Immunomodulatory Potential of Dapphitkhai and Chanthaleela Thai Traditional Recipes in the Context of COVID-19 Pathogenesis

Project description:This study aimed to investigate the effects of two Thai traditional recipes including Dapphitkhai and Chanthaleela on various immune parameters and signaling pathways in human white blood cells. The gene expression of whole blood stimulation was analyzed through RNA-sequencing and pathway analysis. Chanthaleela and Dapphitkhai demonstrated a significant decrease in IL-8 secretion and an increase in IFN-β secretion, suggesting their potential to modulate these signaling pathways. Gene expression analysis of Chanthaleela and Dapphitkhai at 500 µg/mL revealed distinct patterns and identified differentially expressed genes. Gene ontology analysis highlighted the impact of the recipes on immune responses and cytokine regulation, while pathway analysis identified key pathways related to cytokine-cytokine receptor interaction, chemokine signaling, and infection-related processes. This study provides valuable insights into the effects of Thai traditional recipes on immune parameters and signaling pathways in human white blood cells. The findings suggest potential therapeutic applications of Thai traditional medicine in immune-related disorders and viral infections like COVID-19.

Assessment of fish biodiversity in four Korean rivers using environmental DNA metabarcoding.

Project description:Environmental DNA (eDNA) metabarcoding is a cost-effective novel approach to estimate biodiversity in an ecosystem. In this study, the MiFish pipeline was employed to test if the system methodology is sufficiently reliable to estimate fish biodiversity in Korean rivers. A total of 125 unique haplotypes and 73 species were identified at the species level from 16 water samples collected from a single survey in four Korean rivers (Hyeongsan, Taehwa, Seomjin, and Nakdong). Among the four rivers, the highest species richness was recorded in the Seomjin River (52 species), followed by the Taehwa (42 species) and Hyeongsan (40 species) rivers. The Nakdong River (26 species) presented the lowest species richness and number of endemic species, presumably due to its metropolitan location and anthropogenic impacts, such as dams or weirs. We were also able to detect that five exotic species (Carassius cuvieri, Cyprinus carpio, Cyprinus megalophthalmus, Lepomis macrochirus, and Micropterus salmoides) are widely distributed in all surveyed rivers, a situation that might be problematic in terms of conservation. Our findings indicate that the eDNA metabarcoding technique is one of the most cost-effective scientific tools available for the management and conservation of the freshwater fish resources available in Korea. However, the low number of 12S sequences of endemic species in the database and low resolution of the MiFish region for differentiating several taxa should be upgraded for their wide use.

Environmental DNA reveals quantitative patterns of fish biodiversity in large rivers despite its downstream transportation.

Project description:Despite the ecological and societal importance of large rivers, fish sampling remains costly and limited to specific habitats (e.g., river banks). Using an eDNA metabarcoding approach, we regularly sampled 500?km of a large river (Rhône River). Comparisons with long-term electrofishing surveys demonstrated the ability of eDNA metabarcoding to qualitatively and quantitatively reveal fish assemblage structures (relative species abundance) but eDNA integrated a larger space than the classical sampling location. Combination of a literature review and field data showed that eDNA behaves in the water column like fine particulate organic matter. Its detection distance varied from a few km in a small stream to more than 100?km in a large river. To our knowledge, our results are the first demonstration of the capacity of eDNA metabarcoding to describe longitudinal fish assemblage patterns in a large river, and metabarcoding appears to be a reliable, cost-effective method for future monitoring.
