Project description:Heavy payloads in future shuttle journeys to Mars present limiting factors, making self-sustenance essential for future colonies. Therefore, in situ resources utilization (ISRU) is the path to successful and feasible space voyages. This research frames the concept of planting leafy vegetables on Mars regolith simulant, ameliorating this substrate's fertility by the addition of organic residues produced in situ. For this purpose, two butterhead lettuce (Lactuca sativa L. var. capitata) cultivars (green and red Salanova®) were chosen to be cultivated in four different mixtures of MMS-1 Mojave Mars simulant:compost (0:100, 30:70, 70:30 and 100:0; v:v) in a phytotron open gas exchange growth chamber. The impact of compost rate on both crop performance and the nutritive value of green- and red-pigmented cultivars was assessed. The 30:70 mixture proved to be optimal in terms of crop performance, photosynthetic activity, intrinsic water use efficiency and quality traits of lettuce. In particular, red Salanova® showed the best performance in terms of these quality traits, registering 32% more phenolic content in comparison to 100% simulant. Nonetheless, the 70:30 mixture represents a more realistic scenario when taking into consideration the sustainable use of compost as a limited resource in space farming, while still accepting a slight significant decline in yield and quality in comparison to the 30:70 mixture.

Although the mechanisms underlying their effect remain unclear, it has been postulated that miRNAs could be involved. In order to understand these mechanisms we performed NGS sequencing in potato leaves treated or not with KPhi to identify miRNAs responsive to this biocompatible compound. The aim of the present work is to identify miRNAs that are involved in the regulation of potato defense responses after phosphite treatment.

Project description:Crop rotation and intercropping with Allium plants suppresses Fusarium wilt in various crops. However, the mechanisms underlying this phenomenon have not been fully elucidated. This study was designed to assess the role of microorganisms inhabiting Allium rhizospheres and antifungal compounds produced by Allium roots in Fusarium wilt suppression by Allium cultivation. Suppression of cucumber Fusarium wilt and the pathogen multiplication by Allium (Welsh onion and/or onion)-cultivated soils were eliminated by heat treatment at 60?°C, whereas those by Welsh onion-root extract were lost at 40?°C. The addition of antibacterial antibiotics eliminated the suppressive effect of Welsh onion-cultivated soil on pathogen multiplication, suggesting the contribution of antagonistic gram-negative bacteria to the soil suppressiveness. The Illumina MiSeq sequencing of 16S rRNA gene amplicons revealed that genus Flavobacterium was the predominant group that preferentially accumulated in Allium rhizospheres. Flavobacterium species recovered from the rhizosphere soils of these Allium plants suppressed Fusarium wilt on cucumber seedlings. Furthermore, confocal laser scanning microscopy revealed that Flavobacterium isolates inhibited the multiplication of the pathogen in soil. Taken together, we infer that the accumulation of antagonistic Flavobacterium species plays a key role in Fusarium wilt suppression by Allium cultivation.

Project description:Micro RNAs (miRNAs) are small single strand non-coding RNAs that regulate gene expression at the post-transcriptional level, either by translational inhibition or mRNA degradation based on the extent of complementarity between the miRNA and its target mRNAs. Potato (Solanum tuberosum L.) is the most important horticultural crop in Argentina. Achieving an integrated control of diseases is crucial for this crop, where frequent agrochemical applications, particularly fungicides, are carried out. A promising strategy is based on promoting induced resistance through the application of environmentally friendly compounds such as phosphites, inorganic salts of phosphorous acid. The use of phosphites in disease control management has proven to be effective. Although the mechanisms underlying their effect remain unclear, we postulated that miRNAs could be involved. Therefore we performed next generation sequencing (NGS) in potato leaves treated and non treated with potassium phosphite (KPhi). We identified 25 miRNAs that were expressed differentially, 14 already annotated in miRBase and 11 mapped to the potato genome as potential new miRNAs. A prediction of miRNA targets showed genes related to pathogen resistance, transcription factors, and oxidative stress. We also analyzed in silico stress and phytohormone responsive cis-acting elements on differentially expressed pre miRNAs. Despite the fact that some of the differentially expressed miRNAs have been already identified, this is to our knowledge the first report identifying miRNAs responsive to a biocompatible stress resistance inducer such as potassium phosphite, in plants. Further characterization of these miRNAs and their target genes might help to elucidate the molecular mechanisms underlying KPhi-induced resistance.

Project description:Wheat (Triticum aestivum L.) is a strategic agricultural crop that plays a significant role in maintaining national food security and sustainable economic development. Increasing technical performance considering lowering costs, energy, and environmental consequences are significant aims for wheat cultivation. For drylands, which cover approximately 41% of the world's land surface, water stress has a considerable negative impact on crop output. The current study aimed to assess the environmental aspects of chemical fertilizer in combination with compost in dryland and irrigated winter wheat production systems through life cycle assessment (LCA). The cradle-to-farm gate was considered as the system boundary based on one tone of wheat yield and four strategies: D-C (dryland with compost), D (dryland without compost), I-C (irrigated with compost), and I (irrigated without compost). Based on the results, the highest and lowest amounts of wheat yield were related to the I-C and D strategies with 12.2 and 6.7 ton ha-1, respectively. The LCA result showed that the I strategy in comparison with other strategies had the highest negative impact on human health (49%), resources (59%), ecosystem quality (44%), and climate change (43%). However, the D-C strategy resulted in the lowest adverse effect of 6% on human health, 1% on resources, 10% on ecosystem quality, and 11% on climate change. Utilizing a combination of fertilizer and compost in dryland areas could ensure a higher yield of crops in addition to alleviating negative environmental indicators.

Project description:Xylanases are in important class of industrial enzymes that are essential for the complete hydrolysis of lignocellulosic biomass into fermentable sugars. In the present study, we report the cloning of novel xylanases with interesting properties from compost metagenomics libraries. Controlled composting of lignocellulosic materials was used to enrich the microbial population in lignocellulolytic organisms. DNA extracted from the compost samples was used to construct metagenomics libraries, which were screened for xylanase activity. In total, 40 clones exhibiting xylanase activity were identified and the thermostability of the discovered xylanases was assayed directly from the library clones. Five genes, including one belonging to the more rare family GH8, were selected for subcloning and the enzymes were expressed in recombinant form in E. coli. Preliminary characterization of the metagenome-derived xylanases revealed interesting properties of the novel enzymes, such as high thermostability and specific activity, and differences in hydrolysis profiles. One enzyme was found to perform better than a standard Trichoderma reesei xylanase in the hydrolysis of lignocellulose at elevated temperatures.

Project description:Tomato bacterial wilt caused by Ralstonia solanacearum (RS) is one of the most devastating soil-borne diseases, and compost is to be considered as a resource-saving and environment-friendly measure to control the disease. Herein, a pot experiment was implemented to explore the effects of vinegar residue matrix amendments on the growth performances of tomato seedlings and to examine the suppression ability against bacterial wilt under vinegar residue substrate (VRS), and peat substrate (Peat) with RS inoculation. The results revealed that VRS effectively suppressed the disease incidence of bacterial wilt, increased the number of bacteria and actinomycetes, decreased fungi populations, promoted soil microbial populations and microbial activities, enhanced the growths of tomato seedlings, and modulated defense mechanism. In addition, VRS efficiently inhibited the oxidative damage in RS inoculated leaves via the regulation of excess reactive oxide species (O2•- and H2O2) production, lessening of malondialdehyde (MDA) content, and causing less membrane injury; resulting in enhancements of antioxidants enzymes activities accompanying with modulating their encoding gene expression. The transcription levels of NPR1, PIN2, PR1b, ACO1, EDS1, PR1B, MAPK3, PIN2, and RRS1 were also modulated with the pathogens inoculated in tomato leaves both in VRS and Peat treatments, which indicated that systemic-acquired resistance possesses cross-talk between salicylic acid, jasmonic acid, and the ethylene-dependent signaling pathway. Besides, the RS inoculation significantly inhibited the growth of tomato seedlings, and all growth indices of plants grown in VRS were considerably higher than those produced in Peat. Taken together, VRS represents a new strategy to control tomato bacterial wilt through boosting the soil microbial populations and microbial activities. Furthermore, VRS promotes the plant immune response to provide a better growth environment for plants surviving in disease conditions.

Project description:The white button mushroom Agaricus bisporus is the most widely produced edible fungus with a great economical value. Its commercial cultivation process is often performed on wheat straw and animal manure based compost that mainly contains lignocellulosic material as a source of carbon and nutrients for the mushroom production. As a large portion of compost carbohydrates are left unused in the current mushroom cultivation process, the aim of this work was to study wild-type A. bisporus strains for their potential to convert the components that are poorly utilized by the commercial strain A15. Growth profiling suggested different abilities for several A. bisporus strains to use plant biomass derived polysaccharides, as well as to transport and metabolize the corresponding monomeric sugars. Six wild-type isolates with diverse growth profiles were compared for mushroom production to A15 strain in semi-commercial cultivation conditions. Transcriptome and proteome analyses of the three most interesting wild-type strains and A15 indicated that the unrelated A. bisporus strains degrade and convert plant biomass polymers in a highly similar manner. This was also supported by the chemical content of the compost during the mushroom production process. Our study therefore reveals a highly conserved physiology for unrelated strains of this species during growth in compost.
