Microarray analysis of rat hepatic gene expression identifies new genes associated with postprandial triglyceridemia

Project description:The acute response four hours after a fat load of extra virgin olive oil was investigated using DNA microarrays. Hepatic gene expression was analysed in Wistar Rats. Male rats, weighing 250- 300 g (purchased from Charles River, Barcelona, Spain), were used for experiments. Rats, housed in sterile filter-top cages (3-4 per cage), were acclimatized in a room maintained at 20ºC with a 12-h light-dark cycle for 10 days, allowed ad libitum access to water and standard chow diet (Pascual S.A., Barcelona, Spain), and fasted for 18 h before experiments. Two study groups were established: a) The control group did not receive any fat meal. The other group was fed 5 ml of olive oil (Aceites Toledo, Spain) as a bolus and sacrificed 4 hours after the feeding, respectively. This amount represents the use of a dose of 16 mL olive oil/ kg.

Genome Wide Association Study:GR@ACE Stage I

Project description:The Genome Research program at Fundacio ACE (GR@ACE, Barcelona, Spain), represents a new effort to study Alzheimer's Disease susceptibility. The GR@ACE study comprises 7,409 unrelated individuals. 4,120 AD cases were diagnosed at Fundació ACE Memory Clinic (Barcelona, Spain). 3,289 Controls were obtained from three collections (Fundació ACE, Barcelona; DNA National Biobank, Salamanca and HU Valme, Seville). Genotyping was carried out using the 815K Axiom Spain Biobank array (Affymetrix.) in the Spanish national center for genotyping (CEGEN, Santiago de Compostela, Spain). After genotyping, an extensive quality control was conducted, including re-sampling, gender match, stratification, relatedness, heterozygosity, differential missingness and HWD analyses.

Real-time quantitative PCR analysis of human total blood tissue from athletes or sedentary controls

Project description:Thirteen elite handball athletes and 13 sedentary controls. Three timepoints were established: T0 (baseline conditions); T8 (after 8 weeks of supplementation); and T16 (after 8 weeks in the absence of supplementation). The dietary intervention consisted of the oral administration of one daily multivitamin/mineral complex capsule (Multicentrum® Pfizer, Barcelona, Spain) before exercise during the controlled dietary intervention period. Multivitamin/mineral complex intervention adherence/compliance was defined as the percentage of all of the supplement capsules ingested throughout the study period. The expressions of a total 112 of genes were evaluated by RT-qPCR analysis with the QuantStudioTM 12K Flex Real-Time PCR System. 78 genes were finally analized.

Two Low Volume Regimens for Colorectal Cancer Screening Colonoscopy

Project description:Colorectal cancer is the first neoplasm most commonly diagnosed in both sexes and the second leading cause of cancer death in Spain. Colorectal cancer screening Program in Barcelona was implanted in 2009 and approximately 1,500 colorectal cancer screening colonoscopies are performed annually at Hospital Clínic de Barcelona. Adequate colon preparation (anterograde cleansing with laxatives) improves polyp detection, decreases examination time, and complications. There are both low and high volume intestinal regimens with polyethylene glycol (PEG) as the main active ingredient or without it. Hypothesis: Two low volumen regimens, 1L PEG plus ascorbate and magnesium citrate plus picosulphate, at starting doses have the same Adenoma Detection Rate as cleansing solutions in preparation for screening colonoscopy.

Comparative genomic hybridization of strains of Salmonella enterica serovar Enteritidis from Uruguay, other distant geographical regions and other serovars isolated in Uruguay

Project description:Microarray based CGH was conducted over a group of 29 strains of S. Enteritidis spanning different epidemiological periods in Uruguay, plus 6 other S. Enteritidis strains isolated from distant geographical regions. We also included 9 Salmonella enterica strains of other serovars isolated in Uruguay. A S. Enteritidis dispensable genome of 233 chromosomal genes and high extent of variation in virulence plasmid was found. Strains isolated before the epidemic show the highest genomic differences as compared with the PT4 reference strain. Comparison with the gene content of other serovars demonstrate extensive horizontal gene transfer between circulating strains beyond serovar definition. Our results show that the epidemic of S Enteritidis in Uruguay was produced by the introduction of strains closely related to PT4, and corroborate the extensive genetic homogeneity among S. Enteritidis isolates worldwide. Phage SE14 emerges as the only specific region for S. Enteritidis. Genetic differences detected in pre-epidemic strains, mainly associated with the absence of phage SE20, suggest that genetic features encoded in this phage may be related to particular epidemiological behavior.

Experiment Overall Design: Two study groups were established: a) one received a chow diet (Teklad Mouse/Rat Diet no. 2014, Harlan Teklad, Harlan IbÃ©rica, Barcelona, Spain) supplemented with 10% (w/w) olive oil (diet OO) (n=6), and b) the other received the same chow diet but supplemented with 10% (w/w) maslinic acid- enriched olive oil (diet MAOO; n= 6 and).
