Project description:F1 Arabidopsis thaliana plants from parents grown in absence/presence of acute/chronic Cr stress were analyzed in terms of Cr tolerance (evaluated by the half maximal effective concentration, EC50), germination, root length and hydroxide peroxide. F1 generation from parents grown in both acute and chronic stress showed significant higher levels of EC50 than F1 from unstressed parents. In addition, both F1 from Cr stressed parents showed quicker germination under Cr presence and F1 from parents cultivated under chronic stress displayed reduction of hydroxide peroxide levels under Cr stress compared to control. RNA-seq analyses were performed at 1 and 3 weeks after the beginning of stress at two levels of Cr excess for both generation (F0 and F1).

Project description:Organisms can gain information about their environment from their ancestors, their parents, or their own personal experience. “Cue integration” models often start with the simplifying assumption that information from different sources is additive. Here, we test key assumptions and predictions of cue integration theory at both the phenotypic and molecular level in threespined sticklebacks (Gasterosteus aculeatus). We show that regardless of whether cues about predation risk were provided by their father or acquired through personal experience, sticklebacks produced the same set of predator-adapted phenotypes. Moreover, there were nonadditive effects of personal and paternal experience: animals that received cues from both sources resembled animals that received cues from a single source. A similar pattern was detected at the molecular level: there was a core set of genes that were differentially expressed in the brains of offspring regardless of whether risk was experienced by their father, themselves or both. These results provide strong support for cue integration theory because they show that cues provided by parents and personal experience are comparable at both the phenotypic and molecular level, and draw attention to the importance of nonadditive responses to multiple cues.

Project description:Chromium (Cr) is a highly toxic heavy metal for both mono and dicot plants, causing severe cellular damage and disturbances in plant growth, development, and reproduction. Its phytotoxicity results in serious changes in plant physiology, gene expression, and at a transgenerational level in genomic DNA methylation. Herein, transcriptional responses of durum wheat (Triticum turgidum L.) to continuous and basal Cr stress were explored in roots and leaves by RNAseq analysis. Plants cultivated in a hydroponic system supplemented with 2.5 and 10 µM Cr were compared with control plants through transcriptomic analysis after 50 days and for the biomass accumulation and seed yield after senescence.

Project description:A number of environmental factors (e.g. toxicants) have been shown to promote the epigenetic transgenerational inheritance of disease and phenotypic variation. Transgenerational inheritance requires the germline transmission of altered epigenetic information between generations in the absence of direct environmental exposures. The primary periods for epigenetic programming of the germline is associated with primordial germ cell development and during fetal gonadal sex determination. The current study examined the actions of an agricultural fungicide vinclozolin on gestating female (F0 generation) progeny in regards to the primordial germ cell (PGC) epigenetic reprogramming of the F3 generation (i.e. great-grandchildren). The F3 generation primordial germ cell transcriptome and epigenome (DNA methylation) was altered transgenerationally. Interestingly, the differential DNA methylation regions (DMR) and altered transcriptomes were distinct between the onset of gonadal sex determination at embryonic day 13 (E13) and after cord formation in the testis at embryonic day 16 (E16). A larger number of DMR and transcriptional alterations were observed in the E13 PGC than E16 germ cells. Observations demonstrate an altered transgenerational epigenetic reprogramming and function of the primordial germ cells and subsequent male germline is a component of vinclozolin induced epigenetic transgenerational inheritance of disease. Insights into the molecular control of germline transmitted epigenetic inheritance are provided. The combined observations demonstrate ancestral exposure of a gestating female during fetal gonadal sex determination can promote transgenerational alterations in the primordial germ cell and subsequent male germline epigenetic and transcriptional programming. This altered germline programming leads to the epigenetic transgenerational inheritance of disease and phenotypic variation. Observations support the role of the primordial germ cell programming in the molecular mechanism involved and provides insights into the molecular mechanisms that control the epigenetic transgenerational inheritance phenomena. Results suggest a cascade of epigenetic and transcriptional events during germ cell development is needed to obtain the mature germline epigenome that is then transmitted transgenerationally. Project description:Environmentally induced epigenetic transgenerational inheritance of adult onset disease involves a variety of phenotypic changes suggesting a general alteration in genome activity. Investigation of eleven different tissue transcriptomes in male and female F3 generation vinclozolin versus control lineage rats demonstrated all tissues examined had unique transgenerational transcriptomes. Common cellular pathways and processes were identified among the tissues. A bionetwork analysis identified gene modules with coordinated gene expression and each had unique gene networks regulating tissue specific gene expression and function. A large number of statistically significant over-represented clusters of differentially expressed genes were identified and termed M-bM-^@M-^\Epigenetic Control RegionsM-bM-^@M-^]. Combined observations demonstrate that all tissues derived from the epigenetically altered germ line develop transgenerational transcriptomes unique to the tissue, but common epigenetic control regions in the genome appear to in part coordinately regulate these tissue specific transcriptomes. This systems biology approach provides insight into the molecular mechanisms involved in the epigenetic transgenerational inheritance of a variety of adult onset disease phenotypes. We used microarrays to determine genes expressed differentially in rats 11 male or female smatic tissues -male heart, kidney, liver, testis, prostate, seminal vesicles; female heart, kidney, liver, ovary, uterus - due to Vinclozolin treatments of their grand-grandmothers. For each of 11 male or female smatic tissues, RNA samples from 2 treatment groups - control (Con) and vinclozolin (Vin) - were compared to each other. Each treatment groups contained 3 biological replica. RNA for each replica was pooled from 2 individual animals.

Project description:A marine bacterial isolate, Brevibacterium casei #NIOSBA88 was grown separately in Luria Bertini broth supplemented with 750 mg L-1 Arsenic (III) and 100 mg L-1 Chromium respectively. Similarly, the strain was grown in absence of Arsenic and Chromium, which served as an experimental control. The cells were harvested at different time intervals and proteins were extracted from the cell pellets using in-house protocol (Shah and Damare 2018). The extracted proteins were digested using trypsin following standard protocol. Tryptic digests were injected in LCMS QToF for obtaining spectral library which was used to identify the proteins against a species specific database in NCBI.
