Project description:Cellular proteins in central nervous system involved in avian neurotropic virus infection remains completely unknown. To investigate host gene expression profile in NDV infected SPF chicken brains, The microarray initial analysis was performed at LC-Bio (Hangzhou, China). A 44K Agilent chicken whole genome chip (43,803 probes) (Agilent Technologies, USA) was used for gene microarray analysis from F48E9-, LaSota-infected and mock-infected brains through intraocular-nasal routes.The chicken brains were collected at 5 day post infection. The significance analysis was used to evaluate the differences in gene expression. P and fold change (FC) values represent the alteration tendency of gene expression between experimental and control groups. The genes (FC>2) were input in Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis for DEGs.

Project description:N6-methyladenosine (m6A) modification is the most prevalent post-transcriptional modification of mRNA in eukaryotes and has been reported to play an important role in viral infection. However, the role of m6A modification during Newcastle disease virus infection (NDV) is not clear. In this study, we profiled the transcriptome-wide m6A modification of mRNA in NDV-infected chicken macrophages using MeRIP-seq. we identified a total of 9496 significantly changed peaks, of which 7015 peaks distributed across 3320 genes were significantly up-regulated and 2481 peaks distributed across 1264 genes were significantly down-regulated. Combined analysis of m6A peaks and mRNA expression revealed 1234 mRNAs with significantly altered methylation and expression levels after NDV infection, and m6A modification tended to have a negatively correlation with mRNA expression, suggesting that m6A modification may regulate the process of NDV infection by regulating gene expression, especially innate immunity-related genes. To our knowledge, This is the first comprehensive characterization of m6A patterns of the mRNA in chicken macrophages after NDV infection, and provides a valuable basis for further exploring the role of m6A modification in the process of NDV infection.

Project description:B lymphocytes from the chicken Bursa of Fabricius were isolated and cultured in the presence of CD40L. Cells were infected with a GFP fluorescent reporter virus of the very virulent Marek's disease virus (vvMDV) strain RB-1B (RB-1B_UL47-GFP, see doi: 10.1073/pnas.1424420112). After 24h viable infected (GFPpositive) and uninfected (GFPnegative) B-cells were sort purified from these cultures. Viable B cells, purified from uninfected cultures served as control. 3x10^6 cells per sample were subjected to RNA isolation and microarray analysis. Goal of the experiment was to elucidate the reaction of chicken B cells, primary target cells of MDV upon infection with this virus.

Project description:Two environmental factors, Newcastle disease and heat stress, are concurrently negatively impacting poultry worldwide and warrant greater attention into developing genetic resistance within chickens. Using two genetically distinct and highly inbred layer lines, Fayoumi and Leghorn, we explored how different genetic backgrounds affect the bursal response to a treatment of simultaneous Newcastle disease virus (NDV) infection at 6 days postinfection (dpi) while under chronic heat stress. The bursa is a primary lymphoid organ within birds and is crucial for the development of B cells. We performed RNA-seq and ChIP-seq targeting histone modifications on bursa tissue. Differential gene expression revealed that Leghorn, compared to Fayoumi, had significant down-regulation in genes involved in cell proliferation, cell cycle, and cell division. Interestingly, we also found greater differences in histone modification levels in response to treatment in Leghorns than Fayoumis, and biological processes enriched in associated target genes of H3K27ac and H3K4me1 were similarly associated with cell cycle and receptor signaling of lymphocytes. Lastly, we found candidate variants between the two genetic lines within exons of differentially expressed genes and regulatory elements with differential histone modification enrichment between the lines, which provides a strong foundation for understanding the effects of genetic variation on NDV resistance under heat stress. This study provides further understanding of the cellular mechanisms affected by NDV infection under heat stress in chicken bursa and identified potential genes and regulatory regions that may be targets for developing genetic resistance within chickens.

The transcripts of heat shock protein including Hsp 40 and Hsp 90 were significantly changed respond to thermal stress and genes involved in regulation of cell morphogenesis were significantly upregulated in heat stressed group with comparison to control and temperature recovery group. Additionally, the down-regulated genes in both heat stress and temperature recovery groups compared to control group were enriched in muscle organ development, striated muscle tissue development, cardiac muscle tissue development and muscle tissue development, which indicates that muscle development was inhibited during and in short-term after heat treatment. Most of genes dysregulated in heat stress group were found to be recovered in temperature recovery group, which confirmed their roles they could play in coping with heat stress. The present study provides a broader understanding of molecular mechanisms underlying the stress response in chicken and discovery of novel genes that are regulated in a thermal stress specific manner.
