Project description:This study investigated the systemic effects of 0.1 THz irradiation on the inflammatory status of rheumatoid arthritis (RA) in the collagen-induced arthritis (CIA) mouse model. It was found that THz irradiation apparently alleviated the symptoms of arthritis in CIA mice, reducing the joint swelling, mitigating the tissue damage and relieving the expression of pro-inflammatory factors (TNF-, IL-6, RANKL and MMPs) in the swollen joint of the CIA mice. Moreover, the reduction of serum inflammatory cytokines levels together with the reduced CD4+ T cell count and the recovered percentage of regulatory T cells (Treg) indicated that THz irradiation exerted a systemic immunoregulatory activity in CIA mice. The blood leukocyte mRNA-seq GO analysis also enriched several biological processes including the differentiation and adhesion of leukocytes, lymphocyte differentiation and T cell activation, providing additional supporting evidence for the immunoregulatory effects of THz.

Project description:The molecular basis to autoimmune arthritis is unclear. Collagen Induced Arthritis (CIA) in mice, is a model that has many features that resemble Rheumatoid Arthritis (RA), although it does not perfectly duplicate RA. The study of CIA has provided insight into relevant pathogenesis and has aided in the identification of potential therapeutic targets. In this study we used Mouse Cytokine Expression Arrays to examine gene expression levels in joints at early, peak and decline stages during disease in DBA/1 mice. High power of 0.26 Terahertz radiation induced genes involved in immune system and nervous system changes in the nematode Caenorhabditis Elegans

Project description:The biological impacts of terahertz (THz) waves as well as their biosafety have emerged as a new area of concern with the gradual application of terahertz radiation. In recent years, many studies have been conducted to investigate the influence of terahertz on living organisms, such as animals or cell cultures; however, the biological effects of terahertz waves have not yet been fully revealed. In this study, Caenorhabditis elegans (C. elegans), one of the most acceptable model organisms for studying various forms of ionizing and non-ionizing radiation exposure was used to evaluate the effects of terahertz waves on the entire organism. In this study, the integration of transcriptome sequencing and behavioral tests of C. elegans revealed that high-power THz irradiation damaged the epidermal ultrastructures, inhibited the expression of cuticle collagen gene to impair the movement of C. elegans. Our findings offer fresh perspectives on the biological impacts of high-power THz radiation that could cause epidermal damage and provoke a systemic response.

DNA methylation analysis of synovial fibroblasts from mice undergoing collagen-induced arthritis (CIA) treated, or not, with the the helminth-derived immunomodulator, ES-62

Project description:Aim: Synovial fibroblasts (SFs) undergo a rewiring of their DNA methylation landscape that is associated with their development of an aggressive pathogenic phenotype during Rheumatoid Arthritis (RA), both in humans and in CIA, a mouse model of this chronic inflammatory disease. The helminth-derived immunomodulator, ES-62 acts to prevent joint destruction in CIA by suppressing pathogenic SF responses. We therefore investigated whether ES-62 suppressed the changes in DNA methylation observed in SFs from CIA, relative to those from healthy, mice. Methods: The DNA methylation status of SFs from naïve (no induction of CIA), CIA and ES-62-treated mice undergoing CIA (CIA_ES-62) was compared by Reduced Representation Bilsulphite Sequencing (RRBS) analysis performed by the Active Motif service. Results: Synovial fibroblasts from naive, CIA and CIA_ES-62 mice exhibit differential genomic methylation profiles. Conclusion: ES-62 protection against joint protection in CIA is not associated with prevention of the changes in DNA methylation occurring during rewiring of naive SFs to aggressively pathogenic CIA SFs but rather, reflects generation of a distinct ES-62 phenotype of SF genomic methylation.
