Data-independent acquisition proteomics reveals the effects of red and blue light on the growth and development of moso bamboo(Phyllostachys edulis) seedlings

Project description:Moso bamboo is a fast-growing bamboo species with high economic, social and cultural value. The method of transplanting moso bamboo seedlings for afforestation has become a more economical and effective method. The effect of light on the growth of plant seedlings is mainly reflected in the regulation of different light quality on the growth and development of seedlings, including light morphogenesis, photosynthesis and secondary metabolites. Therefore, studying the effects of specific wavelength light on the physiology and proteome of moso bamboo seedlings will play an important role in growing seedlings and seed cultivation of moso bamboo. Here, moso bamboo seeds were germinated in the dark and then were transferred to the blue and red-light conditions. After 14 days, we observed the effects of different light treatments on the growth and development of seedlings, and then compared and analyzed their proteome.

Expression data from Arabidopsis under hydrogen peroxide and light treatment

Project description:Green plants are more robust to hydrogen peroxide (H2O2) stress and contain high endogeneous H2O2 levels which is generated during photorespiration and photosynthesis. Therefore, exgeneous H2O2 application mostly impose oxidative stress. To reduce endogenous H2O2 background, we adopted a strategy which is to grow Arabidopsis seedlings in the dark to eliminate light-induced H2O2 production, thus to reduce the endogenous H2O2 level. Exogenous H2O2 was then applied to induce transcriptome changes. Global gene expression is studied and compared between samples collected under 7d dark, 7d H2O2 treatment under dark and 7d light conditions. We cultured seedlings in the dark to reduce endogenous H2O2. Three conditions were used for transcriptome profiling: dark grown (dark); dark grown with exogenous H2O2 treatment (H2O2); and light grown (light). Three types of conditions were used for Arabidopsis seedling culture: dark, dark with 5 mM H2O2 treatment and light. Each condition was performed with two biological replicates. The seedlings were harvested at 7 days old.

Transcription profiling of light and carbon signaling interactions in Arabidopsis

Project description:A. thaliana seeds of Columbia (Col-0) ecotype were surface-sterilized, plated on designated media and vernalized for 48 hours at 8C. Plants were grown semi-hydroponically under 16-hr-light, 8-hr-dark cycles at a constant temperature of 23C on basal Murashige and Skoog medium supplemented with 2 mM KNO3, 2 mM NH4NO3 and 30 mM sucrose. Two-week-old seedlings were transferred to fresh MS media without nitrogen and sucrose and dark-adapted for 48 hours. To perform specific metabolic treatments manipulating availability of source of carbon (C) and illumination (L): -C-L, +C-L, -C+L, +C+L, two-week-old seedlings were transferred to fresh MS medium containing 0% or 1% sucrose and either placed back into the dark or illuminated with white light for an additional 8 hours. The various treatments were compared to one another using the treatment of -C-L as the background treatment to which all other treatments (-C+L, +C-L, +C+L) were compared.

A light-independent allele of phytochrome B faithfully recapitulates photomorphogenic transcriptional networks

Project description:Dominant gain-of-function alleles of Arabidopsis phytochrome B were recently shown to confer light-independent, constitutive photomorphogenic (cop) phenotypes to transgenic plants (Su & Lagarias 2007 Plant Cell 19, 2124-2139). In the present study, comparative transcript profiling experiments were performed to assess whether the pattern of gene expression regulated by these alleles accurately reflects the process of photomorphogenesis in wild-type Arabidopsis. Whole genome transcriptional profiles of dark-grown phyAphyB seedlings expressing the Y276H mutant of phyB (YHB) revealed that YHB reprograms about 13% of the Arabidopsis transcriptome in a light-independent manner. The YHB-regulated transcriptome proved qualitatively similar to, but quantitatively greater than those of wild-type seedlings grown under 15 or 50 umol m-2 m-1 continuous red light (Rc). Among the 2977 genes statistically significant two-fold (SSTF) regulated by YHB in the absence of light include those encoding components of the photosynthetic apparatus, tetrapyrrole/pigment biosynthetic pathways and early light-responsive signaling factors. Approximately 80% of genes SSTF regulated by Rc were also YHB-modulated. Expression of a notable subset of 346 YHB-regulated genes proved to be strongly attenuated by Rc, indicating compensating regulation by phyC-E and/or other Rc-dependent processes. Since the majority of these 346 genes are regulated by the circadian clock, these results suggest that phyA- and phyB-independent light signaling pathway(s) strongly influence clock output. Together with the unique plastid morphology of dark-grown YHB seedlings, these analyses indicate that the YHB mutant induces constitutive photomorphogenesis via faithful reconstruction of phyB signaling pathways in a light-independent fashion.

Project description:In this study, we have used the comparative transcriptomic and proteomic approaches to decipher the changes in genes and protein abundances in cigar tobacco leaves under LL. In this study, DEGs and DEPs related to glycolysis, starch and sucrose metabolism, tyrosine metabolism, photosynthesis-antenna proteins, and photosynthesis pathways are significantly enriched. This study offers novel insights into both transcriptome and proteome levels response mechanisms under different light intensities.
