Project description:In order to investigate how chronic steroid treatment affects mouse skin at molecular levels, we shaved the back hair of 7 week-old female C57BL/6 mice and applied steroid (5 μg fluocinolone acetonide in 200 μl acetone) or vehicle once every three days for 12 days and then collected the skin samples 1, 15, or 30 days later for transcriptomic analysis.

Project description:Joint pain, either traumatic or degenerative, is a common cause of morbidity. Triamcinolone acetonide (TA), a common glucocorticoid, has been used as symptomatic treatment for joint pain for decades. However, besides its relatively short-lived therapeutic effects, TA may have potentially devastating effects by altering the normal healing process to restore and maintain the cellular entities that contribute to tissue regeneration of the synovial joint. Mesenchymal stem cells (MSCs) have been shown to hold great potential in tissue regeneration due to their multipotency in regenerating osteoblasts, chondrocytes and adipocytes and their immunomodulatory properties. So far, it remains largely unclear how TA treatment affects MSC activities.

Transcriptomic analysis of early-stage basal cell carcinomas in murine skin following ablative fractional laser and vismodegib treatment

Project description:Recent studies have demonstrated that ablative fractional laser (AFL) can inhibit the hedgehog pathway, enhance immune infiltration, and clear basal cell carcinomas (BCCs) in murine models. In this study, we applied RNA sequencing to further characterize the impact of AFL on the transcriptome of murine skin containing early-stage microscopic BCCs, contrasting it with the effects of the FDA approved hedgehog inhibitor vismodegib. Our results showed that BCC induction in murine skin was primarily linked to gene upregulation. Both AFL and vismodegib treatments were able to reverse this upregulation with vismodegib demonstrating superior performance by reversing the most genes (AFL: 59/277; vismodegib 180/277). Surprisingly, Ingenuity Pathway Analysis revealed that both treatments also caused considerable immune cell infiltration. Based on gene set enrichment analysis and cell type deconvolution, AFL treatment resulted in the largest immune cell recruitment, which for both treatments primarily consisted of infiltrating neutrophils, macrophages, and monocytes. In conclusion, the distinct effects observed in BCC skin following AFL and vismodegib treatment highlight key differences between the two interventions. Future applications of AFL or vismodegib treatments could leverage their individual effects, for example by combining AFL’s effects on the immune system with other topical treatments.

Genetic Effects on the Skin Methylome in Healthy Older Twins

Project description:Whole skin DNA methylation variation has been implicated in several diseases, including melanoma, but its genetic basis has not yet been characterised. Using bulk skin tissue samples from 414 healthy female UK twins, we performed twin-based heritability and methylation Quantitative Trait Loci (meQTL) analyses for >400,000 DNA methylation sites. We find that the human skin DNA methylome is on average less heritable than previously estimated in blood and other tissues (mean heritability: 10.02%). MeQTL analysis identified local genetic effects influencing DNA methylation at 18.8% (76,442) of tested CpG sites, as well as 1,775 CpG sites associated with at least one distal genetic variant. As a functional follow-up we performed skin expression QTL (eQTL) analyses in a partially overlapping sample of 604 female twins. Colocalisation analysis identified over 3,500 shared genetic effects affecting thousands of CpG sites (10,067) and genes (4,475). Mediation analysis of putative colocalised gene-CpG pairs identified 114 genes with evidence for eQTL effects being mediated by DNA methylation in skin, including in genes implicated skin disease such as ALOX12 and CSPG4. We further explored the relevance of skin meQTLs to skin disease, and found that skin meQTLs and CpGs under genetic influence were enriched for multiple skin-related genome-wide and epigenome-wide association signals, including for melanoma and psoriasis. Our findings give insights into the regulatory landscape of epigenomic variation in skin.
