Desiccation Tolerance of Arabidopsis thaliana seeds

Project description:Desiccation tolerance (DT) allowed seed plants to conquer ecosystems with long periods of limited water availability. This adaptive features allows seeds to remain dried for very long times without losing their ability to germinate. There is little information about all the signaling components required to achieve DT and on how transcription factors (TFs) modulate global DT processes. We performed RNA-seq experiment and carbohydrates profiles of lec1, lec2, fus3 and abi3, as well as their corresponding wild types, at three stages of seed development 15, 17 and 21 DAF (day after open flower) belonging to the seed desiccation period. A complex experimental design approach and regulatory networks prediction were used to identify differentially expressed genes specifically involved in DT process. In order to identify mechanisms involved in the acquisition of DT during seed development, we designed a comparative transcriptomic analysis between the seed desiccation intolerant (DI) mutants lec1-1, abi3-5 and fus3-3, the desiccation tolerant mutant lec2-1 and the desiccation tolerant weak allele of abi3 (abi3-1) with their respective wild type controls. This analysis should allow to identify genes that are differentially expressed in the desiccation intolerant mutants respect to tolerant mutants and WT controls.

Project description:This study was developed under the framework of the Human Biomonitoring for Europe Initiative (HBM4EU). We evaluated the possible impacts of BPA exposure in adolescent males from the INMA-Granada cohort (Spain) with the goal of identifying new biomarkers that could be useful in biomonitoring and epidemiological studies. Our hypothesis is that alterations in epigenetic regulation could lead to genetic instability in human subjects exposed to environmentally relevant doses of BPA.

Proteomics data analysis for the identification of proteins and derived proteotypic peptides to be used as drought tolerance markers in Quercus ilex

Project description:Drought is considered one of the main causes of mortality of holm oak (Quercus ilex) upon seedling plantations in reforestation programs. Although Q. ilex shows high adaptability to extreme climate conditions prevailing in Southern Spain, the genetic variability inherent to this species could be associated with a differential response depending on populations or individuals. The objective of this work was to identify proteins and derived proteotypic peptides to be used as markers of tolerance to drought in holm oak using a targeted post-acquisition proteomics approach. The study was based on a set of proteins identified by shotgun (LC-MSMS) from a drought experiment carried out in Q. ilex seedlings from four Andalusian provenances (Granada, Huelva, Cadiz and Seville). A double strategy, shotgun protein quantification and target peptides quantification post-acquisition data analysis based on proteotypic peptides, was used. For this purpose, an initial list of target peptides consisting in proteotypic peptides derived of proteins highly represented in drought conditions was compiled. Then, the proteotypic peptides list and the raw files from the shotgun experiment were imported into the Skyline software to quantify the relative abundance of the fragments ion peaks. Those peptides significantly more abundant in drought conditions at least in two populations were selected as putative markers of tolerance to drought. These corresponded to proteins belonged to redox, stress related proteins, synthesis, folding and degradation, and primary and secondary metabolism functional groups. Two proteins were found to be increased in three populations, making them especially interesting for validation in subsequent experiments as markers of drought tolerance.
