This paper describes the construction of a food safety-oriented potato cDNA microarray (FSPM). Microarray analysis was performed on a well-defined set of tuber samples of two different potato varieties, grown under different, well-recorded environmental conditions. Data were analyzed to assess the potential of transcriptomics to detect differences in gene expression due to genetic differences or environmental conditions. The most pronounced differences were found between the varieties Sante and Lady Balfour, whereas differences due to growth conditions were less significant. Transcriptomics results were confirmed by quantitative PCR. Furthermore, the bandwidth of natural variation of gene expression was explored to facilitate biological and/or toxicological evaluation in future assessments. Project description:The use of biofertilizers is becoming an economical and environmentally friendly alternative to promote sustainable agriculture. Biochar from microalgae can be applied to enhance the productivity of food crops through soil improvement, slow nutrient absorption and release, increased water uptake, and long-term mitigation of greenhouse gas sequestration. Therefore, the aim of this study was to evaluate the stimulatory effects of biochar produced from Spirulina platensis biomass on the development and seed production of rice plants. Biochar was produced by slow pyrolysis at 300°C, and characterization was performed through microscopy, chemical, and structural composition analyses. Molecular and physiological analyses were performed in rice plants submitted to different biochar concentrations (0.02, 0.1, and 0.5 mg mL-1) to assess growth and productivity parameters. Morphological and physicochemical characterization revealed a heterogeneous morphology and the presence of K and Mg minerals in the biochar composition. Chemical modification of compounds post-pyrolysis and a highly porous structure with micropores were observed. Rice plants submitted to 0.5 mg mL-1 of biochar presented a decrease in root length, followed by an increase in root dry weight. The same concentration influenced seed production, with an increase of 44% in the number of seeds per plant, 17% in the percentage of full seeds per plant, 12% in the weight of 1,000 full seeds, 53% in the seed weight per plant, and 12% in grain area. Differential proteomic analyses in shoots and roots of rice plants submitted to 0.5 mg mL-1 of biochar for 20 days revealed a fine-tuning of resource allocation towards seed production. These results suggest that biochar derived from Spirulina platensis biomass can stimulate rice seed production.

Project description:Phosphorus (P) is a limiting nutrient for plants and an essential element for all life on Earth. As the resources of phosphate rock are depleting, new management tools for environmentally friendly P fertilizers are needed. In order to achieve this, recent studies have proposed to use biochar, a carbon-rich solid product of thermochemical conversion of biomass with minimal or zero oxygen supply, as slow-release P fertilizer. However, the effects of biochar on plant-available P in soils have been reported to be variable. Therefore, we quantitatively evaluated existing peer-reviewed data using meta-analysis to draw general conclusions. In the present study, we evaluated 108 pairwise comparisons to their response of biochar application on P availability in soils. Our results indicate that biochar can act as a short-, mid-, and long-term P fertilizer with its effect depending on feedstock, pyrolysis temperature and application amount. Overall, the addition of biochar significantly increased the P availability in agricultural soil by a factor of 4.6 (95% confidence interval 3.4-5.9), independent of the used feedstock for biochar production. Only biochar application amounts above 10 Mg ha-1 and biochar produced at temperatures lower than 600 °C significantly increased the P availability of agricultural soils. The application of biochar to acid (pH < 6.5) and neutral soils (pH 6.5-7.5) significantly increased plant-P availability by a factor of 5.1 and 2.4, respectively (95% confidence interval 3.5-6.7 and 1.4-3.4, respectively), while there was no significant effect in alkaline soils (pH > 7.5). Taken together, this meta-analysis shows that biochar significantly enhances plant-available P in biochar-amended soils at least for five years.
