Proteomics characterization of extracellular vesicles derived from fermented food industry by-product: looking for new potential delivery vectors

Project description:In the last decade, the interest in extracellular vesicles (EVs) as safe and biocompatible nano-transporters has risen dramatically, as these nanoparticles can be used to increase the delivery efficiency rate and reduce secondary effects. However, the difficulty for its obtention in large quantities represents a major drawback for its utilization. Also, EVs are currently mainly derived from immortalized cell lines, which may arise safety problems when administered to humans. Hence, the shift from cell lines to safer EVs sources would suppose a step ahead for its use as delivery vectors in the biotechnology and biomedical research fields. Here, we provide proteomics characterization of EVs isolated by molecular weight cut-off filtration (MWCO) from four different fermented food industry by-products (FFIBPs), including: i) yogurt whey (named as milk in the raw data files), symbiotic culture of bacteria and yeast (SCOBY) from kombucha fermented tea (named as Com in the raw data files), wine pomace (named as Wine in the raw data files) and brewer spend yeast (named as beer in the raw data files).This proteomics characterization will provide key information about composition of by-product derived EVs (BP-EV) uncovering a safe and sustainable EVs source with potential as functional EVs delivery vectors.

Project description:The use of high levels of marine fish oil in aquafeeds is a non-sustainable practice. However, more sustainable oils sources from terrestrial plants do not contain long-chain polyunsaturated fatty acids (LC-PUFA). Consequently, feeds based on conventional vegetable oils reduce n-3 LC-PUFA levels in farmed fish. Therefore, the aquaculture industry desperately requires new, sustainable oil sources that contain high levels of n-3 LC-PUFA in order to supply the increasing demand for fish and seafood while maintaining the high nutritional quality of the farmed product. One approach to the renewable supply of n-3 LC-PUFA is metabolic engineering oilseed crops with the capacity to synthesize these essential fatty acids in seeds. In the present study, the oilseed Camelina sativa has been transformed with algal genes encoding the n-3 biosynthetic pathway and expression restricted to the seeds via seed-specific promoters to produce an oil containing > 20% eicosapentaenoic acid (EPA). This oil was investigated as a replacement for marine fish oil in feeds for post-smolt Atlantic salmon. In addition, this study with EPA-rich oil will contribute to our understanding of the biochemical and molecular mechanisms involved in the control and regulation of docosahexaenoic acid (DHA) production from EPA, and will thus better inform our understanding of this key part of the LC-PUFA biosynthetic pathway.

Characterization of Smoke Taint in Hops: Insights from Laboratory Simulation and Chemical Profiling

Project description:The Pacific Northwest (PNW) region contains approximately 97.5% of U.S. commercial hop acreage. In recent years, wildfire events have been increasing in both number and severity within the region. Currently, there is an extensive body of research from the wine industry on the impact of smoke taint in grapes, however, research investigating smoke taint in hops is limited. This study aims to characterize smoke taint in hops through laboratory simulated wildfire smoke exposure coupled with chemical profiling by nontargeted gas chromatography-mass spectrometry (GC-MS). Results reveal that the chemical profiles of smoked hops varied across fuel types. Specifically, several known smoke taint markers, including guaiacol and 4-methylguaiacol, as well as previously unreported compounds such xylopyranose were observed to be elevated in smoked hops compared to controls. This research provides a foundation for future studies to investigate various fuel types and their varying effects on hop quality.
