Project description:Characterization of the transcriptomic responses of grafted tomato seedlings leaves after the root inoculations with the two beneficial microorganisms Paraburkholderia graminis and Azospirillum brasiliensis. Paraburkholderia graminis treatment led to a higher number of differentially expressed genes than Azospirillum brasiliensis, with a higher amount of up-regulated than down-regulated genes for both treatments. These DEGs were manly involved in response to oxidative stress, response to biotic and abiotic stress, water transport, regulation of transcription and hormones. Only few DEGs were shared among the two treatments, including genes involved in flowering time and in tolerance against abiotic stresses.

Negotiation of mutualism: rhizobia and legumes.

Project description:The evolution and persistence of biological cooperation have been an important puzzle in evolutionary theory. Here, we suggest a new approach based on bargaining theory to tackle the question. We present a mechanistic model for negotiation of benefits between a nitrogen-fixing nodule and a legume plant. To that end, we first derive growth rates for the nodule and plant from metabolic models of each as a function of material fluxes between them. We use these growth rates as pay-off functions in the negotiation process, which is analogous to collective bargaining between a firm and a workers' union. Our model predicts that negotiations lead to the Nash bargaining solution, maximizing the product of players' pay-offs. This work introduces elements of cooperative game theory into the field of mutualistic interactions. In the discussion of the paper, we argue for the benefits of such an approach in studying the question of biological cooperation.

How legumes control nitrogen fixation by root nodule bacteria

Project description:Rhizobia are soil bacteria that can enter into complex symbiotic relationships with legumes, where rhizobia induce the formation of nodules on the plant root. Inside nodules, rhizobia differentiate into nitrogen-fixing bacteroids that reduce atmospheric nitrogen into ammonia, secreting it to the plant host in exchange for carbon. During the transition from free-living bacteria to bacteroids, rhizobial metabolism undergoes major changes. To investigate the metabolism of bacteroids and contrast it with the free-living state, we quantified the proteome of unlabelled bacteroids relative to 15N-labelled free-living rhizobia. The data were used to build a core metabolic model of pea bacteroids for the strain Rhizobium leguminosarum bv. viciae 3841.
