Project description:The native microflora associated with mosquitoes have important roles in mosquito development and vector competence. Sequencing of bacterial V3 region from 16S rRNA genes across the developmental stages of Culex mosquitoes (early and late larval instars, pupae and adults) was used to test the hypothesis that bacteria found in the larval stage of Culex are transstadially transmitted to the adult stage, and to compare the microbiomes of field-collected versus laboratory-reared mosquitoes.Beta diversity analysis revealed that bacterial community structure differed among three life stages (larvae, pupae and adults) of Culex tarsalis. Although only ~2% of the total number of bacterial OTUs were found in all stages, sequences from these OTUs accounted for nearly 82% of the total bacterial sequences recovered from all stages. Thorsellia (Gammaproteobacteria) was the most abundant bacterial taxon found across all developmental stages of field-collected Culex mosquitoes, but was rare in mosquitoes from laboratory-reared colonies. The proportion of Thorsellia sequences in the microbiomes of mosquito life stages varied ontogenetically with the greatest proportions recovered from the pupae of C. tarsalis and the lowest from newly emerged adults. The microbiome of field-collected late instar larvae was not influenced significantly by differences in the microbiota of the habitat due to habitat age or biopesticide treatments. The microbiome diversity was the greatest in the early instar larvae and the lowest in laboratory-reared mosquitoes.Bacterial communities in early instar C. tarsalis larvae were significantly more diverse when compared to late instar larvae, pupae and newly emerged adults. Some of the bacterial OTUs found in the early instar larvae were also found across developmental stages. Thorsellia dominated the bacterial communities in field-collected immature stages but occurred at much lower relative abundance in adults. Differences in microbiota observed in larval habitats did not influence bacterial community profiles of late instar larvae or adults. However, bacterial communities in laboratory-reared C. tarsalis larvae differed significantly from the field. Determining the role of Thorsellia in mosquitoes and its distribution across different species of mosquitoes warrants further investigation.

Project description:Investigation of gene expression level differences in Culex pipiens (SB) and C. quinquefasciatus (JHB) at three time points (8, 16, and 24 hours post-exposure) during the early pupal state in standard (25°C; 16 h light/8 h dark) and diapause-inducing (18°C; 8 h light/16 h dark) conditions. A forty-eight chip study (4 Expr12x135K slides) using cDNA from total RNA collected from two species in the Culex pipiens complex at 3 time-points during the early pupal stage to study gene expression differences between standard (25°C; 16 h light/8 h dark) and diapause-inducing (18°C; 8 h light/16 h dark) conditions, and between Culex pipiens (diapause) and C. quinquefasciatus (no diapause). Each chip measures the expression level of 18,692 protein coding genes, with 3 probes per gene and two-fold technical redundancy. Each of the 12-plex slides was used for one of four biological replicates. Probes were designed using the C.quinquefasciatus CpipJ1.2 geneset in VectorBase.

Project description:Determination of miRNA profiles in most prominent mosquitoes will determine the potential targets for mosquito control Some of the most medically important viruses, such as dengue virus, West Nile virus, Zika virus, and yellow fever virus, are transmitted by mosquitoes. These aptly named arboviruses impose a tremendous cost to the health of populations around the world. As a result, much effort has gone into the study of the impact of these viruses in human infections. Comparatively less efforts, however, have been made to study the way these viruses interact with mosquitos themselves. It has long been held that these viruses are introduced into the midgut of mosquitoes upon ingestion of a blood meal before being transmitted within the saliva upon subsequent feeding. This sequence requires that the mosquito be able to defend itself from infection every step along the way-from ingesting bloodmeal to subsequent feeding. The main defense mechanisms employed by the mosquitoes to control viruses is RNA interference (RNAi). Modulation of this facet of the mosquito’s immune system would thereby suggest a practical strategy for vector control. This paper will provide an up to date overview of the mosquito’s immune system along with novel data describing miRNA profiles for Aedes aegypti and Culex quinquefasiatus in Grenada, West Indies.

Project description:Rift valley fever (RVF) is an emerging zoonotic disease and it is caused by Rift valley fever phlebovirus (RVFV). This virus is commonly transmitted in endemic areas between wild ruminants and mosquitoes, mainly by mosquitoes of Culex and Aedes genus. Starting from 2000, several outbreaks have been reported outside the African continent, in countries facing the Mediterranean Sea, such as Saudi Arabia. The available vaccines for ruminants present limited efficacy or residual pathogenic effects. Consequently, new strategies are urgently required to limit the expansion of this zoonotic virus. The main objective of this work is to investigate the molecular responses of Culex pipiens to RVFV focusing mainly on genes implicated in the classical innate immunity pathways, RNAi mechanism and apoptosis process in order to elucidate the implicated genes in viral infection. The immune altered genes here described could be potential targets to control RVFV infection in mosquitoes. Some of the genes related to the immune defense response were previously described in others mosquito-arbovirus models, as also in Drosophila and human. To our knowledge, this study elucidates for the first time the Cx. pipiens-RVFV interaction in terms of defense infection-response, which was largely under studied and provides information to develop new approaches to prevent and control the expansion of the virus in the future.
