Pleiotropic expression of endogenous genes upon fungus infection of wheat plants containing anti-fungal transgenes

Project description:Recent attempts to increase endogenous disease resistance of plants by overexpression of anti-fungal transgenes have shown a potential of this method. However, it has also been shown that such improvements are usually small. One of the obvious reasons for this low anti-fungal effect might be the regulation of endogenous genes in parallel. In this project, we will study the effect of anti-fungal transgenes on the endogenous gene expression. Such effects might relate to substantial equivalence which is a biosafety issue of concern to the public. The GeneChip Wheat Genome Array will be used to detect expression of defence response genes and key genes of metabolic pathways. We will use wheat plants transformed with anti-fungal gene of specific effect against a small group of seed transmitted, pathogenic fungi (KP4 against smuts and bunts). Transformed spring wheat line will be challenged by stinking smut (inhibited by KP4). The effect on the endogenous gene expression will be tested for plants grown in the field in collaboration with the USDA Department. This work will contribute to our understanding of plant defence responses in general and may allow improving strategies to strengthen these responses.

A Fungal Effector Suppresses the Nuclear Export of AGO1-miRNA Complex to Promote Infection in Plants

Project description:Communication between interacting organisms via bioactive molecules is widespread in nature and plays key roles in diverse biological processes. Small RNAs (sRNAs) can travel between host plants and filamentous pathogens to trigger trans-kingdom RNA interference (RNAi) in recipient cells and modulate plant defense and pathogen virulence. However, how trans-kingdom RNAi is regulated has rarely been reported. Here, we show that the secretory protein VdSSR1 (secretory silencing repressor 1) from Verticillium dahliae, a soil-borne phytopathogenic fungus that causes wilt diseases in a wide range of plant hosts, is required for fungal virulence in plants through the suppression of trans-kingdom RNAi.

Cell-type-specific regulation of IL-1-driven inflammation in fungal infection

Project description:Inflammation is a very powerful innate immune response to tissue injury; however, it needs to be tightly regulated to prevent protracted and potentially damaging inflammation. In this project we study the impact of a negative regulator of IL-1 signaling that can be produced by different immune cell subsets. In particular, we intend to identify the contribution of different immune cell subsets, such as neutrophils, macrophages and dendritic cells to the regulation of the anti-fungal immune response. Our prior studies revealed that while macrophage-specific conditional KO mice are strongly protected from the fungal infection, the CD11c-specific conditional KO only shows transient protection. In this RNAseq experiment we aim to identify differentially regulated gene sets and pathways responsible for these effects. In addition, we intend to compare the results of these analyses to a data set generated from a larger cohort of human patiens with disseminated fungal infection.

An ancient symbiotic fungal gene network revealed by comparative transcriptomics [Rirregularis_symbiotic_tissues]

Project description:Purpose: The recent publication of the fungal mutualist R. irregularis genome facilitated transcriptomic studies. We here wanted to understand the large host range of this fungus, throught its gene regulation in divergent plants Methods: mRNA from Medicago truncatula (legume), Brachypodium distachyon (grass) and Lunularia cruciata (liverwort) in association with R. irregularis were sequenced. Reads were mapped on the genome assembly with the software CLC workbench. Fungal gene expression in the different plants was compared to extra radical hyphae as a control. Results: 529, 486 and 523 R. irregularis gene were highly overexpressed (fold change >5 ; FDR <0,05 and experimental value > l10l) in M. truncatula, B. distachyon and L. cruciata, respectively. Among those genes, 262 were induced in all hosts. qPCR validation on 32 genes supported these results in an extended set of hosts (Zea mays spp parviglumis, Pisum sativum, Marchantia paleacea).

Project description:Purpose: We here wanted to describe the gene regulation of Gigaspora rosea in association with phyllogenetically divergent plant hosts, and compare these results with gene regulation occuring in R. irregularis, the model arbuscular mycorrhizal fungus. Methods: mRNA from Medicago truncatula (legume), Brachypodium distachyon (grass) in association with G. rosea, and extra radical mycelium of G.rosea were sequenced by Illumina. Reads were mapped on a in-house de novo transcript assembly with the software CLC workbench. Fungal gene expression in the different host plants was compared to extra radical hyphae as reference. Results: Sets of 1891 and 1566 G. rosea genes were highly overexpressed (fold change >5 ; FDR <0,05 and experimental value difference > 10) , in M. truncatula and B. distachyon respectively compared to ERM, among which 802 of them were up-regulated in both plants. Non common up-regulated genes are mainly found non statistically robust in one condition. Conclusions: Our study represents the first transcriptomic analysis on several hosts for this fungal species. These results showed that the interaction between plants and AMF is highly conserved.

A 5’ tRNA-Ala derived small RNA regulates anti-fungal defense in plants

Project description:Apart from their primordial role in protein synthesis, tRNAs can be cleaved to produce tRNA-derived small RNAs (tsRNAs). The biological functions of tsRNAs in plants remain largely unknown. In this study, we developed RtcB-based RNA sequencing, a method that captures and distinguishes between 2’,3’-cyclic-phosphate (cP)-terminated RNAs and 3’-OH-terminated RNAs, and profiled 5’ tsRNAs and 5’ tRNA halves in Arabidopsis thaliana. We found that Arabidopsis 5’ tsRNAs and 5’ tRNA halves predominantly contain a cP at the 3’ end and require S-like RNase 1 (RNS1) and RNS3 for their production. One of the most abundant 5’ tsRNA, 5’ tsR-Ala, likely by associating with AGO1, negatively regulates Cytochrome P450 71A13 (CYP71A13) expression and camalexin biosynthesis to repress anti-fungal defense. 5’ tsR-Ala is downregulated upon fungal infection. Our study provides a global view of 5’ tsRNAs and 5’ tRNA halves in Arabidopsis and unravels an important role of a 5’ tsRNA in regulating anti-fungal defense.
