Altered Faecal Microbiota Composition and Structure of Ghanaian Children with Acute Gastroenteritis.

Project description:Acute gastroenteritis (AGE) is a disease of global public health importance. Recent studies show that children with AGE have an altered gut microbiota relative to non-AGE controls. Yet, how the gut microbiota differs in Ghanaian children with and without AGE remains unclear. Here, we explore the 16S rRNA gene-based faecal microbiota profiles of Ghanaian children five years of age and younger, comprising 57 AGE cases and 50 healthy controls. We found that AGE cases were associated with lower microbial diversity and altered microbial sequence profiles relative to the controls. The faecal microbiota of AGE cases was enriched for disease-associated bacterial genera, including Enterococcus, Streptococcus, and Staphylococcus. In contrast, the faecal microbiota of controls was enriched for potentially beneficial genera, including Faecalibacterium, Prevotella, Ruminococcus, and Bacteroides. Lastly, distinct microbial correlation network characteristics were observed between AGE cases and controls, thereby supporting broad differences in faecal microbiota structure. Altogether, we show that the faecal microbiota of Ghanaian children with AGE differ from controls and are enriched for bacterial genera increasingly associated with diseases.

Project description:The differences of clinical characteristics in complex seizures induced by influenza A(H1N1)pdm09 and rotavirus gastroenteritis are well known, but the pathogenic mechanisms remain unclear. We analyzed the gene expression profiles in the peripheral whole blood cells isolated from pediatric patients using an Affymetrix oligonucleotide microarray. Results provide insights into the difference of the pathogenesis in the patients with complex seizures induced by influenza A(H1N1)pdm09 and rotavirus infections. The gene expression profiles in the peripheral whole blood of ten patients (n=5; complex seizures, n=5; control) with influenza A(H1N1)pdm09 and six patients (n=3; complex seizures, n=3; control) with rotavirus gastroenteritis were examined. Whole blood samples were collected from patients in the acute phase of the disease and in the recovery phase.

Project description:In small animals, diarrhoea is a regular presentation, the aetiology of which can be varied, from an isolated self-limiting event to more serious episodes requiring symptomatic and supportive treatment. Chronic diarrhoea, defined as diarrhoea lasting longer than 3 weeks, is a relatively common problem often prompting referral for more detailed investigation. Although the potential cause of chronic diarrhoea includes chronic intestinal parasitism and alimentary neoplasia (specifically lymphoma), the majority of these dogs will be diagnosed with idiopathic canine chronic enteropathy (CCE), previously referred to as idiopathic inflammatory bowel disease (IBD). Of those cases diagnosed with CCE, further subdivision based on response to treatment trials leads to a final diagnosis of antibiotic responsive diarrhoea (ARD), food responsive diarrhoea (FRD) or idiopathic inflammatory bowl disease (IBD). The aim of this study was to compare the proteomes of faeces of dogs with chronic enteropathies and a population of healthy dogs, and furthermore to identifying proteins that might be useful in distinguishing FRD from ARD from IBD.
