Project description:The opiate system has long been implicated in the rewarding properties of social interactions. In particular, the μ-opioid receptor (MOR) mediates multiple forms of social attachment, including the attachment of offspring to the mother and social bonding between mates. We have previously shown that MOR in the caudate-putamen is involved in partner preference formation in monogamous prairie voles. Here, using in situ hybridization and receptor autoradiography, we mapped in detail the distribution of MOR mRNA and ligand binding in monogamous prairie vole brains and compared MOR binding density with that of promiscuous meadow vole brains. Comparison of MOR binding in these closely related species with distinctly different social behavior revealed that while the distribution of MOR is similar, prairie voles have significantly higher densities of MOR than meadow voles in a majority of regions in the forebrain, including the caudate-putamen, nucleus accumbens shell, lateral septum and several thalamic nuclei, including the anteroventral and anteromedial thalamic nuclei. These differences in MOR expression between prairie and meadow voles could potentially contribute to species differences in behavior, including social attachment.

Project description:Wood mice of the genus Apodemus are an essential component of small mammal communities throughout Europe. Molecular data suggest the postglacial colonization of current ranges from south European glacial refugia, different in particular species. Yet, details on the course of colonization and Holocene history of particular species are not available, partly because of a lack of reliable criteria for species identification in the fossil record. Using a sample of extant species, we analyzed variation patterns and between-species overlaps for a large set of metric and non-metric dental variables and established the criteria enabling the reliable species identification of fragmentary fossil material. The corresponding biometrical analyses were undertaken with fossil material of the genus (2528 items, 747 MNI) from 22 continuous sedimentary series in the Czech Republic and Slovakia, from LGM to Recent. In Central Europe, the genus is invariantly absent in LGM assemblages but regularly appears during the Late Vistulian. All the earliest records belong to A. flavicollis, the species clearly predominating in the fossil record until the Late Holocene. A. uralensis accompanied it in all regions until the late Boreal when disappeared from the fossil record (except for Pannonia). A few items identified as A. sylvaticus had already appeared in the early Holocene assemblages, first in the western part of the region, yet the regular appearance of the species is mostly in the post-Neolithic age. A. agrarius appeared sparsely from the Boreal with a maximum frequency during the post-Neolithic period. The results conform well to the picture suggested by molecular phylogeography but demonstrate considerable differences among particular species in dynamic of the range colonization. Further details concerning Holocene paleobiogeography of individual species in the medium latitude Europe are discussed.

Project description:Meadow voles (Microtus pennsylvanicus) are permissive to chronic wasting disease (CWD) infection, but their susceptibility to other transmissible spongiform encephalopathies (TSEs) is poorly characterized. In this initial study, we intracerebrally challenged 6 meadow voles with 2 isolates of sheep scrapie. Three meadow voles acquired a TSE after the scrapie challenge and an extended incubation period. The glycoform profile of proteinase K-resistant prion protein (PrP(res)) in scrapie-sick voles remained similar to the sheep inocula, but differed from that of voles clinically affected by CWD. Vacuolization patterns and disease-associated prion protein (PrP(Sc)) deposition were generally similar in all scrapie-affected voles, except in the hippocampus, where PrP(Sc) staining varied markedly among the animals. Our results demonstrate that meadow voles can acquire a TSE after intracerebral scrapie challenge and that this species could therefore prove useful for characterizing scrapie isolates.

Project description:More than 40 species of mammal have been reported to be infected naturally with Schistosoma japonicum (Chinese mainland strain) in China. The reed vole, Microtus fortis, is the only known mammalian host in which the schistosomes are unable to mature and cause significant pathogenic changes. Gene expression profiling of the 10 day old schistosomula was performed. Microarray analysis was also used to identify differences in gene expression between Schistosoma japonicum schistosomula from BALB/c mice and from Microtus fortis. 10 day old schistosomula were isolated, total RNA obtained and Agilent one colour labeling used. A custom designed Agilent microarray was used to determine what differential gene expression occurs between parasites maintained in either a permissive (mouse) or non-permissive (vole) hosts.

Project description:Throughout the Pleistocene, early humans and carnivores frequented caves and large rock-shelters, usually generating bone accumulations. The well-preserved late Early Pleistocene sedimentary sequence at Cueva Negra del Estrecho del Río Quípar (CNERQ) has provided substantial evidence concerning the behavioural and adaptive skills of early humans in Western Europe, such as butchery practices, lithic technology or tending fire, whilst also bearing witness to the bone-altering activities of carnivores. Recent fieldwork has allowed the re-examination of the spatial and taphonomical nature of the macrofaunal assemblage from the upper layers of Complex 2. These layers are somewhat different from most of the underlying sequence, in showing quite a high representation of cranial and post-cranial bones of large mammals, including several Megaloceros carthaginiensis antlers. The presence of Crocuta sp. at Cueva Negra represents one of the earliest instances of this genus in Western Eurasia. Identification of several juvenile Crocuta sp. remains alongside coprolites and bones with carnivore damage, indicates sporadical hyaenid denning activity. Furthermore, the presence of bones with percussion and cut-marks near to several hammerstones suggests a clear albeit limited anthropogenic input. We interpret the available taphonomical and spatial evidence from these layers as reflecting a multi-patterned palimpsest, likely representing the non-simultaneous and short-lived co-existence of hyaenas, humans, and other small carnivores in the Cueva Negra palaeolandscape during the final phase of sedimentation preserved at the site.

Project description:Grey voles (subgenus Microtus) represent a complex of at least seven closely related and partly cryptic species. The range of these species extends from the Atlantic to the Altai Mountains, but most of them occur east of the Black Sea. Using ancient DNA analyses of the Late Pleistocene specimens, we identified a new mtDNA lineage of grey voles in Europe. Phylogenetic analysis of mitochondrial DNA cytochrome b sequences from 23 voles from three caves, namely, Emine-Bair-Khosar (Crimea, Ukraine), Cave 16 (Bulgaria), and Bacho Kiro (Bulgaria), showed that 14 specimens form a previously unrecognized lineage, sister to the Tien Shan vole. The average sequence divergence of this lineage and the extant Tien Shan vole was 4.8%, which is similar to the divergence of grey vole forms, which are considered distinct species or being on the verge of speciation; M. arvalis and M. obscurus or M. mystacinus and M. rossiaemeridionalis. We estimated the time to the most recent common ancestor of the grey voles to be 0.66 Ma, which is over twice the recent estimates, while the divergence of the extant Tien Shan vole and the new lineage to be 0.29 Ma. Our discovery suggests that grey voles may have been more diversified in the past and that their ranges may have differed substantially from current ones. It also underlines the utility of ancient DNA to decipher the evolutionary history of voles.
