Project description:Different populations of the same species survive different environments through local adaptation. Temperature is one of the most important driving forces that could result in local adaptation. Here, we studied the influence of extreme low temperature on the survival of two genetically and geographically distinct populations of the free-living Caenorhabditis briggsae. We found that Caenorhabditis briggsae strains of temperate origin had a cold resistant phenotype, while those originating from a tropical climate had reduced survival after cold treatment. Using this phenotypic difference between geographically diverse populations as a model for how species adapt to their local environment, we then analyzed the transcriptional profiles of two Caenorhabditis briggsae strains of tropical and temperate origin to find genes that are involved in survival after extreme cold. In summary, the response to the extreme low temperature that clearly distinguishes the temperate and tropical Caenorhabditis briggsae strains could serve as an excellent example for studying local adaption of species that show genetic separation associated with their geographical distribution.

Project description:We designed a new specific mRNA microarray targeting a subset of genes (956) of the diazotrophs Richelia intracellularis and Calothrix rhizosoleniae (genomes RintRC01, RintHH01, RintHM01 and CalSC01) which associate with diatom hosts. The aim was to be able to describe the gene expressions of genes related to several metabolic pathways, specifically nitrogen fixation and how they possibly differed between the closely related strains based on environment and host association. In this study we focused on the RintRC01 and RintHH01. To better understand how the gene expression of nitrogen fixation genes relates to nitrogen fixation rates both gene expression and rate measurements were done in parallell.

Project description:Observational analysis suggests that the western tropical Pacific (WTP) sea surface temperature (SST) shows predominant variability over multidecadal time scales, which is unlikely to be explained by the Interdecadal Pacific Oscillation. Here we show that this variability is largely explained by the remote Atlantic multidecadal oscillation (AMO). A suite of Atlantic Pacemaker experiments successfully reproduces the WTP multidecadal variability and the AMO-WTP SST connection. The AMO warm SST anomaly generates an atmospheric teleconnection to the North Pacific, which weakens the Aleutian low and subtropical North Pacific westerlies. The wind changes induce a subtropical North Pacific SST warming through wind-evaporation-SST effect, and in response to this warming, the surface winds converge towards the subtropical North Pacific from the tropics, leading to anomalous cyclonic circulation and low pressure over the WTP region. The warm SST anomaly further develops due to the SST-sea level pressure-cloud-longwave radiation positive feedback. Our findings suggest that the Atlantic Ocean acts as a key pacemaker for the western Pacific decadal climate variability.

Project description:The dispersal of marine organisms can be restricted by a set of isolation mechanisms including hard barriers or hydrological features. In the Western Atlantic Ocean, the Amazon River discharge has been shown to act as a biogeographical barrier responsible for the differences in reef fish communities between Caribbean Sea and Northeast Brazil continental shelves. Here, we compare the diversity of all Animalia phyla from biogeographic ecoregions along the Tropical Western Atlantic continental shelf to test the hypothesis that the Amazon River plume spatially structures species diversity. For that, we used beta diversity estimators and multivariate ecological analysis on a database of species occurrence of the whole animal kingdom including 175,477 occurrences of 8,375 species from six ecoregions along the Western Tropical Atlantic. Results of the whole animal kingdom and the richest phyla showed that the Caribbean Sea and Tropical Brazil ecoregions are isolated by the Amazon River Plume, broadening and confirming the hypothesis that it acts as a soft barrier to animal dispersal in the Western Tropical Atlantic. Species sharing is larger northwestwards, in direction of the Caribbean than the opposite direction. Beyond species isolation due to local characteristics such as low salinity and high turbidity, our results suggest the dominant northwestward currents probably play a major role in animal dispersion: it enhances the flux of larvae and other planktonic organisms with reduced mobility from Brazil to Caribbean and hinders their contrary movement. Thus, the Amazon area is a strong barrier for taxa with reduced dispersal capacity, while species of pelagic taxa with active swimming may transpose it more easily.

Project description:Six species of Serpula and Spiraserpula were identified, mainly, from the material of the expeditions of the Rosenstiel School of Marine and Atmospheric Science, University of Miami, including two new species of Serpula. Serpula madrigalaesp. n. from the Turks and Caicos has a tube with five longitudinal ridges, four rows of alveoli and a medium-sized shallow symmetrical opercular funnel with 17 radii, and an inner surface with opercular tubercles. Serpula vossaesp. n. from the Western Caribbean and Bahamas has a tube with 6-8 longitudinal ridges, and a large, deep symmetrical opercular funnel, with 21-33 radii, and a smooth inner surface. Serpula cf. vermicularis, recorded from the Gulf of Guinea (tropical eastern Atlantic), is distinguished from the nominal species in possessing fewer opercular radii (33-39) and the lack of a proximal rasp in the bayonet chaetae; tubes are missing. The distribution range is extended for the three known Spiraserpula species found in the collections, Spiraserpula caribensis, Spiraserpula karpatensis and Spiraserpula ypsilon.

Project description:Diazotrophic cyanobacteria, those capable of fixing di-nitrogen (N2), are considered one of the major sources of new nitrogen (N) in the oligotrophic tropical ocean, but direct incorporation of diazotrophic N into food webs has not been fully examined. In the Amazon River-influenced western tropical North Atlantic (WTNA), diatom diazotroph associations (DDAs) and the filamentous colonial diazotrophs Trichodesmium have seasonally high abundances. We sampled epipelagic mesozooplankton in the Amazon River plume and WTNA in May-June 2010 to investigate direct grazing by mesozooplankton on two DDA populations: Richelia associated with Rhizosolenia diatoms (het-1) and Hemiaulus diatoms (het-2), and on Trichodesmium using highly specific qPCR assays targeting nitrogenase genes (nifH). Both DDAs and Trichodesmium occurred in zooplankton gut contents, with higher detection of het-2 predominantly in calanoid copepods (2.33-16.76 nifH copies organism-1). Abundance of Trichodesmium was low (2.21-4.03 nifH copies organism-1), but they were consistently detected at high salinity stations (>35) in calanoid copepods. This suggests direct grazing on DDAs, Trichodesmium filaments and colonies, or consumption as part of sinking aggregates, is common. In parallel with the qPCR approach, a next generation sequencing analysis of 16S rRNA genes identified that cyanobacterial assemblage associated with zooplankton guts was dominated by the non-diazotrophic unicellular phylotypes Synechococcus (56%) and Prochlorococcus (26%). However, in two separate calanoid copepod samples, two unicellular diazotrophs Candidatus Atelocyanobacterium thalassa (UCYN-A) and Crocosphaera watsonii (UCYN-B) were present, respectively, as a small component of cyanobacterial assemblages (<2%). This study represents the first evidence of consumption of DDAs, Trichodesmium, and unicellular cyanobacteria by calanoid copepods in an area of the WTNA known for high carbon export. These diazotroph populations are quantitatively important in the global N budget, widespread and hence, the next step is to accurately quantify grazing. Nonetheless, these results highlight a direct pathway of diazotrophic N into the food web and have important implications for biogeochemical cycles, particularly oligotrophic regions where N2 fixation is the main source of new nitrogen.
