Project description:This study aimed to determine sediment contamination in the Beidagang Wetland Nature Reserve to describe atmospheric deposition of trace metals. We analyzed Hg, Cd, Pb, TOC, TN, TP, ?13C, and ?15N, and studied their variations in surface sediments and in the vertical profiles of sediment cores collected from the reserve. Evaluation of environmental trace metal contamination using sediment quality guidelines and geochemical background values indicated that the risk of metal pollution in the reserve sediments was relatively low. Concentrations of Hg, Cd, and Pb in the sediments were much lower than concentrations in sediment samples from Bohai Bay and polluted rivers in Tianjin. Enrichment factors indicate that samples are moderately contaminated with Hg, Cd, and Pb; whereas the geo-accumulation index results classify the sediments as uncontaminated to moderately contaminated with Hg, Cd, and Pb. The distribution patterns of trace metal concentrations in the three core samples were uniform. ?13C and ?15N were used to track the sources of TOC and TN in sediments. Results show that TOC mainly originated from the residue and decaying matter of aquatic plants (e.g., algae, reeds, and Typha), while TN was derived from soil N and elevated atmospheric N deposition. Because domestic and industrial waste is not discharged into the Beidagang Wetland Nature Reserve, trace metals found in sediments mainly originate from atmospheric deposition. The results provide baseline data for analysis of trace metal accumulation in Beijing-Tianjin-Hebei, a region subject to atmospheric deposition in northern China.

Project description:Purpose: The goal of this study was to characterized the transcriptome profiles of whole-blood cells from 388 specimens obtained from 106 individuals before and after the meditation retreat at four-time points (T1–T4) by RNA sequencing (RNA-Seq). T1 samples were collected 5-8 weeks before the retreat, T2 samples were collected on the day of retreat before starting the meditation method, T3 samples were collected immediately after the retreat, and T4 samples were collected three months after the retreat. Methods: The human blood samples were collected into PAXgene Blood RNA Tubes and stored at -80°C freezer. Before RNA extraction the samples were removed from -80°C and incubated overnight at room temperature. The samples were randomized before RNA extraction to eliminate any time point or age or sex or batch effect. Manufacturer’s protocol of manual purification of total RNA from human whole blood was followed (Qiagen, cat #762164). RNA was reverse transcribed to complementary DNA (Lexogen QuantSeq 3′ FWD), and sequenced on a HiSeq 4000 instrument (Illumina). Results: We applied a comprehensive systems biology approach starting with whole blood gene expression profiling combined with multi-level bioinformatic analyses to characterize the co-expression, transcriptional, and protein-protein interaction networks to identify meditation-specific core network after an advanced eight-day Inner Engineering retreat program. We found the response to oxidative stress, detoxification, and cell cycle regulation pathways were downregulated after meditation. Strikingly, 220 genes directly associated with immune response, including 68 genes related to interferon (IFN) signaling were upregulated, with no significant expression changes in the inflammatory genes. This robust meditation-specific immune response network is significantly dysregulated in multiple sclerosis and severe COVID-19 patients. Conclusions: The present proof-of-principle study demonstrates that the immune system can be voluntarily influenced by non-pharmaceutical interventions like yoga and meditation. This suggests that meditation as a behavioral intervention could have important implications for treating various conditions associated with excessive or persistent inflammation with a dampened immune system profile.

Project description:Hydrocarbon vents have recently been reported to contribute considerable amounts of dissolved organic carbon (DOC) to the oceans. Many such hydrocarbon vents widely exist in the northern South China Sea (NSCS). To investigate if these hydrocarbon vent sites release DOC, we used a real-time video multiple-corer to collect bottom seawater and surface sediments at vent sites. We analyzed concentrations of DOC in these samples and estimated DOC fluxes. Elevated DOC concentrations in the porewaters were found at some sites suggesting that DOC may come from these hydrocarbon vents. Benthic fluxes of DOC from these sediments were 28 to 1264 μmol m(-2 )d(-1) (on average ~321 μmol m(-2 )d(-1)) which are several times higher than most DOC fluxes in coastal and continental margin sediments. The results demonstrate that the real-time video multiple-corer can precisely collect samples at vent sites. The estimated benthic DOC flux from the methane venting sites (8.6 × 10(6 )mol y(-1)), is 24% of the DOC discharge from the Pearl River to the South China Sea, indicating that these sediments make an important contribution to the DOC in deep waters.

Project description:Perennial cropping has been an alternative land use type due to its widely accepted role in increasing soil carbon sequestration. However, how soil organic carbon (SOC) changes and its underlying mechanisms under different cropping years are still elusive. A chronosequence (0-, 3-, 6-, 20-year) of perennial mugwort cropping was chosen to explore the SOC dynamics and the underlying mechanisms in agricultural soils of Northern China Plain. The results revealed that SOC first increased and then decreased along the 20-year chronosequence. The similar patterns were also found in soil properties (including soil ammonium nitrogen, total nitrogen and phosphorus) and two C-degrading hydrolytic enzyme activities (i.e., α-glucosidase and β-glucosidase). The path analysis demonstrated that soil ammonium nitrogen, total nitrogen, and plant biomass affected SOC primarily through the indirect impacts on soil pH, total phosphorus availability, and C-degrading hydrolytic enzyme activities. In addition, the contributions of soil properties are greater than those of biotic factors (plant biomass) to changes in SOC across the four mugwort cropping years. Nevertheless, the biotic factors may play more important roles in regulating SOC than abiotic factors in the long run. Moreover, SOC reached its maximum and was equaled to that under the conventional rotation when cropping mugwort for 7.44 and 14.88 years, respectively, which has critical implications for sustainable C sequestration of agricultural soils in Northern China Plain. Our observations suggest that short-term but not long-term perennial mugwort cropping is an alternative practice benefiting soil C sequestration and achieving the Carbon Neutrality goal in China.

Project description:Atmospheric brown carbon (BrC) can impact the radiative balance of the earth and form photooxidants. However, the light absorption and photochemical properties of BrC from different sources remain poorly understood. To address this gap, dilute water extracts of particulate matter (PM) samples collected at Davis, CA over one year were analyzed using high resolution aerosol mass spectrometry (HR-AMS) and UV-vis spectroscopy. Positive matrix factorization (PMF) on combined AMS and UV-vis data resolved five water-soluble organic aerosol (WSOA) factors with distinct mass spectra and UV-vis spectra: a fresh and an aged water-soluble biomass burning OA (WSBBOAfresh and WSBBOAaged) and three oxygenated OA (WSOOAs). WSBBOAfresh is the most light-absorbing, with a mass absorption coefficient (MAC365 nm) of 1.1 m2 g-1, while the WSOOAs are the least (MAC365 nm = 0.01-0.1 m2 g-1). These results, together with the high abundance of WSBBOAs (∼52% of the WSOA mass), indicate that biomass burning activities such as residential wood burning and wildfires are an important source of BrC in northern California. The concentrations of aqueous-phase photooxidants, i.e., hydroxyl radical (·OH), singlet molecular oxygen (1O2*), and oxidizing triplet excited states of organic carbon (3C*), were also measured in the PM extracts during illumination. Oxidant production potentials (PPOX) of the five WSOA factors were explored. The photoexcitation of BrC chromophores from BB emissions and in OOAs is a significant source of 1O2* and 3C*. By applying our PPOX values to archived AMS data at dozens of sites, we found that oxygenated organic species play an important role in photooxidant formation in atmospheric waters.

Project description:BackgroundExtensive studies have been conducted to evaluate the effect of external organic Carbon on native soil organic carbon (SOC) decomposition. However, the direction and extent of this effect reported by different authors is inconsistent.ObjectiveThe objective was to provide a synthesis of existing data that comprehensively and quantitatively evaluates how the soil chemical properties and incubation conditions interact with additional external organic C to affect the native SOC decomposition.Data sourceA meta-analysis was conducted on previously published empirical studies that examined the effect of the addition of external organic carbon on the native SOC decomposition through isotopic techniques.Results and conclusionsThe addition of external organic C, when averaged across all studies, enhanced the native SOC decomposition by 26.5%. The soil with higher SOC content and fine texture showed significantly higher priming effects, whereas the soil with higher total nitrogen content showed an opposite trend. The soils with higher C:N ratios had significantly stronger priming effects than those with low C:N ratios. The decomposition of native SOC was significantly enhanced more at early stage of incubation (<15d) than at the later stages (>15d). In addition, the incubation temperature and the addition rate of organic matter significantly influenced the native SOC decomposition in response to the addition of external organic C.

Project description:Metagenome-assembled genomes (MAGs) have revealed the existence of novel bacterial and archaeal groups and provided insight into their genetic potential. However, metagenomics and even metatranscriptomics cannot resolve how the genetic potential translates into metabolic functions and physiological activity. Here, we present a novel approach for the quantitative and organism-specific assessment of the carbon flux through microbial communities with stable isotope probing-metaproteomics and integration of temporal dynamics in 13C incorporation by Stable Isotope Cluster Analysis (SIsCA). We used groundwater microcosms labeled with 13CO2 and D2O as model systems and stimulated them with reduced sulfur compounds to determine the ecosystem role of chemolithoautotrophic primary production. Raman microspectroscopy detected rapid deuterium incorporation in microbial cells from 12 days onwards, indicating activity of the groundwater organisms. SIsCA revealed that groundwater microorganisms fell into five distinct carbon assimilation strategies. Only one of these strategies, comprising less than 3.5% of the community, consisted of obligate autotrophs (Thiobacillus), with a 13C incorporation of approximately 95%. Instead, mixotrophic growth was the most successful strategy, and was represented by 12 of the 15 MAGs expressing pathways for autotrophic CO2 fixation, including Hydrogenophaga, Polaromonas and Dechloromonas, with varying 13C incorporation between 5% and 90%. Within 21 days, 43% of carbon in the community was replaced by 13C, increasing to 80% after 70 days. Of the 31 most abundant MAGs, 16 expressed pathways for sulfur oxidation, including strict heterotrophs. We concluded that chemolithoautotrophy drives the recycling of organic carbon and serves as a fill-up function in the groundwater. Mixotrophs preferred the uptake of organic carbon over the fixation of CO2, and heterotrophs oxidize inorganic compounds to preserve organic carbon. Our study showcases how next-generation physiology approach like SIsCA can move beyond metagenomics studies by providing information about expression of metabolic pathways and elucidating the role of MAGs in ecosystem functioning.
