Chemopreventive effect of flaxseed-enriched diet on ovarian cancer in laying hen.

Project description:The objective of this study was to identify the molecular mechanisms and biological pathways associated with the anticancer effects of flaxseed (richest plant source of Omega-3 fatty acid) in laying hen model of ovarian cancer. miRNA expression profiling in liver of juvenile and egg-laying hens

Project description:A total of 565 miRNAs annotated in miRBase 20.0 were identified to be expressed in the liver of hen by high-throughput sequencing three biological reduplication libraries in juvenile and egg-laying hens, respectively. Compared with juvenile hen, 80 miRNAs (67 down-regulated and 13 up-regulated) were verified to be significant differential expression (SDE) in egg-laying physiological stage. Among these, miR-22-3p has the highest abundant expression, and miR-146b-5p has the highest fold-change. Additionally, 19 of the 71 novel miRNAs was significantly expressed. Furthermore, 648 putative target genes of the SDE miRNAs were obtained, and among these, FADS1, FADS2, ELOVL6, ACSL5, etc which are lipid metabolism related critical regulators are targeted by some SDE miRNAs. Gene Ontology (GO) analyses to the putative target genes of all the SDE miRNAs showed significantly enriched in Steroid biosynthesis, Glycerophospholipid metabolism, Biosynthesis of unsaturated fatty acids, and PPAR signaling pathway (P ≤ 0.05). Meanwhile, GO terms are also significantly enriched in lipid related biological processes.

Associations between antimicrobial use and the faecal resistome on broiler farms from nine European countries.

Project description:ObjectivesTo determine associations between farm- and flock-level antimicrobial usage (AMU), farm biosecurity status and the abundance of faecal antimicrobial resistance genes (ARGs) on broiler farms.MethodsIn the cross-sectional pan-European EFFORT study, conventional broiler farms were visited and faeces, AMU information and biosecurity records were collected. The resistomes of pooled faecal samples were determined by metagenomic analysis for 176 farms. A meta-analysis approach was used to relate total and class-specific ARGs (expressed as fragments per kb reference per million bacterial fragments, FPKM) to AMU (treatment incidence per DDD, TIDDDvet) per country and subsequently across all countries. In a similar way, the association between biosecurity status (Biocheck.UGent) and the resistome was explored.ResultsSixty-six (38%) flocks did not report group treatments but showed a similar resistome composition and roughly similar ARG levels to antimicrobial-treated flocks. Nevertheless, we found significant positive associations between β-lactam, tetracycline, macrolide and lincosamide, trimethoprim and aminoglycoside antimicrobial flock treatments and ARG clusters conferring resistance to the same class. Similar associations were found with purchased products. In gene-level analysis for β-lactams and macrolides, lincosamides and streptogramins, a significant positive association was found with the most abundant gene clusters blaTEM and erm(B). Little evidence was found for associations with biosecurity.ConclusionsThe faecal microbiome in European broilers contains a high diversity of ARGs, even in the absence of current antimicrobial selection pressure. Despite this, the relative abundance of genes and the composition of the resistome is positively related to AMU in European broiler farms for several antimicrobial classes.
