Project description:The Sda histo-blood group antigen [GalNAcβ1-4(NeuAcα2-3)Galβ-R] is present in colon, kidney and body fluids among 96-98% of Europeans whilst 90% have Sda-positive erythrocytes. Sda is implicated in various infections and constitutes a potential biomarker for colon cancer. The 2-4% truly Sd(a‒) individuals may produce anti-Sda, which can lead to incompatible blood transfusions, especially if donors with the high-expressing Sda phenotype, Sd(a++) or Cad, are involved. It was hypothesized that defects in the B4GALNT2-encoded β4GalNAc-T2 glycosyltransferase underlies the null phenotype. We recently reported the association of B4GALNT2 mutations with the Sd(a‒) phenotype, which formally established the SID blood-group system. In the present study, we provide causal proof and glycoprotein profiling underpinning this correlation. Phenotypically Sd(a‒) HEK293 cells were transfected with different B4GALNT2 constructs and evaluated by immunostaining and LC-MS/MS-based glycoproteomics. The pre¬dominant SIDnull allele with SNP rs7224888:T>C (p.Cys406Arg) abolished Sda synthesis, while this antigen was detectable as N- or O-glycans on multiple glycoproteins following transfection of wildtype B4GALNT2. Surprisingly, two rare missense SNPs, rs148441237:A>G and rs61743617:C>T, found in a Sd(a‒) compound heterozygote gave results similar to wildtype. To elucidate if Sd(a++)/Cad is also due to B4GALNT2 alterations, its coding region and 2 kbp upstream were sequenced in five Cad individuals. No genetic changes were associated with this phenotype but a detailed erythroid Cad glycoprotein profile was obtained, especially for GLPA (O-glycosylation) and, for the first time, B3AT (N-glycosylation). In conclusion, the p.Cys406Arg β4GalNAc-T2 variant causes Sda-deficiency in humans, while the enigmatic Cad phenotype remains unresolved, albeit further characterized.

Project description:To increase our knowledge of the effects of Fructo oligosaccharides (FOS) on Salmonella infection in fats, a controlle rat infection study was performed. Two groups of 12 rats were adapted for 14 days to a cellulose diet and one group of 12 rats to a FOS diet. One cellulose-fed group and the FOS-fed group were infected with Salmonella. Two days post infection mRNA was collected from the mucosa of the colon and changes in gene expression were assessed using an Agilent rat whole genome microarray (G4131A Agilent Technologies). Project description:Protein turnover affects protein abundance and phenotypes. Comprehensive investigation of protein turnover dynamics has the potential to provide substantial information about gene expression. Here we report a large-scale protein turnover study in Salmonella Typhimurium during infection by quantitative proteomics. Murine macrophage-like RAW 264.7 cells were infected with SILAC labeled Salmonella. Bacterial cells were extracted after 0, 30, 60, 120 and 240 min. Mass spectrometry analyses yielded information about Salmonella protein turnover dynamics and a software program named Topograph was used for the calculation of protein half lives. The half lives of 311 proteins from intracellular Salmonella were obtained. For bacteria cultured in control medium (DMEM), the half lives for 870 proteins were obtained. The calculated median of protein half lives was 69.13 min and 99.30 min for the infection group and the DMEM group respectively, indicating an elevated protein turnover at the initial stage of infection. Gene ontology analyses revealed that a number of protein functional groups were significantly regulated by infection, including proteins involved in ribosome, periplasmic space, cellular amino acid metabolic process, ion binding and catalytic activity. The half lives of proteins involving in purine metabolism pathway were found to be significantly shortened during infection.

Project description:Poultry products are an important source of Salmonella enterica. An effective way to reduce food poisoning due to Salmonella would be to breed chickens more resistant to Salmonella. Unfortunately resistance to Salmonella is a complex trait with many factors involved. To learn more about Salmonella resistance mechanisms in young chickens, a cDNA microarray analysis was performed to compare gene expression profiles between a Salmonella susceptible and a more resistant chicken line. Newly hatched chickens were orally infected with S. enterica serovar Enteritidis. Since the intestine is the first barrier the bacteria encountersbacteria encounter after oral inoculation, gene expression was investigated in the intestine, from day 1 until day 21 post infection. Differences in gene expression between the susceptible and resistant chicken line were found in control as well as Salmonella infected conditions. In response to the Salmonella infection, the expression of different sets of genes seemed to be affected in the jejunum of the two chicken lines. In the susceptible line this included genes that affect T-cell activation, whereas in the more resistant line, at day 1, macrophage activation seemed to be more affected. At day 7 and 9 most gene expression differences between the two chicken lines were identified under control conditions, indicating a difference in the intestinal development between the two chicken lines which might be linked to the difference in Salmonella susceptibility. The findings in this study have lead to the identification of novel genes and possible cellular pathways of the host involved in Salmonella susceptibility. Keywords: timecourse, disease

Project description:Environmental stress contributes to the outcome of infection by impacting both microbial virulence and host susceptibility to infection. Thermal processing, commonly used for decontamination of poultry in the food industry, may elicit sublethal stress on resistant serovars of Salmonella. We employed traditional heat shock temperatures (42 and 48ºC), similar to avian body temperature and poultry processing conditions, to study gene expression of Salmonella enterica serovar Typhimurium. Microarray analysis indicated that thermal shock at 42°C and 48°C induced expression of SPI-2 and SPI-5 genes, whose products are required for adhesion and survival. However, SPI-1 genes, responsible for invasion of Salmonella into host cells, were down-regulated following exposure to 42°C and 48°C. Bacterial adhesion assays showed greater adhesion of heat-stressed S. Typhimurium to Caco-2 cells compared to non-stressed bacteria. In addition, subjecting Caco-2 cells to mild heat shock (39°C), which is similar to human fever, enhanced host cell susceptibility to bacterial adhesion. Data indicate that thermal stress enhances bacterial colonization and host cell susceptibility to adhesion during S. Typhimurium infection.

RNA was isolated from colonic mucosal scrapings. mRNA samples of 12 rats per group were pooled. Each group-sample was hybridised in duplicate on Agilent rat whole genome microarrays containing 44290 60-mer spots.

Abstract:
BACKGROUND: Infections with Salmonella cause significant
morbidity and mortality worldwide. Replication of Salmonella
typhimurium inside its host cell is a model system for studying
the pathogenesis of intracellular bacterial infections.
Genome-scale modeling of bacterial metabolic networks provides
a powerful tool to identify and analyze pathways required for
successful intracellular replication during host-pathogen
interaction. RESULTS: We have developed and validated a
genome-scale metabolic network of Salmonella typhimurium LT2
(iRR1083). This model accounts for 1,083 genes that encode
proteins catalyzing 1,087 unique metabolic and transport
reactions in the bacterium. We employed flux balance analysis
and in silico gene essentiality analysis to investigate growth
under a wide range of conditions that mimic in vitro and host
cell environments. Gene expression profiling of S. typhimurium
isolated from macrophage cell lines was used to constrain the
model to predict metabolic pathways that are likely to be
operational during infection. CONCLUSION: Our analysis suggests
that there is a robust minimal set of metabolic pathways that
is required for successful replication of Salmonella inside the
host cell. This model also serves as platform for the
integration of high-throughput data. Its computational power
allows identification of networked metabolic pathways and
generation of hypotheses about metabolism during infection,
which might be used for the rational design of novel
antibiotics or vaccine strains.

Project description:Knockout mice were generated that harbour a mutation in the gene encoding gastric intrinsic factor (GIF) a transporter protein necessary for the absorption of vitamin B12/cobalamin (Cbl) from the intestine. The levels of Cbl in the blood of these gif-/- mice exibited a maternal influence dependent on whether they were bred from heterozygous (high Cbl) or homozygous (low Cbl) mothers and this correlated with their susceptibility to infection challenge with Salmonella enterica Serovar Typhimurium (S.Typhimurium). Microarrray analysis was conducted on total messenger RNA (mRNA) extracted fron the homzygous-bred gif-/- and wild-type mice with the aim to identify any abnormal genomic expression patterns.

Project description:Salmonella enteritidis is suggested to translocate in the small intestine. Previously we identified that prebiotics, fermented in the colon, increased Salmonella translocation in rats, suggesting involvement of the colon in translocation. Effects of Salmonella on colonic gene expression in vivo are largely unknown. The aim of this study was to characterize time dependent Salmonella induced changes of colonic mucosal gene expression in rats using whole genome microarrays. Rats were orally infected with Salmonella enteritidis to mimic a foodbore infection and colonic gene expression was determined at day 1, 3 and 6 post-infection (n=8 per timepoint). Agilent rat whole genome microarray (G4131A Agilent Technologies) were used. Results indicate that colon is clearly a target tissue for Salmonella considering the abundant changes in mucosal gene expression observed. Experiment Overall Design: In the present study, large-scale gene expression analysis was performed to reveal whether Salmonella induced changes of colonic mucosal gene expression in rats. Wistar rats were infected with Salmonella enteritidis. Non-infected control rats were sham-treated. Rats were sacrificed on day 1, 3 or 6 post infection or sham-treatment (n=8 rats per treatment and per time point). RNA was isolated from colonic mucosal scrapings. mRNA samples of 8 rats per group were pooled. Each pooled group-sample was hybridised in duplicate on Agilent rat whole genome microarrays containing 44290 60-mer spots. From the 12 arrays one duplicate array (Colon mucosa non-infected day6) did not pass quality control and was left out from further analysis.
