditions show that N2 fixation takes place both in euphotic and aphotic waters, with rates reaching 155 to 509 µmol N m(-2) d(-1) in 2010 and 24±14 to 118±87 µmol N m(-2) d(-1) in 2011. In the aphotic layers, volumetric N2 fixation rates were relatively low (<1.00 nmol N L(-1) d(-1)), but when integrated over the whole aphotic layer, they accounted for 87-90% of total rates (euphotic+aphotic) for the two cruises. Phylogenetic studies performed in microcosms experiments confirm the presence of diazotrophs in the deep waters of the Oxygen Minimum Zone (OMZ), which were comprised of non-cyanobacterial diazotrophs affiliated with nifH clusters 1K (predominantly comprised of α-proteobacteria), 1G (predominantly comprised of γ-proteobacteria), and 3 (sulfate reducing genera of the δ-proteobacteria and Clostridium spp., Vibrio spp.). Organic and inorganic nutrient addition bioassays revealed that amino acids significantly stimulated N2 fixation in the core of the OMZ at all stations tested and as did simple carbohydrates at stations located nearest the coast of Peru/Chile. The episodic supply of these substrates from upper layers are hypothesized to explain the observed variability of N2 fixation in the ETSP.

Project description:The eastern North Pacific (ENP) has the highest density of tropical cyclones (TCs) on earth, and yet the controls on TCs, from individual events to seasonal totals, remain poorly understood. One effect that has not been fully considered is the unique geography of the Central American mountains. Although observational studies suggest these mountains can readily fuel individual TCs through dynamical processes, here we show that these mountains indeed play the opposite role on the seasonal timescale, hindering seasonal ENP TC activity by up to 35%. We found that these mountains significantly interrupt the abundant moisture transport from the Caribbean Sea to the ENP, limiting deep convection over the open ocean area where TCs preferentially occur. This study advances our fundamental understanding of ENP TC genesis mechanisms across the weather-to-climate timescales, and also highlights the importance of topography representation in improving the ENP regional climate simulations, as well as TC seasonal predictions and future projections.

Project description:Mechanisms by which tropical Pacific and Indian Ocean sea surface temperatures (SST) influence vegetation in eastern Africa have not been fully explored. Here, we use a suite of idealized Earth system model simulations to elucidate the governing processes for eastern African interannual vegetation changes. Our analysis focuses on Tanzania. In the absence of ENSO-induced sea surface temperature anomalies in the Tropical Indian Ocean (TIO), El Niño causes during its peak phase negative precipitation anomalies over Tanzania due to a weakening of the tropical-wide Walker circulation and anomalous descending motion over the Indian Ocean and southeastern Africa. Resulting drought conditions increase the occurrence of wildfires, which leads to a marked decrease in vegetation cover. Subsequent wetter La Niña conditions in boreal winter reverse the phase in vegetation anomalies, causing a gradual 1-year-long recovery phase. The 2-year-long vegetation decline in Tanzania during an ENSO cycle can be explained as a double-integration of the local rainfall anomalies, which originate from the seasonally-modulated ENSO Pacific-SST forcing (Combination mode). In the presence of interannual TIO SST forcing, the southeast African precipitation and vegetation responses to ENSO are muted due to Indian Ocean warming and the resulting anomalous upward motion in the atmosphere.

Project description:Changes in chromium (Cr) isotope ratios due to fractionation between trivalent [Cr(III)] and hexavalent [Cr(VI)] are being utilized by geologists to infer oxygen conditions in past environments. However, there is little information available on Cr in the modern ocean to ground-truth these inferences. Transformations between the two chromium species are important processes in oceanic Cr cycling. Here we present profiles of hexavalent and trivalent Cr concentrations and stable isotope ratios from the eastern tropical North Pacific (ETNP) oxygen-deficient zone (ODZ) which support theoretical and experimental studies that predict that lighter Cr is preferentially reduced in low-oxygen environments and that residual dissolved Cr becomes heavier due to removal of particle-reactive Cr(III) on sinking particles. The Cr(III) maximum dominantly occurs in the upper portion of the ODZ, implying that microbial activity (dependent on the sinking flux of organic matter) may be the dominant mechanism for this transformation, rather than a simple inorganic chemical conversion between the species depending on the redox potential.
