A Listeria monocytogenes Bacteriocin Can Target the Commensal Prevotella copri and Modulate Intestinal Infection.

Project description:Understanding the role of the microbiota components in either preventing or favoring enteric infections is critical. Here, we report the discovery of a Listeria bacteriocin, Lmo2776, which limits Listeria intestinal colonization. Oral infection of conventional mice with a ?lmo2776 mutant leads to a thinner intestinal mucus layer and higher Listeria loads both in the intestinal content and deeper tissues compared to WT Listeria. This latter difference is microbiota dependent, as it is not observed in germ-free mice. Strikingly, it is phenocopied by pre-colonization of germ-free mice before Listeria infection with Prevotella copri, an abundant gut-commensal bacteria, but not with the other commensals tested. We further show that Lmo2776 targets P. copri and reduces its abundance. Together, these data unveil a role for P.copri in exacerbating intestinal infection, highlighting that pathogens such as Listeria may selectively deplete microbiota bacterial species to avoid excessive inflammation.

Proteome of Listeria monocytogenes FBUNT biofilm cells and response to bacteriocin AL705

Project description:L. monocytogenes FBUNT is able to form biofilm after six days at 10 ºC. Proteome under planktonic growth, lactocin AL705-treated and untreated biofilms was analyzed. Compared to planktonic cells, protein abundance shifts associated with ribosomes function and biogenesis, cell membrane functionality, carbohydrate and amino acid metabolism and transport were observed in sessile cells. When sessile cells were treated with lactocin AL705, proteins up-regulation were mostly related to carbohydrate metabolism and nutrient transport in an attempt to compensate for impaired energy generation caused by bacteriocin interacting with the cytoplasmic membrane. Notably, transport systems as β-glucosidase IIABC (lmo0027), cellobiose (lmo2763) and trehalose (lmo1255) specific PTS proteins were highly overexpressed.

Project description:The foodborne pathogen Listeria monocytogenes uses a number of transcriptional regulators, including the negative regulator HrcA, to control gene expression under different environmental conditions and in response to stress. Gene expression patterns of DhrcA stationary phase cells were compared to wt to identify hrcA-dependent genes. We identified 61 HrcA-dependent genes that showed significant expression ratios (adj. P < 0.05), with ≥ 1.5-fold differential expression between ΔhrcA and wt. Combined with microarray analysis, Hidden Markov Model searches show HrcA directly repress at least 8 genes. Project description:This study investigated the alternative gene, protein expression and morphological changes of L. monocytogenes 08-5923 following 15min, 4h and 30h exposure to bacteriocin carnocyclin A (cclA). The microarray experiment studied the gene expression of untreated and treated Listeria cells following 4h exposure to cclA. A six chip study using total RNA recovered from six separate cultures of L. monocytogenes 08-5923; three grown in TSB (untreated) and three grown in TSB with cclA (treated); Each chip measures the expression level of 2,857 genes from L. monocytogenes 08-5923 with five 60-mer probe pairs (PM/MM) per gene, with 1-fold technical redundancy.
