Project description:Gut immunocompetence involves immune, stress, and regenerative processes. To investigate the determinants underlying inter-individual variation in gut immunocompetence, we perform enteric infection of 140 Drosophila lines with the entomopathogenic bacterium Pseudomonas entomophila and observe extensive variation in survival. Using genome-wide association analysis, we identify several novel immune modulators. Transcriptional profiling further shows that the intestinal molecular states of resistant and susceptible lines differ, already before infection, with one transcriptional module involving genes linked to reactive oxygen species (ROS) metabolism contributing to this difference. This genetic and molecular variation is physiologically manifested in lower ROS activity, lower susceptibility to ROS-inducing agent, faster pathogen clearance and higher stem cell activity in resistant versus susceptible lines. This study provides novel insights into the determinants underlying population-level variability in gut immunocompetence, revealing how relatively minor, but systematic genetic and transcriptional variation can mediate overt physiological differences that determine enteric infection susceptibility.

Project description:Insect gut microbiota plays important roles in acquiring nutrition, preventing pathogens infection, immune responses, and communicating with the environment. Gut microbiota can be affected by some external factors such as foods, temperature, and antibiotics. Spodoptera frugiperda (Lepidoptera: Noctuidae) is an important destructive pest of grain crops all over the world. The function of gut microbiota in S. frugiperda remains to be investigated. In this study, we fed the S. frugiperda with the antibiotic mixture (penicillin, gentamicin, rifampicin, and streptomycin) to perturb the gut microbiota, and further examined the effect of dysbiosis in gut microbiota on the gene expression of S. frugiperda by RNA sequencing. We found the composition and diversity of the gut bacterial community were changed in S. frugiperda after antibiotics treatmen, and the expression of genes related to energy and metabolic process were affected after antibiotics exposure in S. frugiperda. Our work will help understand the role of gut microbiota in insects.

Project description:Equine atypical myopathy (AM) is a severe environmental intoxication linked to the ingestion of protoxins contained in seeds and seedlings of the sycamore maple (Acer pseudoplatanus) in Europe. The toxic metabolites cause a frequently fatal rhabdomyolysis syndrome in grazing horses. Since toxic metabolites can also be present in co-grazing horses, it is still unclear as to why, in a similar environmental context, some horses show signs of AM whereas others remain clinically healthy. Label-free proteomics analyses on the serum of 26 diseased AM, 23 co-grazers and 11 control horses were performed to provide insights into biological processes and pathways. A total of 43 and 44 differentially expressed proteins between “AM vs co-grazing horses” and “AM vs control horses” were found. Disease-linked changes in the proteome of different groups were found to correlate with detected amounts of toxins and principal component analysis was performed to identify the 29 proteins with the greatest impact on the proteomic differences between groups. Among the pathway-specific changes the glycolysis/gluconeogenesis pathway, the coagulation/complement cascade, and the biosynthesis of amino acid were affected. Sycamore maple poisoning results in a combination of inflammation, oxidative stress, and high energy demand, which is trying to be met by enhanced glycolysis.

Project description:Background: Gastrointestinal nematodes are a serious cause of morbidity and mortality in grazing ruminants. The major ovine defence mechanism is acquired immunity, which develops over time in response to infection. Nematode resistance varies both within and between breeds and is moderately heritable (h ~ 0.3). A detailed understanding of the genes and mechanisms involved in protective immunity, and the factors that regulate this response, is required to aid future breeding strategies as well as the development of effective and sustainable nematode control methods. The aim of this study was to compare the abomasal lymph node transcriptome of resistant and susceptible lambs in order to determine biological processes differentially expressed between resistant and susceptible lambs. Results: Scottish Blackface lambs, with divergent phenotypes for resistance, were challenged with 30,000 Teladorsagia circumcincta larvae (L3), and abomasal lymph node recovered at 7 and 14 days post-infection (dpi). High-throughput sequencing of abomasal lymph node cDNA was used to quantitatively sample the transcriptome with an average of 32 million reads per sample. A total of 194 and 144 genes were differentially expressed between resistant and susceptible lambs at 7 and 14 dpi respectively. Differentially expressed networks and biological processes were identified using Ingenuity Pathway Analysis. Resistant animals appear to generate a more rapid immune response as at 7 dpi processes relating to homing of lymphocytes, leukocyte migration and migration of antigen presenting cells were up-regulated. In susceptible animals this response appears to be delayed until approximately 14 dpi. Twenty-four Single Nucleotide Polymorphims (SNP), within 11 differentially expressed genes were tested for association with gastrointestinal nematode resistance in the Scottish Blackface lambs. Four SNPs in two genes (SLC30A2, and ALB) were suggestively associated with faecal egg count. Conclusions: A large number of genes were differentially expressed in the abomasal lymph node of resistant and susceptible lambs responding to gastrointestinal nematode challenge. Resistant Scottish Blackface lambs appear to generate a more rapid immune response to T. circumcincta. In susceptible lambs this response appears to be delayed until approximately 14 days post infection. SNP in two differentially expressed genes were suggestively associated with faecal egg count indicating that differentially expressed genes can be considered as candidate loci for mediating nematode resistance.

Project description:Antibiotics have long-lasting consequences on the gut microbiota with the potential to impact host physiology and health. However, little is known about the transgenerational impact of an antibiotic-perturbed microbiota. Here we demonstrated that adult pregnant female mice inoculated with a gut microbial community shaped by antibiotic exposure passed on their dysbiotic microbiota to their offspring. This dysbiotic microbiota remained distinct from controls for at least 5 months in the offspring without any continued exposure to antibiotics. By using IL-10 deficient mice, which are genetically susceptible to colitis, we showed mice that received an antibiotic-perturbed gut microbiota from their mothers had increased risk of colitis. Taken together, our findings indicate that the consequences of antibiotic exposure affecting the gut microbiota can extend to a second generation.

Project description:Whereas canines are susceptible to mammary cancer at a frequency similar to humans, equines are largely resistant to the disease. We treated mammary stem cells from both species with the chemical carcinogen, DMBA, and found that equine cells are significantly more susceptible to treatment. These RNA-seq results represent samples from three individuals of dogs and horses in order to reveal transcriptional differences in response to treatment that may explain the phenotypic differences we observed.
