Project description:Oil rapeseed (Brassica napus L.) is a typical winter biennial plant, with high cold tolerance during vegetative stage. In recent years, more and more early-maturing rapeseed varieties were planted across China. Unfortunately, the early-maturing rapeseed varieties with low cold tolerance have higher risk of freeze injury in cold winter and spring. Little is known about the molecular mechanisms for coping with different low-temperature stress conditions in rapeseed. In this study, we investigated 47,328 differentially expressed genes (DEGs) of two early-maturing rapeseed varieties with different cold tolerance treated with cold shock at chilling (4°C) and freezing (−4°C) temperatures, as well as chilling and freezing stress following cold acclimation or control conditions. Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis indicated that two conserved (the primary metabolism and plant hormone signal transduction) and two novel (plant-pathogen interaction pathway and circadian rhythms pathway) signaling pathways were significantly enriched with differentially-expressed transcripts. Our results provided a foundation for understanding the low-temperature stress response mechanisms of rapeseed. We also propose new ideas and candidate genes for genetic improvement of rapeseed tolerance to cold stresses.

Project description:Brassica oleracea and Brassica napus are comprised of diverse cultivars that collectively constitute an important global food source. Of those, the Brassica oleracea convar. acephala cultivar group containing var. sabellica and var. palmifolia and Brassica napus var. pabularia, collectively known as kale, are nutritious leafy greens consumed for their abundance of vitamins and micronutrients. Typified by their curly, serrated or wavy leaves, kale varieties have been primarily defined based on their leaf morphology and geographic origin, despite maintaining complex genetic backgrounds. With changes in the diel molecular environment directly tied to multiple agronomic traits across the food production landscape (e.g. time-of-day nutritional content) and kale representing a candidate crop for vertical farming, we selected nine diverse kale varieties encompassing a wide swath of consumer kale varieties for growth under LED lights using precise real-world dawn/dusk growth conditions followed by quantitative GC-MS metabolomic and LC-MS proteomic analyses. With plant growth and development driven by the day-to-day molecular activities of plants, we harvested kale leaf tissue at end-of-day (ED) and end-of-night (EN) time-points for all metabolomic and proteomic analyses. Our results reveal that kale forms 2 distinct groups, defined by their diel metabolome and proteome signatures primarily involving amino acids and sugars along, with proteome changes in carbon and nitrogen metabolism, mRNA splicing, protein translation and light harvesting. Together, our analysis have derived robust quantitative insights into the diel growth and development landscape of kale, significantly advancing our fundamental understanding of this nutritious leafy green for next-generation breeding and biotechnology.

Project description:C3-C4 intermediate Moricandia suffruticosa showed tolerance to drought and heat stresses, and high photosynthetic capacity under these abiotic stresses as comparing with C3 relative crop rapeseed (Brassica napus). In our study, systematic analysis was conducted to reveal photosynthetic difference between C3-C4 Moricandia suffruticosa and its relative C3 rapeseed from the same Brassiceae tribe. It was found that Moricandia leaf photosynthesis and anatomy were significantly changed compared to rapeseed under drought and heat stress conditions. De novo transcriptome of Moricandia was assembled by next generation sequencing, and unigenes were mapped to respective rapeseed gene locus. Then comparative transcriptome analysis was conducted in leaf tissues of Moricandia and rapeseed under both drought and heat stresses. Main pathways and candidate genes were revealed from this analysis, which may be associated with the stress induced change in Moricandia.

Project description:Maize (Zea mays L.) is one of the major cereal crops worldwide. Increasing planting density is an effective way to improve crop yield. However, plants grown under high-density conditions compete for water, nutrients, and light, which often leads to changes in productivity. To date, few studies have determined the transcriptomic differences in maize leaves in response to different planting densities. This study examined the whole-genome expression patterns in the leaves of maize planted under high and low densities to identify density-regulated genes. Leaves at upper, ear, and lower stem nodes were collected at the grain-filling stage of the maize hybrid Xianyu335 grown under low-density planting and high-density planting. In total, 72, 733, and 1,739 differentially expressed genes (DEGs) were identified in the respective upper, ear, and lower leaves under HDP. Upregulated and downregulated DEGs in the upper and lower leaves were similar in number, whereas upregulated DEGs in the ear leaves were significantly higher in number than the downregulated DEGs. Functional analysis indicated that genes responding to HDP-related stresses were mediated by pathways involving four phytohormones responsible for metabolism and signaling, osmoprotectant biosynthesis, transcription factors, and fatty acid biosynthesis and protein kinases, which suggested that these pathways are affected by the adaptive responses mechanisms underlying the physiological and biochemical responses of the leaves of maize planted at high density.

Project description:Salinity is a major abiotic stress that adversely affects plant growth and development. Canola (Brassica napus L.) is an important oilseed crop in the world, and its yield decreases drastically with increasing salinity. To date, little is known about the molecular mechanisms underlying its salt stress response and tolerance. This study combines physiological assays with comparative proteomics to understand how B. napus plants respond to salt stress. The changes in relative water content, electrical conductance, stomata conductance, intercellular CO2 concentration, transpiration rate, photosynthesis rate, water usage efficiency, respiration rate, chlorophyll fluorescence parameters, antioxidant enzyme activities, soluble sugar, proline and betaine in B. napus plants under 0, 50, 100, 200 and 400 mM NaCl conditions were analyzed. Proteomic profiles of B. napus plants under 100, 200 and 400 mM NaCl treatment at 7 day and 14 day were acquired using iTRAQ LC-MS/MS based quantitative proteomics. A total of 2316 proteins were identified in B. napus leaves, of which 614 proteins showed differential expression under salt stress. These proteins were mainly involved in 10 processes, of which proteins in stress and defense, metabolism and photosynthesis pathways ranked the top three. Subcellular localization analysis showed that more proteins were located in chloroplast, cytoplasm, mitochondria and nucleus. A total of 138 differentially expressed proteins were predicted to interact with each other. These results have provided a comprehensive view of the physiological and molecular processes taken place in B. napus leaves under salt stress, and revealed the molecular mechanisms underlying salt tolerance of B. napus plants.
