Project description:To understand transcriptional aberrations induced by oncogenic Calreticulin (Calr) in myeloproliferative neoplasms we used a cell line – 32D, generated from the murine myeloid precursor cells, to overexpress human thrombopoietin receptor (MPL) together with human Calr. Cells expressing oncogenic Calr show increased JAK/STAT signaling independent of extrinsic stimuli like IL3.

Project description:We established a model for myeloproliferative disease in mice, that is based on primary bone marrow expressing human Thrombopoietin receptor and human mutant Calrdel52. To learn more about cellular and transcriptional rearrangements in the bone marrow we performed single cell RNA sequencing comparing bone marrow cells of empty vector control mice with mice that received MPL-Calrdel52 bone marrow.

Project description:Transcriptional profiling of human MCF7 cells comparing mock control MCF7cells with MCF7cells transfected by siRNA against calreticulin. The latter was shown to be of having significantly decreased expression of calreticulin followed by significant decrease in migrative and invasive potentials of the MCF7 cells. Goal was to determine the effect of calreticulin knockdown on the global gene expression of MCF7 cells. Two-condition experiment, Mock control MCF7 cells vs. calreticulin siRNA knockdown MCF7 cells. Biological replicates: 3 mock control replicates, 3 transfected replicates.

Study the effect of calreticulin knockdown on genome-wide transcript expression in HepG2 cells

Project description:The objective was to evaluate the status of the global hepatic transcriptome in the presence of inhibited calreticulin levels. Results identify a total of 324 genes were altered of which 245 genes were downregulated and 79 genes were upregulated. This suggests that such altered genes might be associated with altered hepatic physiology that are observed during calreticulin deficiency. Total RNA was isolated from HepG2 cells subjected to calreticulin (CRT) siRNA for 24 hours compared to control HepG2 cells subjected to scramble siRNA for 24 hours. This was then hybridized to Illumina HumanHT-12 v4 Expression BeadChip arrays.
