Project description:Adolescence is a critical period in cognitive and emotional development, characterized by high levels of social interaction and increases in risk-taking behavior including binge drinking. Adolescent exposure to social stress and binge ethanol have individually been associated with the development of social, emotional, and cognitive deficits, as well as increased risk for alcohol use disorder. Disruption of cortical development by early life social stress and/or binge drinking may partly underlie these enduring emotional, cognitive, and behavioral effects. The study goal is to implement a novel neighbor housing environment to identify the effects of adolescent neighbor housing and/or binge ethanol drinking on (1) a battery of emotional and cognitive tasks (2) adult ethanol drinking behavior, and (3) the nucleus accumbens and prefrontal cortex transcriptome. Adolescent male and female C57BL/6J mice were single or neighbor housed with or without access to intermittent ethanol. One cohort underwent behavioral testing during adulthood to determine social preference, expression of anxiety-like behavior, cognitive performance, and patterns of ethanol intake. The second cohort was sacrificed in late adolescence and brain tissue was used for transcriptomics analysis. As adults, single housed mice displayed decreased social interaction, deficits in the novel object recognition task, and increased anxiety-like behavior, relative to neighbor-housed mice. There was no effect of housing condition on adolescent or adult ethanol consumption. Adolescent ethanol exposure did not alter adult ethanol intake. Transcriptomics analysis revealed that adolescent housing condition and ethanol exposure resulted in differential expression of genes related to synaptic plasticity in the nucleus accumbens and genes related to methylation, the extracellular matrix and inflammation in the prefrontal cortex. The behavioral results indicate that social interaction during adolescence via the neighbor housing model may protect against emotional, social, and cognitive deficits. In addition, the transcriptomics results suggest that these behavioral alterations may be mediated in part by dysregulation of transcription in the frontal cortex or the nucleus accumbens

Project description:Orofacial inflammation could lead to transcriptional alterations in the trigeminal ganglion (TG). In this study, we performed single-cell RNA-sequencing (scRNA-seq) analysis of mouse TG to identify all cell types and profile transcriptomic alterations of TG cells under inflammatory conditions. A total of 7 types of cells including endothelial cells, fibroblasts, glial cells, granulocytes, lymphocytes, monocyte-macrophages and several subtypes of neurons were identified. In addition, we performed annotation of neuronal subtypes and differential gene expression analysis among TG neurons, identifying several differential genes involved in pain modulation such as Scn10a, Zbtb20 and Runx1. Collectively, our study revealed the heterogeneity of TG cells and diverse neuronal transcriptomic responses to orofacial inflammation, which aids in the development of novel therapeutics for orofacial inflammatory pain.

Project description:BACKGROUND: Orofacial development is a multifaceted process involving precise, spatio-temporal expression of a panoply of genes. MicroRNAs (miRNAs) constitute the largest family of noncoding RNAs involved in gene silencing, and represent critical regulators of cell and tissue differentiation. MicroRNA gene expression profiling is an effective means of acquiring novel and valuable information regarding the expression and regulation of genes, under the control of miRNA, involved in mammalian orofacial development. RESULTS: To identify differentially expressed miRNAs during mammalian orofacial ontogenesis, miRNA expression profiles from gestation day (GD) -12, -13 and -14 murine orofacial tissue were compared utilizing miRXplore™ microarrays from Miltenyi Biotech GmbH. TaqManTM quantitative Real-Time PCR was utilized for validation of gene expression changes. Cluster analysis of the microarray data was conducted with the clValid R package and the UPGMA (hierarchical) clustering method. Functional relationships between selected miRNAs were investigated using Ingenuity Pathway Analysis. Expression of over 26% of the approximately 588 murine miRNA genes examined was detected in murine orofacial tissues from GD 12, 13 and 14. Among these expressed genes several clusters were seen to be developmentally regulated. Differential expression of genes encoding miRNAs within such clusters were shown to target genes encoding proteins involved in cell proliferation, cell adhesion, differentiation, apoptosis and epithelial-mesenchymal transformation, all processes critical for normal orofacial development. Functional relationships between miRNAs differentially expressed were investigated using Ingenuity Pathway Analysis (IPA; Ingenuity Systems). CONCLUSIONS: Using miRNA microarray technology, unique gene expression signatures of hundreds of miRNAs in embryonic orofacial tissue were defined. Gene targeting and functional analysis revealed that the expression of numerous protein-encoding genes, crucial to normal orofacial ontogeny, may be regulated by specific miRNAs. Project description:Dental pain from apical periodontitis is an infection induced-orofacial pain condition that presents with diversity in pain phenotypes among patients. While 60% of patients with a full-blown disease present with the hallmark symptom of mechanical allodynia, nearly 40% of patients experience no pain. Furthermore, a sexual dichotomy exists, with females exhibiting lower mechanical thresholds under basal and diseased states. We conducted a transcriptomic assessment of periapical biopsies (peripheral diseased tissue) from patients with persistent apical periodontitis. our study found several unique differentially expressed genes (DEGs) between symptomatic and asymptomatic patients, as well as novel candidate genes between sexes within the same pain group.

Project description:Female fish are known to be sensitive to temperature during reproduction, but the long-term consequences on offspring adaptive behaviour and their underlying intergenerational mechanisms remain unknown. We studied the intergenerational consequences of female rainbow trout (Oncorhynchus mykiss) exposure to high (17°C) or normal temperature (12°C) on offspring behavioural phenotypes. We also analysed genome-wide gene expression in eggs and embryos to elucidate the mechanisms by which thermal maternal exposure impacts offspring behaviour. Here we show that a thermal maternal stress induces emotional and cognitive disorders in offspring. Fear responses to a novel environment were inhibited in 17°C offspring indicating global emotional blunting. Thermal stress in mothers also decreased spatial learning abilities in progeny. Behavioural phenotypes were associated with the dysregulation of several genes known to play major roles in neurodevelopment. This is especially true for auts2, a key gene for neurodevelopment in fish and mammals, more specifically neuronal migration and neurite extension, and critical for the acquisition of neurocognitive function in fish and mammals. In addition to auts2, our analysis revealed the dysregulation of another neurodevelopment gene (dpysl5) as well as genes associated with human cognitive disorders (arv1, plp2). Our study also revealed major differences in maternal mRNA abundance in the eggs following maternal exposure to high temperature indicating that some of the observed intergenerational effects are mediated by maternally-inherited mRNAs accumulated in the egg. Together, our observations shed new light on the intergenerational determinism of fish behaviour and associated underlying mechanisms. They also stress the importance of maternal history on fish adaptive capacities in a context of global climate changes.
