Project description:Durum wheat is an important cereal crop grown mainly in semi-arid environments (e.g. Mediterranean regions) characterized by water scarcity and high temperatures often occurring at the same time. Identification of Candidate Master Regulators of the Response to Early Heat Stress in Climate-adapted Wheat Landraces via Transcriptomic and Co-expression Network Analyses

Project description:Climate change is not only causing a rise of global temperature but also more variable climates. In major wheat-producing countries, spring temperatures have increased up to 40°C in recent years. Considering the wheat optimal growth temperature of around 20°C, wheat is particularly prone to damage by heat stress. To combat this threat, it is imperative to understand the response of wheat to early heat stress. This research is focus on understanging the transcriptional reprogramming of wheat when exposed to heat stress during the three-leaf stage. Differential expression and weighted gene co-expression network analysis have been used here before and after the stress to to identify candidate master-regulators of this transcriptional response.

Wheat "Chinese Spring" natural antisense transcription survey

Project description:The Affymetrix GeneChip Wheat Genome Array currently provides the most comprehensive coverage of the wheat genome for a microarray. In addition to using this resource for transcript expression studies and hybridization-based DNA marker discovery, we endeavored to use the GeneChip to discover the expression of natural antisense transcript (NAT) pairs. By using alternative target preparation schemes, both the sense- and antisense-strand derived transcripts were labeled and hybridized to the Wheat GeneChip. To enable maximum discovery, five different tissue types were selected for assay, and the wheat cultivar ‘Chinese Spring’ was used considering that most of the GeneChip probe sequences were based on sequencing of this genome. Genome-wide sequencing small RNAs in response to powdery mildew and heat stress in wheat

Project description:To test whether non-coding RNAs play roles in regulating response to powdery mildew infection and heat stress in wheat, by using Solexa high-throughput sequencing and computational analysis and experimental approach we cloned the small RNAs and identified 125 putative long npcRNAs from wheat leaves infected by preponderant physiological strain Erysiphe graminis f. sp. tritici (Egt) or by heat stress treatment. Among long non-coding RNAs, some were precursors of small RNAs such as microRNAs and siRNAs, two long npcRNAs were identified as signal recognition particle (SRP) 7S RNA variants, and three were characterized as U3 snoRNAs. Wheat long npcRNAs showed tissue dependent expression patterns and were responsive to powdery mildew infection and heat stress.

Deep sequencing of wheat sRNA transcriptome reveals distinct temporal expression pattern of miRNAs in response to heat, light and UV

Project description:Understanding of plant adaptation to abiotic stresses has implications in plant breeding, especially in the context of climate change. MicroRNAs (miRNAs) and short interfering RNAs play a crucial role in gene regulation. Here, wheat plants were exposed to one of the following stresses: continuous light, heat or ultraviolet radiations over five consecutive days and, leaf tissues from three biological replicates were harvested at 0, 1, 2, 3, 7 and 10 days after treatment (DAT). A total of 72 small RNA libraries were sequenced on the Illumina platform generating ~524 million reads corresponding to ~129 million distinct tags from which 232 conserved miRNAs were identified. The expression levels of 1, 2 and 79 miRNAs were affected by ultraviolet radiations, continuous light and heat, respectively. Approximately 55% of the differentially expressed miRNAs were downregulated at 0 and 1 DAT including miR398, miR528 and miR156 that control mRNAs involved in activation of signal transduction pathways and flowering. Other putative targets included histone variants and methyltransferases. These results suggest a temporal miRNA-guided post-transcriptional regulation that enables wheat to respond to abiotic stresses, particularly heat. Designing novel wheat breeding strategies such as regulatory gene-based marker assisted selection depends on accurate identification of stress induced miRNAs.

Heat-adaptive proteome in filling wheat grain

Project description:Wheat (T. aestivum L. Chinese Spring) plants were cultured in 25-cm buckets at a density of 16 plants per bucket in a greenhouse at a temperature of 15–30 °C. Plants were watered once a week, and artificial light was applied when there was fewer than 12 h of natural daylight. The flowering date of each spike was recorded when the first flower appeared on the spike. Comparable wheat plants were selected depending on the flowering time and growth status and were transferred into growth chambers 12 DAF for a three-day adaptation period to the growth chamber environment (24/17 °C, 14/10 h day/night cycle). Wheat plants were illuminated with a photosynthetic photon flux density of 360 µmol m˗2 s˗1 at canopy height, and relative humidity ranged from 40% to 70%. Half of the wheat plants (about 50 plants) were transferred to heated conditions (37/17 °C, 14/10 h day/night cycle, 40% to 70% humidity), while the others were maintained under the standard conditions described above as a control. After three day/night cycles, the heated samples were collected after the plants had been exposed to heat for 4 h, and the control samples from the unheated chambers were collected at the same time for and proteomic analyses. A minimum of 20 spikes were collected from each condition, immediately immersed in liquid nitrogen and stored at ˗80 °C until further analysis.
