Project description:Adult skeletal muscle regeneration is mainly driven by muscle stem cells (MuSCs), which are highly heterogeneous. Although recent studies have started to characterize the heterogeneity of MuSCs, whether a subset of cells with distinct exists within MuSCs remains unanswered. Here, we found that a population of MuSCs, marked by Gli1 expression, is required for muscle regeneration. The Gli1+ MuSC population displayed advantages in proliferation and differentiation both in vitro and in vivo. Depletion of this population led to delayed muscle regeneration, while transplanted Gli1+ MuSCs supported muscle regeneration more effectively than Gli1- MuSCs. Further analysis revealed that even in the uninjured muscle, Gli1+ MuSCs had elevated mTOR signaling activity, increased cell size and mitochondrial numbers compared to Gli1- MuSCs, indicating Gli1+ MuSCs are displaying the features of primed MuSCs. Moreover, Gli1+ MuSCs greatly contributed to the formation of GAlert cells after muscle injury. Collectively, our findings demonstrate that Gli1+ MuSCs represent a distinct MuSC population which is more active in the homeostatic muscle and enters the cell cycle shortly after injury. This cell population functions as the tissue-resident sentinel that rapidly responds to injury and initiates muscle regeneration.

Project description:Our studies used scRNA-seq analysis to get a full map of transcriptional profiles of enthesis cells and also a seprate Gli1-lineage enthesis stem cells. By harvesting enthesis cells from different development stages for scRNA-seq analysis, we revealed enthesis cell heterogeneity and identified six cell sub-populations using scRNA-seq. We infered cell differentiation trajectories for enthesis stem cells differentiating into mineralizing chondrocytes. A gene regulatory network analysis combined fluorescent in situ hybridization were then used to identify a number of transcription factors coordinating tenogenesis, chondrogenesis, and osteogenesis to form an enthesis with spatially graded mineralization.To further define the enthesis stem cell population, enthesis Gli1-lineage cells were isolated and their transcriptomes were characterized at single cell resolution. These specific Gli1-responsive cells had a linear trajectory and a capacity of chondrogenesis and osteogenesis. The full characterization of transcriptional landscape of tendon enthesis stem cells demonstrates a promising therapeutic strategies using this cell source for enthesis regeneration.

Project description:In the vertebrate neural tube, regional Sonic hedgehog (Shh) signaling invokes a time- and concentration-dependent induction of six different cell populations mediated through Gli transcriptional regulators. Elsewhere in the embryo, Shh/Gli responses invoke different tissue appropriate regulatory programs. To elucidate Shh/Gli regulation of neural fate sepcification, we performed Gli1 ChIP-Seq analysis. We further analyzed two transcription factors whose motifs were enriched in Gli1 ChIP data (Sox2 and Foxa2). Two active histone marks (H3K4me2 and H3K27ac) were additionally analyzed to study activity status of Shh-responsive cis-elements. Active enhancer histone marks and transcription factor binding patterns were obtained from neuralized emrbyoid bodies. Biological replicates were performed for Gli1 and mock FLAG chips. Histone profiling for enhancer marks were taken from time course experiment performed in parallel.

Project description:Traumatic injury often results in muscle loss and impairment, which is worsened under aged and diseased conditions. Activation of resident stem cells or transplantation of myogenic stem cells can promote muscle regeneration. However, major challenges remain in harvesting sufficient autologous myogenic stem cells and expanding such cells efficiently for muscle regeneration therapies. Here, we identified a chemical cocktail that selectively induced a robust expansion of myogenic stem cells from readily-obtainable dermal cells and from muscle stromal cells. By differential plating and lineage tracing, we showed that Pax7+ cells were the major source for chemical-induced myogenic stem cells (CiMCs). We further performed single-cell RNA sequencing (scRNA-seq) analysis to characterize the transcriptomic profile of CiMCs and demonstrate a specific expansion of myogenic cells from heterogeneous dermal cell population. Upon transplantation into the injured muscle, CiMCs were efficiently engrafted and improved functional muscle regeneration in both adult and aged mice. In addition, CiMC transplantation rescued muscle function in mice with Duchenne muscular dystrophy (DMD). Furthermore, an in situ therapeutic modality using this cocktail was developed by loading the chemical cocktail into injectable nanoparticles, which enabled a sustained release of the cocktail in injured muscle and a local expansion of resident satellite cells for muscle regeneration in adult and aged mice. These findings will lead to the development of novel in vitro and in situ stem cell therapies for effective skeletal muscle repair.

Project description:Regeneration of skeletal muscle depends on a population of adult stem cells (satellite cells) that remain quiescent throughout life. Satellite cell regenerative functions decline with aging. We report that geriatric satellite cells, compared to old and adult cells, are incapable of maintaining their normal quiescent state in muscle homeostatic conditions, and this irreversibly affects their intrinsic regenerative and self-renewal capacities. We analyzed the global changes in gene expression occurring within muscle stem cells (satellite cells) in homeostatic conditions during physiological aging. Pure satellite cell populations from dissociated skeletal muscle from Young (2-3 months) and Adult (6 months) mice were isolated using a well-established flow cytometry protocol gating on integrin a7(+)/CD34(+) (positive selection) and Lin- (CD31, CD45, CD11b, Sca1) (negative selection).
