Project description:The length of internodes is critical in determining the height of the castor plant (Ricinus communis L.), and is closely associated with internode elongation. However, the exact mechanisms underlying internode elongation, particularly in the main stem of the castor plant, remain uncertain. To investigate further, we conducted a study using the dwarf castor variety 071113, comparing it with the homologous high-stalk Zhuansihao as a control. Our research included cytological observation, physiological measurement, transcriptome sequencing, and metabolic determination. By integrating these findings, we discovered that the dwarf 071113 undergoes earlier main stem lignification development and has a more active lignin synthesis pathway in internode intermediate development. The plant hormone IAA also plays a role in this process. Furthermore, potential enzymes and regulators have been identified, including the auxin influx carrier AUX1 LAX, auxin response protein IAA13, ARF3, auxin-responsive protein SAUR50, peroxidase, and EXPs that regulate cell cycle, cell wall synthesis, as well as growth and development, were also. Based on these findings, we developed a model for castor internode elongation and gained a better understanding of the dwarfing mechanism of the 071113 variety. Our work lays a theoretical foundation for the future breeding of dwarf castor varieties.

Project description:The toxic protein ricin (also known as RCA60), found in the seed of the castor plant (Ricinus communis) is frequently encountered in law enforcement investigations. The ability to detect ricin by analyzing its proteolytic (tryptic) peptides by liquid chromatography-tandem mass spectrometry (LC-MS/MS) is well established. However, ricin is just one member of a family of proteins in R. communis with closely related amino acid sequences, including R. communis agglutinin I (RCA120) and other ricin-like proteins (RLPs). Inferring the presence of ricin from its constituent peptides requires an understanding of the specificity, or uniqueness to ricin, of each peptide. Here we describe the set of ricin-derived tryptic peptides that can serve to uniquely identify ricin in distinction to closely-related RLPs and to proteins from other species. Other ricin-derived peptide sequences occur only in the castor plant, and still others are shared with unrelated species. We also characterized the occurrence and relative abundance of ricin and related proteins in an assortment of forensically relevant crude castor seed preparations. We find that whereas ricin and RCA120 are abundant in castor seed extracts, other RLPs are not represented by abundant unique peptides. Therefore, the detection of peptides shared between ricin and RLPs (other than RCA120) in crude castor seed extracts most likely reflects the presence of ricin in the sample.

Physiological, Biochemical, and Transcriptomic Analyses Reveal the Response to Oxidative Stress Under Drought Stress in Castor (Ricinus communis L.)

Project description:Castor is an important industrial raw material. Drought leads to delayed development and reduced yield of castor plants. However, the molecular mechanism of the response of castor plants to drought stress has not been determined. In this study, physiological, biochemical, and RNA-seq analyses were conducted on castor roots under PEG-6000 stress for 3, 7, and 4 d (11 d) of rehydration. The activities of GR, APX, SOD, and CAT in the roots of the castor plants under PEG-6000 stress increased at 3 d and decreased at 7 d, while the activity of POD continued to increase. A total of 2926, 1507, and 111 DEGs were identified in castor roots stressed with PEG-6000 for 3 d and 7 d and rehydrated for 4 d (11 d), respectively. GO analysis revealed that antioxidant activity was significantly enriched under PEG stress. In addition, KEGG enrichment analysis revealed that the DEGs were significantly involved in glutathione metabolism, fatty acid metabolism, and plant hormone signal transduction. KEGG combined with WGCNA was used to screen two drought stress-related genes, PP2C39 and GA2ox4. We propose an oxidative stress model for castor plants under drought stress based on GO terms, KEGG pathway, and WGNA analyses. This study identified high-quality genes for the genetic and molecular breeding of drought-resistant castor beans.
