Project description:Leaf senescence, the last step of leaf development, is a highly regulated process, modulated by a number of internal and external factors. During the senescence process resources like nitrogen (N) are remobilized from senescent tissues to sink tissues. This intrinsically depends on the accurate dispersion of resources according to sink strength of various organs competing with each other. Consequently, N deficiency accelerates barley leaf senescence and its resupply can delay the senescence progression. In order to identify genetic and metabolic factors that regulate leaf senescence in response to N supply, transcriptomic and global metabolic rearrangements were analyzed in barley primary at early and later stages of N deprivation, and after N resupply to N-deficient plants.

Project description:Deprivation of mineral nutrients causes significant retardation of plant growth. This slow growth is assumed to be associated with both nutrient specific transcriptional responses and additionally with common transcription patterns. In this study we adjusted the external supply of iron, potassium and sulfur to cause a similar retardation of growth. Global transcriptome analyses were performed to investigate whether the growth limitation by the different nutrient deficiencies triggered specific or similar transcriptional responses. The global transcriptome responded specifically to sulfur, iron or potassium deprivation. Arabidopsis thaliana plants were grown hydroponically under short-day conditions (8h light / 16h dark cycles) under full nutrient supply or under the limitation of sulfur, iron or potassium. Arabidopsis root material was harvested when the plants reached the age of 7 weeks (from sowing) and used for RNA extraction and hybridization on Affymetrix microarrays. Four biological replicates from each condition were analyzed.

Project description:Plants will meet various abiotic stresses during their growth and development. One of the important strategies for plants to deal with the stress is involved in metabolic regulation, causing the dramatic changes of metabolite profiles. Metabolomic studies have been intensively conducted to reveal the responses of plants to abiotic stress, but most of them were limited to one or at most two abiotic stresses in a single experiment. In this study, we compared the metabolite profiles of barley seedlings exposed to seven abiotic stresses simultaneously, including drought, salt stress, aluminum (Al), cadmium (Cd), deficiency of nitrogen (N), phosphorus (P) and potassium (K). The results showed that metabolite profiles of barley under these stresses could be classified into three types: osmotic stresses (drought and salt); metal stresses (Al and Cd) and nutrient deficiencies (N, P and K deficiencies). Compared with the control, some metabolites (including polyamines, raffinose and piperonic acid) in plants exposed to all abiotic stresses changed significantly, while some other metabolites showed the specific change only under a certain abiotic stress, such as proline being largely increased by osmotic stress (drought and salinity), the P-containing metabolites being largely decreased under P deficiency, some amino acids (lysine, tyrosine, threonine, ornithine， glutamine and so on) showing the dramatic reduction in the plants exposed to N deficiencies, respectively. The current meta-analysis obtained a comprehensive view on the metabolic responses to various abiotic stress, and improved the understanding of the mechanisms for tolerance of barley to abiotic stress.

Project description:The supply of soluble silicon (Si) to plants has been associated with many benefits that remain poorly explained and often contested. In this work, the effect of Si was studied on wheat plants under both control and pathogen stress (Blumeria graminis f.sp. tritici (Bgt)) conditions by conducting an exhaustive transcriptomic analysis (55,000 genes) aimed at comparing the differential response of plants under four treatments. The response to the supply of Si on control (uninfected) plants was limited to 47 genes providing little evidence of regulation of a specific metabolic process. Plants reacted to inoculation with Bgt by an up-regulation of many genes linked to stress and metabolic processes and a down-regulation of genes linked to photosynthesis. Supplying Si to inoculated plants largely prevented disease development, a phenotypic response that translated into a nearly perfect reversal of genes regulated by the effect of Bgt alone. These results suggest that Si plays a limited role on a plant’s metabolism in absence of stress, even in the case of a high-Si accumulating monocot such as wheat. On the other hand, the benefits of Si, in the form of biotic stress alleviation, were remarkably aligned with a counter-response to transcriptomic changes induced by the pathogen Bgt. Project description:Potassium (K+) is a crucial macronutrient in high biomass plants, especially in banana.we comparatively studyed the phenotypic traits and transcriptomic profiles of banana leaves and roots between low potassium group (LK) and normal-potassium group (NK). In our study, the K+ content and biomass index of banana seedling were all significantly decreased under the stress of low potassium group. Moreover, thirty differentially expressed genes (DEGs) related to potassium transport and uptake and transcription factors were analyzed deeply. DEGs about ABC transporters, protein kinases and ion transporters were also detected, these genes may play important roles during potassium deficiency. These results provide valuable information about banana response to low potassium conditions.

Project description:Maintaining genome integrity presents a particular challenge for plants due to their sedentary lifestyle, which disables direct avoidance of unfavorable external conditions. Additionally, plants’ metabolic processes generate reactive molecules as by-products of e.g. photosynthesis, creating internal DNA-damaging conditions. Due to this, plants have developed a unique and strictly regulated web of DNA damage responses (DDR). We initiated analysis of the DDR system in cultivated barley (Hordeum vulgare), a temperate cereal model with a large and repeat rich genome. A series of DNA damaging assays was established to describe barley plants’ phenotypic response to chemically induced DNA double-strand breaks. The efficacy of assays as a tool to be used for assessing potential new DDR barley mutants was demonstrated. DNA damage response network activation in barley was assessed by transcriptome analysis using RNA sequencing for wild type and mutant in the DDR signaling kinase ATAXIA TELANGIECTASIA MUTATED AND RAD3-RELATED (ATR). Considering the barley genome had only recently been sequenced, and it lacks the in-depth gene analysis, a list of potential DNA damage response genes in barley was compiled based on their homology with Arabidopsis genes. Project description:Drought is a destructive abiotic stress, with plants possessing diverse strategies to survive periods of limited water resources. Previous results have described connections between strigolactone (SL) and drought, however, here we used the barley (Hordeum vulgare) SL-insensitive mutant hvd14 (dwarf14) to investigate the SL-dependent mechanisms related to water deficit response. By combining transcriptome, proteome with phytohormone analyses and physiological data, we describe the drought-mediated differences between wild-type and hvd14 plants. Our findings indicate that the drought sensitivity of hvd14 is related to weaker induction of abscisic acid-responsive genes/proteins, lower jasmonic acid content, higher reactive oxygen species content, and lower wax biosynthic and deposition mechanisms then wild-type plants. In addition, we identify a series of transcription factors (TFs) that are exclusively drought-induced in wild-type barley. Critically, we resolve a comprehensive series interestions between the drought-induced barley transcriptome and proteome responses that allow us to understand the impacts of SL in mitigating water limiting conditions. These data provide a number of new angles for the development of drought-resistant barley.
