Project description:Clinical heterogeneity of gastric cancer reflected in unequal outcome of treatment is poorly defined in molecular level, and molecular subtypes and their associated biomarkers have not been established to improve prognostification and treatment of gastric cancer. Using microarray technologies, we analyzed gene expression profiling data from patients with advanced gastric cancer and uncovered potential prognostic subtypes and identify gene expression signature associated with prognosis. Project description:Gastric cancer is one of the leading causes of cancer mortality worldwide, and peritoneal metastasis is a hallmark of incurable advanced gastric cancer. The identification of molecular vulnerability for such conditions is imperative to improve the prognosis of gastric cancer. Here, we comprehensively analysed cancer cells purified from malignant ascitic fluid samples and their corresponding cell lines from 98 patients, through whole-genome sequencing, whole transcriptome sequencing, methylation analyses, and genome-wide enhancer analyses.

EGFR expression status associated with lymph node ratio

Project description:We have tried to identify molecules associated with lymph node ratio expression by microarray. Expression microarray data prioritized gene candidates according to average expression ratio and its frequency. Among these genes, we examined particular EGFR expression by immunohistochemistry (IHC) in 167 advanced gastric cancer. We compared tumor specimens with high LNR (n=4) with those with low one (n=4) at mRNA level by Affymetrix microarray (harboring 54,675 genes) in stage III gastric cancer. We then prioritized the expression data, and enriched genes correlated with LNR, and finally validated EGFR as such candidates in primary advanced gastric cancer who underwent standard treatment (n=167).
