Project description:Human-induced pluripotent stem cells (iPSCs) are derived from differentiated somatic cells using defined factors and provide a renewable source of autologous cells for cell therapy. Many reprogramming methods have been employed to generate human iPSCs, including the use of integrating vectors and non-integrating vectors. Maintenance of the genomic integrity of iPSCs is highly desirable if the cells are to be used in clinical applications. Here, using the Affymetrix Cytoscan HD array, we investigated the genomic aberration profiles of 19 human cell lines: 5 embryonic stem cell (ESC) lines, 6 iPSC lines derived using integrating vectors (“integrating iPSC lines”), 6 iPSC lines derived using non-integrating vectors (“non-integrating iPSC lines”), and the 2 parental cell lines from which the iPSCs were derived. The genome-wide copy number variation (CNV), loss of heterozygosity (LOH) and mosaicism patterns of integrating and non-integrating iPSC lines were investigated. The maximum sizes of CNVs in the genomes of the integrating iPSC lines were 20 times higher than those of the non-integrating iPSC lines. Moreover, the total number of CNVs was much higher in integrating iPSC lines than in other cell lines. The average numbers of novel CNVs with a low degree of overlap with the DGV and of likely pathogenic CNVs with a high degree of overlap with the ISCA database were highest in integrating iPSC lines. Different SNP calls revealed that, using the parental cell genotype as a reference, integrating iPSC lines displayed more single nucleotide variations and mosaicism than did non-integrating iPSC lines. This study describes the genome stability of human iPSCs generated using either a DNA-integrating or non-integrating reprogramming method, of the corresponding somatic cells, and of hESCs. Project description:BackgroundThe combined microbial fuel cell-microbial nutrient recovery system has lately been thoroughly explored from an engineering standpoint. The relevance of microbial communities in this process, on the other hand, has been widely underestimated.ResultsA lab-scale microbial nutrients recovery system was created in this work, and the microbial community structure was further defined, to give a thorough insight into the important microbial groups in the present system. We reported for the first-time different hybrid anodes of activated carbon and chitosan that were used in the microbial nutrient recovery system for bioenergy production, and, for the removal of COD and recovery of nutrients present in the wastewater. The hybrid anodic materials were studied to adapt electrochemically active bacteria for the recovery of nutrients and energy generation from wastewater without the need for an external source of electricity. The potential of the created hybrid anodes in terms of nutrients recovery, chemical oxygen demand elimination, and energy generation from municipal wastewater was thoroughly examined and compared with each other under similar operating conditions. When the COD loading was 718 mg/L, a total COD removal of ~ 79.2% was achieved with a hybrid activated carbon and chitosan anode having an equal ratio after 10 days of the operation cycle. The maximum power density estimated for hybrid anode (~ 870 mWm-2) was found.ConclusionOverall, this work reveals a schematic self-driven way for the collection and enrichment of nutrients (~ 72.9% phosphorus recovery and ~ 73% ammonium recovery) from municipal wastewater, as well as consistent voltage production throughout the operation.

Project description:Novel heterotrophic bacterial strains-Bzr02 and Str21, effective in nitrogen transformation, were isolated from sequential sedimentation-biofiltration systems (SSBSs). Bzr02, identified as Citrobacter freundii, removed up to 99.0% of N-NH4 and 70.2% of N-NO3, while Str21, identified as Pseudomonas mandelii, removed up to 98.9% of N-NH4 and 87.7% of N-NO3. The key functional genes napA/narG and hao were detected for Bzr02, confirming its ability to reduce nitrate to nitrite and remove hydroxylamine. Str21 was detected with the genes narG, nirS, norB and nosZ, confirming its potential for complete denitrification process. Nitrogen total balance experiments determined that Bzr02 and Str21 incorporated nitrogen into cell biomass (up to 94.7% and 74.7%, respectively), suggesting that nitrogen assimilation was also an important process occurring simultaneously with denitrification. Based on these results, both strains are suitable candidates for improving nutrient removal efficiencies in nature-based solutions such as SSBSs.
