Project description:Depletion of ZC3H41 has minor effects on the transcriptome of procyclic trypanosomes. In general, ZC3H41-regulated transcripts seem to be expressed at low abundance, and almost half of them (43%) are probably non-coding, since the corresponding encoded proteins have not been detected in proteomic surveys . A transcript encoding a trans-sialidase (Tb927.11.11410) was clearly upregulated in ZC3H41-depleted cells .

Nuclear pore complex evolution: A trypanosome Mlp analog functions in chromosomal segregation but lacks transcriptional barrier activity

Project description:The nuclear pore complex (NPC) has dual roles in nucleocytoplasmic transport and chromatin organisation. In many eukaryotes the coiled coil Mlp/Tpr proteins of the NPC nuclear basket have specific roles in interactions with chromatin and defining specialised regions of active transcription, while Mlp2 associates with the mitotic spindle in a cell-cycle dependent manner. We previously identified two putative Mlp-related proteins in African trypanosomes, TbNup110 and TbNup92, the latter of which associates with the spindle. We now provide evidence for independent ancestry for TbNup92/TbNup110 and Mlp/Tpr proteins. However, TbNup92 is required for correct chromosome segregation, with knockout cells exhibiting microaneuploidy and low fidelity telomere segregation. Further, TbNup92 is intimately associated with the mitotic spindle and spindle anchor site, but apparently has minimal roles in the control of gene transcription, indicating that TbNup92 lacks major barrier activity. TbNup92 therefore acts as a functional analog of Mlp/Tpr proteins, and together with the lamina analog NUP-1, represents a cohort of novel proteins operating at the nuclear periphery of trypanosomes, uncovering complex evolutionary trajectories for the NPC and nuclear lamina.

Single-cell transcriptome profiling of mouse bone marrow-derived immune cells in response to trypanosome infection

Project description:Bone marrow is an extremely important organ as a primary hematopoietic organ where new red blood cells and immune cells are produced. Parasitic infections showed disruption of red blood cell production and changes in immune responses in bone marrow. Therefore, we generated single-cell RNA sequencing (scRNA-seq) data to reveal transcriptomic profile changes to parasitic infection by trypanosomes. These data will ultimately help us understand the mechanisms of the host immune response to trypanosomosis.

The trypanosome UDP-glucose pyrophosphorylase is active in both the glycosomal and cytosolic compartments and is imported into glycosomes by piggybacking

Project description:Trypanosoma brucei, the etiological agent of sleeping sickness, has particular peroxisome-like organelles called glycosomes where several metabolic pathways are compartmentalized, including glycolysis. Many enzymes from sugar nucleotide biosynthetic pathways are located in this organelle, contrasting with the cytosolic localisation reported in the eukaryota domain. The UDP-glucose pyrophosphorylase (UGP), the enzyme responsible for UDP-glucose (UDP-Glc) production, is essential for growth and survival of trypanosomes. In addition, UGP is localized in both glycosomes and cytosol, raising questions about why would these parasites maintain a pathway functional in two different subcellular compartments. UGP is imported into glycosomes by piggybacking on the glycosomal protein PEPCK, which is the first evidence of piggybacking involving two proteins not functionally related, and the first report in trypanosomatids. The dataset demonstrates that UGP is present in the total cellular extract and the glycocomal fraction of the wild-type trypanosomes, while only in the total cellular extract of the PEPCK null mutant.
