Project description:Obesity is a chronic inflammatory condition which leads to a change in the production of adipokines and cytokines that can be found in blood plasma and saliva. It increases the risk for periodontal and cardiovascular diseases, hypertension, dyslipidemia, type 2 Diabetes Mellitus, cancer and others. This study aims to analyze the plasma and saliva proteomes of young adults with obesity and overweight comparing to normal weight individuals, to reveal the influence of body index in the proteome profiles and to identify early biomarkers of obesity-associated diseases.

CDKN2A-CDK4-E2F3 dependent oxidative skeletal muscle fiber transition in myogenesis, movement, and metabolism

Project description:Skeletal muscle function is vital to movement, thermogenesis and metabolism. Muscle fibers differ in contractile ability, mitochondrial content and metabolic properties and muscle fiber transition influences muscle function. However, the molecular mechanisms regulating muscle fiber transition in muscle function are unclear. Here, in over 150 human muscle samples, we observed that markers of oxidative muscle fiber and mitochondria correlate positively with PPARGC1 and CDK4, and, negatively with CDKN2A, a locus significantly associated with type 2 diabetes. Mice expressing an overactive Cdk4 that cannot bind its inhibitor p16INK4a, a product of the CDKN2A locus, are longer, leaner, exhibit increased oxidative myofibers with superior mitochondrial energetics, display enhanced muscle glucose uptake, and are protected from obesity and diabetes. In contrast, Cdk4-deficiency, or skeletal muscle-specific deletion of Cdk4’s transcriptional target, E2F3, reduces oxidative myofiber numbers, deteriorates mitochondrial function and exercise capacity, while increasing diabetes susceptibility. E2F3 activates the PPARGC1 promoter and CDK4/E2F3/PPARGC1 levels correlate positively with exercise and fitness, and negatively with adiposity, insulin resistance and lipid accumulation in muscle. These findings provide insight into oxidative muscle fiber transition and function that is of relevance to metabolic and muscular diseases.

Linking gut microbiome metabolism of fiber-snack glycans to multifunctional changes in human plasma proteomes

Project description:The worldwide increase in snack-food consumption associated with adoption of Westernized lifestyles provides an opportunity to introduce new nutritious foods into non-nutritious diets. We describe two 10-week-long pilot studies of obese or overweight individuals measured the effects of pea fiber- or orange fiber-containing snack food prototypes on the abundances of fecal microbiome glycoside hydrolases (GH) and polysaccharide lyases (PL), fecal glycan metabolism, and >1300 plasma proteins. Computational methods for feature selection plus contextual language processing software that extracts information from all PubMed abstracts referencing features of interest, identified treatment-discriminatory changes in GH and PL gene abundances that correlated with levels of fecal glycosidic linkages related to fiber metabolism, and plasma proteins representing diverse biological themes, including TGFÃŽÂ²/BMP-mediated fibrotic responses, VEGF-related angiogenic responses, P38/MAPK-associated immune cell biomarkers, and obesity-associated hormonal regulators. These results disclose fiber-snack prototype effects on microbiome function and host physiology in the context of varying background dietary practices.

Proteomics analysis of skeletal muscle from leptin-deficient ob/ob mice reveals adaptive remodeling of metabolic characteristics and fiber type composition

Project description:Skeletal muscle insulin resistance, an early metabolic defect in the pathogenesis of type 2 diabetes, may be a cause or consequence of altered protein expressions profiles. Proteomics technology offers enormous promise to investigate molecular mechanisms underlying pathologies, however, the analysis of skeletal muscle is challenging. Using a state-of-the-art mass spectrometry (MS) based workflow, we performed a global proteomics analysis of skeletal muscle from leptin-deficient, obese, type 2 diabetic (ob/ob) and lean mice, identifying more than 6,000 proteins with 118 proteins differentially regulated in obesity. This included protein kinases, phosphatases, and secreted and fiber type associated proteins. Enzymes involved in lipid metabolism in skeletal muscle from ob/ob mice were increased, providing evidence against reduced fatty acid oxidation in lipid-induced insulin resistance. Mitochondrial and peroxisomal proteins, as well as components of pyruvate and lactate metabolism were likewise increased. Finally, the skeletal muscle proteome from ob/ob mice displayed a shift towards the ‘slow fiber type’. This detailed characterization of obese rodent models of type 2 diabetes demonstrates an efficient workflow for skeletal muscle proteomics, which may easily be adapted to other complex tissues.

Insulin resistance rewires the metabolic gene program and glucose utilization in human white adipocytes

Project description:The aim of this study is to define the molecular alterations occurring in human adipose tissue leading to its dysfunction. While obesity (defined by BMI>30) is strongly associated with insulin resistance and metabolic disorders, some individuals with obesity remain insulin sensitive. Therefore, in this study, we have considered a control group (lean, insuln sensitive: BMI<25, HOMA-IR<2), a group with overweight/obesity but without insulin resistance (BMI>25, HOMA-IR<2) and a group with overweight and insulin resistance (BMI>25, HOMA-IR>3) to dissect the transcriptional changes in viscreal adipose tissue due to increased fat mass, and those associated with metabolic disorder.

Fiber-rich Foods to Treat Obesity and Prevent Colon Cancer

Project description:The study will target a population at high risk for colorectal cancer, overweight and obese participants with a history of a colon polyp in the past 3 years to test whether a high-legume, high-fiber diet will simultaneously increase weight loss and suppress intestinal biomarkers of cancer risk compared to a control diet (healthy American). In addition, it will explore potential mechanisms through which the high-legume intervention diet facilitates weight loss and intestinal health.
