Project description:The transcriptomic analysis was evaluated using RNA-seq of salmon challenged with the birnavirus IPNv under two different thermal conditions: (a) constant temperature (mean temperature 15 ± 0.9 ºC, “No-Fever”) and (b) temperature gradient (mean temperature 15 ± 7.4 ºC, “Fever”). In parallel, fish in another tank were treated by adding 100 ml virus free cell culture supernatant to the water (mean temperature 15 ± 7.4 ºC, "mock-infected"). We identified a specific transcriptomic signature for each group in the RNA-seq data and identifying a set of key components that control inflammatory modulation during behavioural fever upon viral infection in mobile ectotherms.

Project description:Fifteen adult male Cyprinodon variegatus were exposed individually to 10µg/L of bisphenol A (BPA) for 7 days in 1.8L vessels and other fifteen to 10 µg/L of BPA metabolite, 4-methyl-2,4-bis(4-hydroxyphenyl)pent-1-ene (MBP) under the same conditions. Dimethyl sulfoxide (DMSO) was used as a carrier at 1µL/100mL and was added at the same concentration to the solvent control (n = 15). 90% daily water renewal was done after monitoring the culture conditions, that were: temperature 18.8 ± 0.2°C, pH 7.94±0.03, dissolved oxygen saturation 99.3% ± 1.1 and 16:8 hours of light:dark photoperiod. BPA, MBP and DMSO concentrations were adjusted daily after water renewal.

Project description:Thermotolerant Kluyveromyces marxianus possesses intrinsic abilities to ferment and assimilate a wide variety of substrates including xylose and to efficiently produce proteins. The transcriptome analysis clarified distinctive metabolic pathways under three different growth conditions, static culture, high temperature and xylose medium, in comparison to the control condition of glucose medium under a shaking condition at 30°C. Interestingly, the yeast appears to overcome the issue of reactive oxygen species, which tend to accumulate under all three conditions.

Project description:Due to the increase in stocking density, eutrophication of water quality, and high-temperature conditions aggravate the possibility of hypoxia, and inevitably lead to hypoxic stress, affecting the development of its aquaculture industry. We simulated severe hypoxia stress conditions and constructed two different hypoxia tolerance groups. We used next-generation sequencing to analyze the mRNA expression profiles of the brain and liver under hypoxia to clarify the main pathways and metabolic pathways involved in hypoxia, to systematically reveal the reasons for the differences in hypoxia tolerance among different groups.
