Project description:Persistent free-running circannual (circa 1-year) rhythms have evolved in animals to regulate hormone cycles and time annual reproduction. In mammals, these are synchronized by environmental photoperiod and the melatonin signal. Long summer-like photoperiods (LP) activates thyrotrophs and thyroid-stimulating hormone (TSH) expression in the melatonin target tissue, the pars tuberalis (PT) region of the pituitary gland, driven by the transcriptional co-activator EYA3. TSH in turn stimulates thyroid hormone (TH) conversion in hypothalamic ependymal tanycytes, thus timing breeding seasons. In contrast to our new knowledge defining photoperiodic input, it is still not known which cells, tissues and pathways generate the underlying circannual cycle. We used seasonal sheep to characterise the circannual cycle, transferring animals from short winter-like photoperiods (SP) to prolonged LP conditions, and assaying prolactin to track circannual phase in individual animals. Using RNA-seq, we profiled PT cells at different phases of the circannual cycle, and defined extensive changes of cellular-remodeling pathways and genes encoding synaptic guidance proteins in the PT.

Project description:Across evolutionary time, nearly all animal species have harnessed photoperiod to initiate processes that ultimately influence seasonal behavior and life history traits. In the freshwater cladoceran Daphnia magna, the effect of photoperiod on various life history traits has generally been investigated in conjunction with additional environmental stimuli. In the present study, we sought to untangling responses directly attributable to photoperiod in D. magna and identify the molecular processes underlying resultant behavioral and life history responses using functional analysis of global transcriptomic expression. D. magna were exposed to five different photoperiods immediately post-hatch for 21d where a standard long-day photoperiod of 16 hours light and 8 hours dark (16L:8D) served as the control relative to 4L:20D, 8L:16D, 12L:12L, and 20L:4D photoperiods. Entrainment to short-day photo-periods (4L:20D, 8L:16D, and 12L:12L) resulted in significantly increased light-avoidance behaviors relative to the control photoperiod where young Daphnia (7d old) displayed the most pronounced avoidance responses. Correspondingly, functional transcriptomics identified differential transcriptional expression of genes involved in glutamate signaling, which is critical in arthropod light-avoidance responses, as well as period circadian protein and proteins coding F-box/LRR-repeat domains, all of which contribute to establishing circadian rhythms in arthropods. Short-day photoperiods also induced increased metabolic rates which corresponded with broad-scale changes in transcriptional expression across multiple systems-level energy metabolism pathways. The most striking observation was increased male production across short-day photoperiods (4L:20D, 8L:16D, and 12L:12D). Transcriptional expression consistent with multiple putative mechanisms of male production were observed including expression suggestive of increased glutamate signaling; a response observed to induce male production in D. pulex via photo-period sensitive mechanisms. Overall, the results demonstrate the importance of photoperiod on behavior and life history trajectories in D. magna where we have now established multiple putative mechanistic pathways underlying several critical responses.

Project description:This experiment was set up to evaluate the effect of the duration of a short photoperiod (winter-like conditions, 8 hours light, 14 hours dark) on the smolt transcriptome in liver and gill. Fish were kept at constant light before fish were split between treatment tank or control tanks. Treatment tanks either experienced 2 weeks of short photoperiod or 8 weeks of short photoperiod. The control fish were kept in constant winter-like conditions. After winter, the fish were brought back to constant light for 4 weeks. Sampling was done before transition to short photoperiod, at the end of winter period and four weeks after transition back to constant light. Control fish were sampled at the same time points as the treatment group.

Project description:MicroRNAs (miRNAs) are single strand small non-coding RNAs that regulate target mRNAs at post-transcription level. Winter wheat (Triticum aestivum L.), is an important crop plant all over the world. Long term cold exposure (vernalization) is necessary for winter wheat transition from vegetative growth to reproductive growth, yet the involvement of miRNAs in these stages remains unknown. Therefore, we performed next generation sequencing of small RNAs profiles in crown tissues at three-leaf stage, winter dormancy stage, spring greenup stage and jointing stage.

Project description:The circadian clock synchronizes metabolic and behavioral cycles with the rotation of the Earth by integrating environmental cues, such as light. Nutrient content also regulates the clock, though how and why this environmental signal affects the clock remains incompletely understood. Here, we elucidate a role for nutrient in regulating circadian alignment to seasonal photoperiods. High fat diet (HFD) promoted entrainment to a summer light cycle and inhibited entrainment to a winter light cycle by phosphorylating PER2 on serine 662. PER2-S662 phospho-mimetic mutant mice were incapable of entraining to a winter photoperiod, while PER2-S662 phospho-null mutant mice were incapable of entraining to a summer photoperiod, even in the presence of HFD. Multi-omic experimentation in conjunction with isocaloric hydrogenated-fat feeding, revealed a role for polyunsaturated fatty acids in nutrient-dependent seasonal entrainment. Altogether, we identify the mechanism whereby nutrient content shifts circadian rhythms to anticipate seasonal photoperiods in which that nutrient state predominates.

Project description:This study investigated the molecular mechanisms involved in ovarian transcriptomic responses in Nile tilapia under different photoperiod regimes. Histological analysis indicated that ovarian development was significantly affected by photoperiod. The photoperiods tested were as follows: LD (12 h light:12 h dark), LL (24 h light:0 h dark), and DD (0 h light:24 h dark). The longer photoperiod (LL) was shown to induce ovary development earlier than LD and DD. Next, ovary transcriptome levels were sequenced and analyzed. These data indicated that 988, 992, and 1,036 differentially expressed genes (DEGs) were detected by comparing LD-LL, LD-DD, and LL-DD. A number of genes that may be involved in photoperiod-specific regulation of ovarian development were observed. These findings may be useful for investigating the molecular mechanisms underlying light-induced ovarian development in Nile tilapia.
