Project description:The aim of this study was to evaluate the potential effects of methyl donor supplementation of pregnant animals in the presence or absence of a concomitant lactation on the methylome of the offspring. Twenty Holstein cows, 10 nulliparous (not lactating while pregnant) and 10 multiparous (lactating while pregnant) were blocked by parity and randomly assigned to an i.m. weekly injections of a placebo (CTRL) or a solution containing methyl donors (MET). After calving, 5 calves randomly selected from each treatment (two born to primiparous and three to multiparous dams) were blood-sampled to determine their full methylome. There were more than 2,000 CpG differentially methylated between calves born to CTRL and those born to MET, and also between calves born to multiparous and nulliparous dams. Most of the differences affected genes involved in immune function, cell growth regulation and differentiation, kinase activity, and ion channeling. We conclude that the coexistence of pregnancy and lactation affects the methylome of the offspring, and that supplementation of methyl donors early in gestation has also consequences on the methylome.

Project description:Infertility in lactating dairy cows is explained partially by the metabolic state associated with high milk production. The hypothesis was that lactating and non-lactating cows would differ in endometrial and placental transcriptomes during early pregnancy (day 28 to 42) and this difference would explain the predisposition for lactating cows to have embryonic loss at that time. Cows were either milked or not milked after calving. Reproductive [endometrium (caruncular and intercarunclar) and placenta] and liver tissues were collected on day 28, 35, and 42 of pregnancy. The primary hypothesis was rejected because no effect of lactation on mRNA abundance within reproductive tissues was found. Large differences within liver demonstrated the utility of the model to test an effect of lactation on tissue gene expression. Major changes in gene expression in reproductive tissues across time were found. Greater activation of the transcriptome for the recruitment and activation of macrophages was found in the endometrium and placenta. Changes in glucose metabolism between day 28 and 42 included greater mRNA abundance of rate-limiting genes for gluconeogenesis in intercaruncular endometrium and evidence for the establishment of aerobic glycolysis (Warburg effect) in the placenta. Temporal changes were predicted to be controlled by CSF1, PDGFB, and JUN. Production of nitric oxide and reactive oxygen species by macrophages was a mechanism to promote angiogenesis in the endometrium. Reported differences in pregnancy development for lactating versus non-lactating cows could be explained by systemic glucose availability to the conceptus and appear to be independent of the endometrial and placental transcriptomes.

Project description:Growth hormone (GH) has a well established galactopoietic effect on ruminant lactation, however the molecular mechanisms that mediate these effects are not fully understood. This study aimed to define the effects of GH on the production of milk protein by the bovine mammary gland and to establish the potential molecular mechanisms mediating these effects. Elevated levels of GH increased milk yield with corresponding increases in protein, fat and lactose yield, while milk composition remained unaffected by treatment. Increased yield (but not concentration) of all the individual milk proteins was not accompanied by changes in the levels of the milk protein transcripts, with the exception of S2-casein, suggesting post-transcriptional regulation of the milk protein genes is a main factor in the GH-mediated effects on bovine lactation. There were significant transcriptional changes in a wide range of pathways including cell signaling, cell death, cell growth and proliferation, cell cycle regulation and metabolism, consistent with the broad effects of GH. Differential expression of genes involved in the PI3K, ERK/MAPK, insulin receptor, JAK/STAT and IGF-1 signaling pathways were also evident in response to GH treatment supporting potential cross-talk between the GH and insulin signaling pathways in the bovine mammary gland. Transcript levels of the SHC1 gene were elevated in GH-treated cows. SHC1 activates many pathways including those associated with translational regulation and gene transcription and has been implicated in mediating cross-talk between the insulin and GH signaling pathways, providing some insights into the potential pathways mediating the effects of GH on milk protein synthesis in the bovine mammary gland.

Project description:The protein profile of bovine milk serum was characterised as milk transitions from colostrum to transition milk over the first 5 days of lactation. Samples were collected from first and third parity cows at days 0, 2, 5 (D0, D2, D5) after calving. Following isolation of the milk serum fraction, label-free quantitative proteomics was carried out following normalisation by total protein concentration. Protein profiles indicated samples clustered by day postpartum, but not by parity. Proteins (n = 471) were identified and relative quantification was performed, with 199 protein groups showing altered abundance by day of lactation (fold change ≥ 2, P < 0.05). Elevated levels of immune proteins, including immunoglobulins and complement proteins were detected in colostrum with levels significantly decreasing by D2. These findings provide an outline of the dynamics of the protein profile of bovine milk and colostrum in early lactation.

Project description:Breast cancer risk is influenced by parity in an age-dependant manner, however human tissue remodelling induced by pregnancy and lactation is not well understood. In most cases, it is difficult to acquire human breast tissue during these key stages of development. Here, we present an approach to overcome this using single-cell RNA sequencing to examine viable primary mammary epithelial cells isolated from human milk (n=4) compared to resting, non-lactating human breast tissue (n=4). Overall, we examined 54,323 viable high-quality cells from mixed age and parous individuals consisting of 29,078 lactation derived mammary cells (LMCs) and 25,245 non-lactation derived mammary cells (NMCs). We identified all documented mammary subpopulations within our breast tissue samples and found that milk contains distinct secretory luminal cells together with myeloid and lymphocytic hematopoietic lineage cells. Comparing the luminal transcriptional profiles of cells isolated from the resting and lactating state identified differences in mammary cell function and metabolism between these maturation states. In future this data may be used to provide an insight into how parity influences human luminal cell metabolism and breast cancer risk.

Project description:The objective of this study was to apply a quantitative proteomic approach to investigate changes in the plasma proteome of normal and over-conditioned dairy cows during the transition period. We used an animal model with a targeted difference in body condition score (BCS) at calving. For this, multiparous Holstein cows were allocated to two groups 15 weeks before anticipated calving and were fed differently to reach either high (HBCS: 7.2 NEL MJ/kg dry matter (DM) or normal BCS (NBCS: 6.8 NEL MJ/kg DM) until dry-off. Thereafter, both groups were fed identical diets. The targeted BCS difference was sustained during the dry period (7 weeks before calving) and the subsequent 12 weeks of lactation. Plasma samples (n = 5) from d -49, +7, and +21 relative to parturition were used from both groups (each n = 5) for proteome profiling in aby applying quantitative Tandem Mass Tags (TMT) approach.

Project description:Targeting the gastrointestinal microbiome for improvement of feed efficiency and reduction of production costs is a potential promising strategy. However little progress has been made in manipulation of the gut microbiomes in dairy cattle to improve milk yield and milk quality. Even less understood is the milk microbiome. Understanding the milk microbiome may provide insight into how the microbiota correlate with milk yield and milk quality. The objective of this study was to characterize similarities between rumen, fecal, and milk microbiota simultaneously, and to investigate associations between microbiota, milk somatic cell count (SCC), and milk yield. A total of 51 mid-lactation, multiparous Holstein dairy cattle were chosen for sampling of ruminal, fecal, and milk contents that were processed for microbial DNA extraction and sequencing. Cows were categorized based on low, medium, and high SCC; as well as low, medium, and high milk yield. Beta diversity indicated that ruminal, fecal, and milk populations were distinct (p < 0.001). Additionally, the Shannon index demonstrated that ruminal microbial populations were more diverse (p < 0.05) than were fecal and milk populations, and milk microbiota was the least diverse of all sample types (p < 0.001). While diversity indices were not linked (p > 0.1) with milk yield, milk microbial populations from cows with low SCC demonstrated a more evenly distributed microbiome in comparison to cows with high SCC values (p = 0.053). These data demonstrate the complexity of host microbiomes both in the gut and mammary gland. Further, we conclude that there is a significant relationship between mammary health (i.e., SCC) and the milk microbiome. Whether this microbiome could be utilized in efforts to protect the mammary gland remains unclear, but should be explored in future studies.

Project description:Fat metabolism is a complicated process regulated by a series of factors. microRNAs (miRNAs) are a class of negative regulator of proteins and play crucial roles in many biological processes, including fat metabolism. Although there have been some researches indicating that miRNAs could influence the milk fat metabolism through targeting some factors, little is known about the effect of miRNAs on goat milk fat metabolism. Here we utilized an improved miRNA detection assay, S-Poly(T), to profile the expression of miRNAs in the goat mammary gland in early-lactation and dry-lactation. Overall, we found 146 miRNAs expression changed between early-lactation and dry-lactation, with 81 up-regulation and 65 down-regulation, among which the miR-27a, miR-200a and miR-200c have been reported to be involved in the milk fat metabolism process.

Project description:The fertility of dairy cows is challenged during early lactation and better nutritional strategies need to be developed to address this issue. Combined supplementation of folic acid and vitamin B12 improves energy metabolism in the dairy cow during early lactation. Therefore, the present study was undertaken to explore the effects of this supplement on gene expression in granulosa cells from the dominant follicle during the postpartum period. Multiparous Holstein cows received weekly intramuscular injection of 320 mg folic acid and 10 mg vitamin B12 (treated group) beginning 24 (SD 4) d before calving until 56 d after calving, whereas the control group received saline. The urea plasma concentration was significantly decreased during the pre-calving period, and the concentration of both folate and vitamin B12 were increased in treated animals. Milk production and dry matter intake were not significantly different between the two groups. Plasma concentrations of folates and vitamin B12 were increased in vitamin-treated animals. Daily dry matter intake was not significantly different between the 2 groups before (13.5 kg SE 0.5) and after (23.6 kg SE 0.9) calving. Average energy-corrected milk tended to be greater in vitamin-treated cows, 39.7 (SE 1.4) and 38.1 (SE 1.3) kg/d for treated and control cows, respectively. After calving, average plasma concentration of BHBA tended to be lower in cows injected with the vitamin supplement, 0.47 (SE 0.04) vs. 0.55 (SE 0.03) for treated and control cows, respectively. The ovarian follicle ? 12 mm in diameter was collected by ovarian pick-up after estrus synchronization. Recovered follicular fluid volumes were greater in the vitamin-treated group. A microarray platform was used to investigate the impact of treatment on gene expression of granulosa cells. Lower expression of genes involved in the cell cycle and higher expression of genes associated with granulosa cell differentiation prior to ovulation were observed. Selected candidate genes were analyzed by reverse transcription quantitative polymerase chain reaction. Although the effects of intramuscular injections of folic acid and vitamin B12 on lactational performance and metabolic status of animals were limited, Ingenuity Pathway Analysis of gene expression in granulosa cells suggests a stimulation of cell differentiation in vitamin-treated cows, which may be the result of an increase in LH secretion.
