Project description:Increased root H+ secretion is known as a strategy of plant adaption to low phosphorus (P) stress by enhancing mobilization of sparingly soluble P-sources. However, it remains fragmentarywhether enhanced H+ exudation could reconstruct the plant rhizosphere microbial community under low P stress. The present study found that P deficiency led to enhanced H+ exudation from soybean (Glycine max) roots. Three out of all eleven soybean H+-pyrophosphatases (GmVP) geneswere up-regulated by Pi starvation in soybean roots. Among them, GmVP2 showed the highest expression level under low P conditions. Transient expression of a GmVP2-green fluorescent protein chimera in tobacco (Nicotiana tabacum) leaves, and functional characterization of GmVP2 in transgenic soybean hairy roots demonstrated that GmVP2 encoded a plasma membrane transporter that mediated H+ exudation. Meanwhile, GmVP2-overexpression in Arabidopsis thaliana resulted in enhanced root H+ exudation, promoted plant growth, and improved sparingly soluble Ca-P utilization. Overexpression of GmVP2 also changed the rhizospheric microbial community structures, as reflected by a preferential accumulation of acidobacteria in the rhizosphere soils. These results suggested that GmVP2 mediated Pi-starvation responsive H+ exudation,which is not only involved in plant growth and mobilization of sparingly soluble P-sources, but also affects microbial community structures in soils.

Project description:Most land plants form beneficial associations with arbuscular mycorrhizal (AM) fungi which improves mineral nutrition, mainly phosphorus and nitrogen in the host plant in exchange for photosynthetically fixed carbon. Most of our knowledge on the AM symbiosis derives from dicotyledonous species. We show that inoculation with the AM fungus Funneliformis mossease stimulates growth and increases Pi content in leaves of the rice cultivar Loto (O. sativa ssp japonica). Although rice is a host for AM fungi, the molecular mechanism underlying the AM symbiosis, in particular the systemic transcriptional responses of shoots to AM inoculation, remain largely elusive. Transcript profiling of the shoots indicated the systemic induction of genes involved in the biosynthesis of phospholipids (phosphoinositides, inositol polyphosphates) and down-regulation of non-phosphorus lipids (galactolipids, sulfolipids) in leaves of mycorrhizal rice. Regulation of phospholipid biosynthesis genes appears to be coordinated with a reduced expression of genes involved in jasmonic acid and ethylene biosynthesis and signaling. Genes involved in phosphate starvation responses and remobilization of Pi were also found to be down-regulated in leaves of mycorrhizal rice. These results demonstrated that the AM symbiosis is accompanied by complex alterations in gene expression in shoots which are potentially important to maintain a stable symbiotic relationship in rice plants.

Project description:The microbial community in the rhizosphere environment is critical for the health of land plants and the processing of soil organic matter. The objective of this study was to determine the extent to which rice plants shape the microbial community in rice field soil over the course of a growing season. Rice (Oryza sativa) was cultivated under greenhouse conditions in rice field soil from Vercelli, Italy and the microbial community in the rhizosphere of planted soil microcosms was characterized at four plant growth stages using quantitative PCR and 16S rRNA gene pyrotag analysis and compared to that of unplanted bulk soil. The abundances of 16S rRNA genes in the rice rhizosphere were on average twice that of unplanted bulk soil, indicating a stimulation of microbial growth in the rhizosphere. Soil environment type (i.e., rhizosphere versus bulk soil) had a greater effect on the community structure than did time (e.g., plant growth stage). Numerous phyla were affected by the presence of rice plants, but the strongest effects were observed for Gemmatimonadetes, Proteobacteria, and Verrucomicrobia. With respect to functional groups of microorganisms, potential iron reducers (e.g., Geobacter, Anaeromyxobacter) and fermenters (e.g., Clostridiaceae, Opitutaceae) were notably enriched in the rhizosphere environment. A Herbaspirillum species was always more abundant in the rhizosphere than bulk soil and was enriched in the rhizosphere during the early stage of plant growth.

Photosynthetic electron-transport chain switches to use molecular oxygen as an electron carrier, generating photo-reactive oxygen species (photo-ROS) in chloroplast. In rice (Oryza sativa), DEEP GREEN PANICLE1 (DGP1) is robustly up-regulated in response to Pi-deficiency stress. DGP1 decreases the DNA-binding activities of the photosynthetic activators GLK1/2 on the genes involved in chlorophyll biosynthesis, light harvesting and electron transport. This Pi-starvation-induced mechanism dampens electron transport rates (ETRI and ETRII) and alleviates the electron-excessive stress in mesophyll cells. Meanwhile, DGP1 hijacks glycolytic enzymes GAPC1/2/3, redirecting glucose metabolism toward pentose phosphate pathway with superfluous NADPH production. Phenotypically, light irradiation induces O2– accumulation in Pi-starved WT leaves, but was observably accelerated in dgp1 mutant and impaired in GAPCsRNAi line and glk1glk2 double mutant. Interestingly, overexpression of DGP1 in rice caused hyposensitivity to the ROS-inducers (catechin and methyl viologen) and dgp1 mutant shows a similar inhibitory growth with the WT plants. We conclude that DGP1 gene serves as a specific antagonizer against Pi-starvation-induced photo-ROS, which integrates light-absorbing and anti-oxidative systems by orchestrating transcriptional and metabolic regulations, respectively.

Project description:Rice has developed several morphological and physiological strategies to adapt to phosphate starvation stress. In order to elucidate the molecular bases of response due to phosphate stress particularly the transcriptional profile of genotypes with variation in tolerance to phosphate starvation, we analyzed the gene expression profiles of 3 japonica rice cultivars and an indica cultivar with different levels of tolerance to phosphate starvation using the RNA-Seq method. We constructed a total of 48 libraries corresponding to root and shoot of the 4 cultivars for control (0 d) and âP treatment (22 d) with three biological replicates for root and shoot samples in each cultivar. Approximately 254 million sequenced tags were mapped onto the reference rice genome sequence (IRGSP1.0) and an average of about 5,000 transcripts in each genotype were found to be responsive to Pi-starvation. Comparative analysis of the responsive transcripts revealed an overall similarity in the transcriptome signatures resulting from phosphate starvation as well as distinct differences that distinguish the degree of tolerance among the 4 cultivars. We elucidated a set of core responsive transcripts commonly expressed in both root and shoot with different levels of expression reflecting variation as well genotype specificity in tolerance to Pi-starvation. De novo assembly of unmapped reads generated a large set of sequence assemblies that represent potential new transcripts that may be involved in tolerance to phosphate deficiency. Characterization of 88 assemblies unaligned in the reference genome revealed several dozen transcripts which correspond to plant ESTs. This study provides an overview of the diversity in response to P-starvation stress that can be used to identify the major genes for improving Pi acquisition and utilization in rice and other cereal crops. Project description:OsPI1 was isolated from rice and revealed to belong to the TPSI1/Mt4 family, a phosphorus deficiency responsive gene family. The importance of this family gene for phosphorus deficiency responses of plants had been already reported, though its actual role is still unclear. To understand the plants response mechanisms for phosphorus deficiency and the role of OsPI1 in it, we constructed a OsPI1 RNAi line (ospi1-2) for this study. We used the 22K rice Oligoarray from Agilent technologies to study the transcript profile in the shoots and roots of ospi1-2 and Oryza sativa L. cv. Project description:This study aims to deal with a comparative proteome analysis of shoot and root tissues of contrasting phosphorus (P) responsive rice genotypes (Pusa-44 and its near-isogenic line (NIL)-23 harboring Pup1 QTL). Proteins were isolated from shoot and root tissues collected from 45-day-old rice plants grown hydroponically in PusaRicH medium with P (16ppm, +P) or without P (0 ppm, -P) under controlled environmental conditions. Following protein quantification using the Bradford method, and quality check using 1D SDS-PAGE, trypsin was used for in-solution digestion and Nano ACQUITY UPLC-MS/MS was used to separate and identify peptides. This study aimed at deciphering the molecular aspect of Pup1 QTL in P-starvation stress tolerance in rice.
