Phosphoproteomic analysis of activated P. falciparum gametocytes, wild type NF54 and PfCDPK4 knockout

Project description:Gametocytes of the malaria parasite Plasmodium are taken up by the mosquito vector with an infectious blood meal, representing a critical stage for parasite transmission. It is well known that calcium dependent protein kinases (CDPKs) play key roles in calcium-mediated signaling across the complex life cycle of the parasite and we sought to understand their role in transmission from the mammalian host to the mosquito vector. Here, we investigated the role of CDPK4 in the life cycle of the human-infective parasite Plasmodium falciparum . We created Plasmodium falciparum cdpk4 knockout parasites by targeted gene deletion, and this deletion had no effect on blood stage development or gametocyte development. However, cdpk4 knockout parasites showed a severe defect in male gametogenesis and the emergence of male flagellated gametes. To understand this defect, we performed mass spectrometry-based phosphoproteomic analysis of wild type and Plasmodium falciparum cdpk4 knockout late gametocyte stages in order to identify key CDPK4-mediated phosphorylation events that may be important for the regulation of male gametogenesis.

Metabolic Biomarkers of Ageing in C57BL/6J Wild-Type and Flavin-Containing Monooxygenase 5 (FMO5)-Knockout Mice

Project description:It was recently demonstrated in mice that knockout of the flavin-containing monooxygenase 5 gene, Fmo5, slows metabolic ageing via pleiotropic effects. We have now used an NMR-based metabonomics approach to study the effects of ageing directly on the metabolic profiles of urine and plasma from male, wild-type C57BL/6J and Fmo5−/− (FMO5 KO) mice back-crossed onto the C57BL/6J background. The aim of this study was to identify metabolic signatures that are associated with ageing in both these mouse lines and to characterize the age-related differences in the metabolite profiles between the FMO5 KO mice and their wild-type counterparts at equivalent time points. We identified a range of age-related biomarkers in both urine and plasma. Some metabolites, including urinary 6-hydroxy-6-methylheptan-3-one (6H6MH3O), a mouse sex pheromone, showed similar patterns of changes with age, regardless of genetic background. Others, however, were altered only in the FMO5 KO, or only in the wild-type mice, indicating the impact of genetic modifications on mouse ageing. Elevated concentrations of urinary taurine represent a distinctive, ageing-related change observed only in wild-type mice.

RNA-sequencing analysis reveals the different gene expression in male and female wild type and TECRL knockout mice.

Project description:Purpose: In previous study, we generated the TECRL knockout mice. The purpose of this study is to indentify the implication function of TECRL on cardiac myocytes in male and female TECRL knockout(TECRL-/-) mice compared with wild type(WT) mice. Methods: Total RNA profiles of 5 males WT mice and 5 males TECRL-/- mice, 3 females WT mice and 7 females TECRL-/- mice were generated by RNA sequencing. The first strand cDNA was synthesized using random hexamer primer and the second strand cDNA synthesis was performed using DNA PolymeraseⅠand RNase H. Results: 1249 genes were identified between TECRL-/- mice and WT mice through RNA-seq analysis. GO analysis and KEGG analysis revealed some pathways, especially lipid metabolism and Ca2+ handling were closedly related to TECRL deficiency.
