Project description:Aims: Evaluate the genome-wide gene expression and epigenomic alterations caused by transient high glucose (HG) in endothelial cells (ECs), and to assess NRF2 pathway activation as a potential treatment for the diabetes-associated transcriptional memory. Methods and results: We exposed human umbilical vein endothelial cells (HUVEC) to three different glucose treatments: control (5.5 mM for 8 days), HG (30 mM for 8 days) and memory (5.5 mM for 4 days followed by 30 mM for 4 days). After these treatments we conducted a series of experiments, first we found that cells in the HG treatment had sustained alterations in both glycolytic and respiratory rates that persisted in the memory treatment, this without changes in proliferation or cell viability. Similarly, HG provoked substantial changes in the expression of genes belonging to pathways known to be impaired in diabetes, like the TGF-beta, TNF, foxO, p53 and NRF2 pathways, many of which were still present in memory. In addition, we found persistent changes in chromatin accessibility located mainly in non-promoter regions, some of which we identified as putative enhancer regions neighboring genes altered in memory. Finally, we demonstrated that activation of the NRF2 pathway through either NRF2 overexpression or sulforaphane treatment was able to substantially revert the glucose-induced transcriptional memory in ECs. Conclusions: Our results demonstrate that transient HG induces persistent changes in both the transcriptomic and chromatin accessibility profiles of ECs, and that pharmacological NRF2 pathway activation through sulforaphane supplementation is able to prevent and revert the glucose-induced transcriptional memory.

Project description:Nrf2 (NF-E2-related-factor-2) contributes to the maintenance of glucose homeostasis in vivo. Nrf2 suppresses blood glucose levels by protecting pancreatic β-cells from oxidative stress and improving peripheral tissue glucose utilization. To elucidate the molecular mechanisms by which Nrf2 contributes to the maintenance of glucose homeostasis, we generated skeletal muscle (SkM)-specific Keap1-knockout (Keap1MuKO) mice that express abundant Nrf2 in SkM and then examined Nrf2-target gene expression in this tissue. In Keap1MuKO mice, blood glucose levels were significantly downregulated, and the levels of glycogen branching enzyme (Gbe1) mRNA, along with those of glycogen branching enzyme (GBE) protein, were significantly upregulated in mouse SkM. Consistent with this result, chemical Nrf2-inducers promoted Gbe1 mRNA expression in both mouse SkM and C2C12 myotubes. Chromatin-immunoprecipitation analysis demonstrated that Nrf2 binds the Gbe1 upstream promoter regions. In Keap1MuKO mice, muscle glycogen content was strongly reduced, and forced GBE expression in C2C12 myotubes promoted glucose uptake. Therefore, our results demonstrate that Nrf2-induction in SkM increases GBE expression and reduces muscle glycogen content, resulting in improved glucose tolerance. Project description:The transcription factor NF-E2-related factor 2 (Nrf2) induces cytoprotective genes, but has also been linked to the regulation of hepatic energy metabolism. In order to assess the pharmacological potential of hepatic Nrf2 activation in metabolic disease, Nrf2 was activated over 8 weeks in mice on Western diet using two different siRNAs against kelch-like ECH-associated protein 1 (Keap1), the inhibitory protein of Nrf2. Whole genome expression analysis followed by pathway analysis demonstrated that the suppression of Keap1 expression induced genes that are involved in anti-oxidative stress defense and biotransformation, pathways proving the activation of Nrf2 by the siRNAs against Keap1. The expression of neither fatty acid- nor carbohydrate-handling proteins was regulated by the suppression of Keap1. Metabolic profiling of the animals did also not show effects on plasma and hepatic lipids, energy expenditure or glucose tolerance by the activation of Nrf2. The data indicate that hepatic Nrf2 is not a major regulator of intermediary metabolism in mice.
