Enrichment tests show genes related to migration are over-represented in the highly expressed genes of TMK vs. EK.

Project description:The data set compares the microRNA expression of two types of keratinocytes from the tympanic membrane and epidermis of the body. Healthy human samples were harvested for the analysis. Cells were cultivated to evaluate the effect of keratinocytes only. Few Chip Ids (11) were considered differentially expressed between the cell types. Six samples were analyzed in total. Each cell type in biological triplicates. Genes of p-value <0.05 and fold change of â¥ 1.5 were considered significant.

Single-cell RNA sequencing elucidates cell populations in the murine and human tympanic membrane

Project description:Previous descriptions of the cellular makeup of the tympanic membrane (TM) describe three basic cell types: keratinocytes, fibrofblasts, and muscosal cells. To better understand the diversity of cells in this tissue, we performed single-cell RNA sequencing on dissociated murine and human TMs. We identified the basic cell types previously known, as well as subtypes of cells in each group. Notably, keratinocyte cell clusters spanning the known differentiation kierarchy were identified, including two groups of undifferentiated keratinocytes which we propose make distinct contributions to maintaining the TM epidermis.

Hypoxia reveals the new function of Foxn1 in keratinocytes antioxidant defense system

Project description:Skin exposed to environmental threats including injuries and oxidative stress developed an efficient but not fully recognized systems of repair and antioxidant protection. In this project we provide evidence that Foxn1 in keratinocytes regulates elements of electron transport chain and participates in thioredoxin system (Txn2, Txnrd3, Srxn1) induction, particularly under hypoxic environment. It is shown that Foxn1 in keratinocytes upregulate glutathione thioredoxin reductase 3 (Txnrd3) protein, and high levels of Txnrd3 mRNA were detected in injured skin of Foxn1+/+ mice. It also shown that Foxn1 strongly downregulate CCN2 protein participating in reconstruction of the epidermis after injury. As in vitro assay revealed that Foxn1 controls keratinocytes migration stimulating it under normoxia and suppressing under hypoxia. Keratinocytes overexpressing Foxn1 and exposed to hypoxia showed reduced ability to promote angiogenesis by downregulation of Vegfa. In conclusion, this study showed a new mechanism in which Foxn1, along with hypoxia, participates in the activation of antioxidant defense and controls functional properties of keratinocytes.
