Project description:Drought affects avian communities in complex ways. We used our own and citizen science-generated reproductive data acquired through The Cornell Lab of Ornithology's NestWatch Program, combined with drought and vegetation indices obtained from governmental agencies, to determine drought effects on Eastern Bluebird (Sialia sialis L.) reproduction across their North American breeding range for the years 2006-2013. Our results demonstrate that some aspects of bluebird reproductive success vary with the timing and severity of drought. Clutch size was unaffected by any level of drought at the time of clutch initiation or during the 30 to 60 days prior to clutch initiation. Hatching and fledging rates decreased as drought severity increased. Drought conditions occurring at least 30 days prior to the date eggs should have hatched and 60 days prior to the date offspring should have fledged negatively affected reproduction. We also demonstrate the value of datasets generated by citizen scientists in combination with climate data for examining biotic responses at large temporal and spatial scales.

Project description:There is mounting evidence that, across taxa, females breeding in competitive environments tend to allocate more testosterone to their offspring prenatally and these offspring typically have more aggressive and faster-growing phenotypes. To date, no study has determined the mechanisms mediating this maternal effect's influence on offspring phenotype. However, levels of estrogen receptor alpha (ER α) gene expression are linked to differences in early growth and aggression; thus, maternal hormones may alter gene regulation, perhaps via DNA methylation, of ER α in offspring during prenatal development. We performed a pilot study to examine natural variation in testosterone allocation to offspring through egg yolks in wild Eastern Bluebirds (Sialia sialis) in varying breeding densities and percent DNA methylation of CG dinucleotides in the ER α promoter in offspring brain regions associated with growth and behavior. We hypothesized that breeding density would be positively correlated with yolk testosterone, and prenatal exposure to maternal-derived yolk testosterone would be associated with greater offspring growth and decreased ER α promoter methylation. Yolk testosterone concentration was positively correlated with breeding density, nestling growth rate, and percent DNA methylation of one out of five investigated CpG sites (site 3) in the diencephalon ER α promoter, but none in the telencephalon (n = 10). Percent DNA methylation of diencephalon CpG site 3 was positively correlated with growth rate. These data suggest a possible role for epigenetics in mediating the effects of the maternal environment on offspring phenotype. Experimentally examining this mechanism with a larger sample size in future studies may help elucidate a prominent way in which animals respond to their environment. Further, by determining the mechanisms that mediate maternal effects, we can begin to understand the potential for the heritability of these mechanisms and the impact that maternal effects are capable of producing at an evolutionary scale.

Project description:How organisms respond to variation in environmental conditions and whether behavioral responses can mitigate negative consequences on growth, condition, and other fitness measures are critical to our ability to conserve populations in changing environments. Offspring development is affected by environmental conditions and parental care behavior. When adverse environmental conditions are present, parents may alter behaviors to mitigate the impacts of poor environmental conditions on offspring. We determined whether parental behavior (provisioning rates, attentiveness, and nest temperature) varied in relation to environmental conditions (e.g., food availability and ectoparasites) and whether parental behavior mitigated negative consequences of the environment on their offspring in Eastern Bluebirds (Sialia sialis). We found that offspring on territories with lower food availability had higher hematocrit, and when bird blow flies (Protocalliphora spp.) were present, growth rates were reduced. Parents increased provisioning and nest attendance in response to increased food availability but did not alter behavior in response to parasitism by blow flies. While parents altered behavior in response to resource availability, parents were unable to override the direct effects of negative environmental conditions on offspring growth and hematocrit. Our work highlights the importance of the environment on offspring development and suggests that parents may not be able to sufficiently alter behavior to ameliorate challenging environmental conditions.

Project description:The timing and amplitude of reproductive effort are central life history variables for all organisms. In social insects, reproductive effort is collectively controlled at the colony level but little is known about the mechanisms that determine how much colonies invest in reproduction. As part of their female reproductive investment, honey bee colonies raise multiple new queens by feeding royal jelly to female larvae. Artificial selection for commercial royal jelly production in China has generated over the past 40 years a stock of royal jelly bees that raises an order of magnitude more queens and provisions each queen with >3x more royal jelly than unselected stock. Here we establish in a reciprocal cross-fostering experiment that this dramatic shift in social phenotype is due to changes in the nurse bees that care for the brood. We demonstrate higher electrophysiological responsiveness to brood pheromones in royal jelly bees than in unselected bees. Comparing the antennal proteome of unselected and royal jelly bees, we identify proteins involved in chemosensation and energy metabolism as candidates for the observed differences. We confirm several candidates, most prominently OBP16 and CSP4, with quantitative differences of corresponding mRNA levels and functional binding assays between the brood pheromones and the chemosensory proteins. Furthermore, we complement analyses of brood volatiles and electrophysiological recordings with behavioral attraction assays to confirm the presumed biological function of one newly discovered and two existing larval pheromones. Together, these findings help our understanding of pheromonal communication in honey bees and explain how sensory changes in nurse honey bees as alloparental caregivers have evolved in response to artificial selection, leading to a profound shift in colony-level resource allocation to sexual reproduction.

Project description:To study the underlying molecular mechanisms during the Varroa destructor life cycle, we carried out transcriptomic profiling of seven stages: young mites (collected from P8 to P9 brood cells), phoretic mites (collected on adult bees), arresting mites (collected in unsealed L5 brood cells), pre-laying mites (collected from sealed brood cells containing moving larva), laying mites (collected from sealed brood cells containing pre-pupae), post-laying mites (collected from capped brood cells containing purple-eye and white-body pupae P5), emerging mites (collected from P8 to P9 brood cells). In addition, we sampled non-reproducing mites (collected from P5 brood cells, but without offspring), males (collected from P8 to P9 brood cells), and phoretic mites artificially reared in cages with adult bees. This study was performed using Apis mellifera L. honey bee colonies naturally infested by Varroa destructor mites. Adult mites were collected from 4 unrelated colonies.

Project description:Over last decade several studies on oral cancer patients from eastern India have identified alterations in copy numbers in many regions of the chromosome such as 3p21.3, 8q24.21, 9p21, 9p22, 11q13, 11q21-24. However, all these studies employed microsatellite markers to map these CNV regions. This resulted in large map intervals (10-12 megabases) between the adjacent markers studied. As these regions contain a large number of genes, a high resolution CNV map of these regions was necessary to precisely identify novel genes affected by the amplifications and deletions. We thus used custom made Agilent 4X44K oligonucleotide array CGH platform to map the identified CNV regions in a resolution of 3 Kb in oral cancer patients from eastern India. Twenty-nine defined genomic intervals of ten male and two female oral cancer patients were compared with a normal male and normal female sample, respectively, by Agilent's custom oligo CGH array of 4X44K format.

Project description:The hypopharyngeal gland (HG) is the main site of the synthesis and secretion of royal jelly protein (RJP), and shows high plasticity. To identify differentially expressed genes (DEGs) that affect the development of HG and the synthesis and secretion of RJP, we performed a digital gene expression analysis of 9 d Apis mellifera under different conditions of nutrition and exposure to brood pheromone.Six RNA-seq libraries were generated using RNA extracted from 9 d bee HGs. A total of 2801 DEGs were identified on the basis of at least one pairwise comparison, among which 205, 1617 and 2328 genes were differentially expressed in comparisons between the Pollen group and the Honey group, the Brood group and Pollen group and the Brood group and Honey group respectively. The Brood group exhibited the highest number of DEGs, suggesting that brood pheromone plays a key role in the HG ontogeny. A total of 1991 genes were mapped to 129 canonical signaling pathways found in the Kyoto Encyclopedia of Genes and Genomes (KEGG), and the pathways associated with ribosome function and protein processing were significantly enriched.
