Project description:In this study, adult male rainbow darter (Etheostoma caeruleum) were collected from nine sites located at different distances from two wastewater treatment plants (WWTPs) in the Grand River, Ontario, Canada. The goal of this project was to see if the liver transcriptome of fish residing downstream of the two WWTPs differs from the non-exposed fish. Further, this research was also addressing the affects of differnent reference site selection on gene expression patterns, and their subsequent interpretation.The RNA from male rainbow darter was exctracted from liver tissue and used for further gene expression analysis.

Project description:In this study, adult male rainbow darter were collected from three sites in the Grand River in the fall season for four consecutive years (2011-2014), from 3 sites: a reference site (US2) located in the non-urbanized area, and two exposure sites (DSW and DSK1) located dowstrem of two secondary wastewater treatment palnts (WWTP) (Waterloo and Kitchener). Kitchener WWTP has been undergone infrastructure upgrades that were initiated in the summer of 2012. The goal of this project was to see whether the implemented upgrades can be reflected in the liver transcriptome response indicative of recovery.The RNA from male rainbow darter was exctracted from liver tissue and used for further gene expression analysis.

Project description:Ecological flow is an important indicator for reflecting the stability of a watershed ecosystem. The calculation of ecological discharge under hydrological variation has become a research hot-spot. The Ganjiang River south of Poyang Lake in China was taken as an example in this study. Hydrological Alteration Diagnosis System methods were used to detect the change-points. The Distributed Time Variation Gain Model (DTVGM) was used to carry out runoff restoration. The Probability-weighted Flow Duration Curve was applied to calculate the ecological flow. The results showed that: (1) The hydrological alteration of the Waizhou Station occurred in 1991, the annual runoff increased by 10%, and the Gini coefficient (GI) increased by 0.07 after the change-point. The change in precipitation was the main driving factors. (2) The R value and NSE of the DTVGM were greater than 0.84, which represents the feasibility of the model used to restore runoff. (3) Compared to the traditional hydrological method, the proposed method can better reflect the inter-annual difference of ecological flow, flow ranges for high, normal, and low flow years are 398-3771 m3/s, 352-2160 m3/s, and 277-1657 m3/s, respectively. The calculation method of ecological flow in rivers considering hydrological variation can more scientifically reflect the impact of hydrological variation on ecological flow process, ecological flow under different human activities that can be calculated, such as dam control, water intake and water transfer, furthermore, it also provides a scientific basis for water resources planning and allocation under changing environment.

The small RNA profile is unique for each plant tissue at a given point of time. High throughput sequencing method allows the study of differential expression of these small RNAs. Project description:This study aimed to determine the variability of carbon and nitrogen elemental content, stoichiometry and diet proportions of invertebrates in two sub-tropical estuaries in South Africa experiencing seasonal changes in rainfall and river inflow. The elemental ratios and stable isotopes of abiotic sources, zooplankton and macrozoobenthos taxa were analyzed over a dry/wet seasonal cycle. Nutrient content (C, N) and stoichiometry of suspended particulate matter exhibited significant spatio-temporal variations in both estuaries, which were explained by the variability in river inflow. Sediment particulate matter (%C, %N and C:N) was also influenced by the variability in river flow but to a lesser extent. The nutrient content and ratios of the analyzed invertebrates did not significantly vary among seasons with the exception of the copepod Pseudodiaptomus spp. (C:N) and the tanaid Apseudes digitalis (%N, C:N). These changes did not track the seasonal variations of the suspended or sediment particulate matter. Our results suggest that invertebrates managed to maintain their stoichiometry independent of the seasonality in river flow. A significant variability in nitrogen content among estuarine invertebrates was recorded, with highest % N recorded from predators and lowest %N from detritivores. Due to the otherwise general lack of seasonal differences in elemental content and stoichiometry, feeding guild was a major factor shaping the nutrient dynamics of the estuarine invertebrates. The nutrient richer suspended particulate matter was the preferred food source over sediment particulate matter for most invertebrate consumers in many, but not all seasons. The most distinct preference for suspended POM as a food source was apparent from the temporarily open/closed system after the estuary had breached, highlighting the importance of river flow as a driver of invertebrate nutrient dynamics under extreme events conditions. Moreover, our data showed that estuarine invertebrates concentrated C and N between 10-100 fold from trophic level I (POM) to trophic level II (detritivores/deposit feeders) and thus highlighted their importance not only as links to higher trophic level organisms in the food web, but also as providers of a stoichiometrically homeostatic food source for such consumers. As climate change scenarios for the east coast of South Africa predict increased rainfall as a higher number of rainy days and days with higher rainfall, our results suggest that future changes in rainfall and river inflow will have measurable effects on the nutrient content and stoichiometry of food sources and possibly also in estuarine consumers.

Project description:IntroductionThe fractional concentration of exhaled nitric oxide (FeNO) is a biomarker of airway inflammation. Repeat FeNO maneuvers at multiple fixed exhalation flow rates (extended NO analysis) can be used to estimate parameters quantifying proximal and distal sources of NO in mathematical models of lower respiratory tract NO. A growing number of studies use extended NO analysis, but there is no official standard flow rate sampling protocol. In this paper, we provide information for study planning by deriving theoretically optimal flow rate sampling designs.MethodsFirst, we reviewed previously published designs. Then, under a nonlinear regression framework for estimating NO parameters in the steady-state two compartment model of NO, we identified unbiased optimal four flow rate designs (within the range of 10-400 ml s-1) using theoretical derivations and simulation studies. Optimality criteria included NO parameter standard errors (SEs). A simulation study was used to estimate sample sizes required to detect associations with NO parameters estimated from studies with different designs.ResultsMost designs (77%) were unbiased. NO parameter SEs were smaller for designs with: more target flows, more replicate maneuvers per target flow, and a larger range of target flows. High flows were most important for estimating alveolar NO concentration, while low flows were most important for the proximal NO parameters. The Southern California Children's Health Study design (30, 50, 100 and 300 ml s-1) had ≥1.8 fold larger SEs and required 1.1-3.2 fold more subjects to detect the association of a determinant with each NO parameter as compared to an optimal design of 10, 50, 100 and 400 ml s-1.ConclusionsThere is a class of reasonable flow rate sampling designs with good theoretical performance. In practice, designs should be selected to balance the tradeoffs between optimality and feasibility of the flow range and total number of maneuvers.

Project description:The Grand River (Ontario, Canada) is impacted by wastewater treatment plants (WWTPs) that release ammonia (NH3 and NH4+) into the river. In-river microbial communities help transform this ammonia into more oxidized compounds (e.g., NO3- or N2), although the spatial distribution and relative abundance of freshwater autotrophic ammonia-oxidizing prokaryotes (AOP) are not well characterized. This study investigated freshwater N cycling within the Grand River, focusing on sediment and water columns, both inside and outside a WWTP effluent plume. The diversity, relative abundance, and nitrification activity of AOP were investigated by denaturing gradient gel electrophoresis (DGGE), quantitative real-time PCR (qPCR), and reverse transcriptase qPCR (RT-qPCR), targeting both 16S rRNA and functional genes, together with activity assays. The analysis of bacterial 16S rRNA gene fingerprints showed that the WWTP effluent strongly affected autochthonous bacterial patterns in the water column but not those associated with sediment nucleic acids. Molecular and activity data demonstrated that ammonia-oxidizing archaea (AOA) were numerically and metabolically dominant in samples taken from outside the WWTP plume, whereas ammonia-oxidizing bacteria (AOB) dominated numerically within the WWTP effluent plume. Potential nitrification rate measurements supported the dominance of AOB activity in downstream sediment. Anaerobic ammonia-oxidizing (anammox) bacteria were detected primarily in sediment nucleic acids. In-river AOA patterns were completely distinct from effluent AOA patterns. This study demonstrates the importance of combined molecular and activity-based studies for disentangling molecular signatures of wastewater effluent from autochthonous prokaryotic communities.
