Comparative proteomic analysis of the defense response to Gibberella stalk rot in maize

Project description:Fusarium graminearum is the causal agent of Gibberella stalk rot in maize stem, resulting maize lodging, yield, quality, and mechanical harvesting capacity. To date, little is known about the maize stem defense mechanism in response to invasion of F. graminearum. This study represents a global proteomic approach to document the infection by F. graminearum. A total of 1,894 differentially accumulated proteins (DEPs) were identified after inoculation of maize plants with F. graminearum. Functional categorization analysis indicated that proteins involved in plant-pathogen interaction were inducible at early stages of infection. We also found that the expression of proteins involved in phenylpropanoid, flavonoid, and terpenoid biosynthesis were up-regulated in response to F. graminearum infection, which may reflect a particular contribution of secondary metabolism in protection against the fungal attack in maize stem. Together, our results indicated that the defense response of maize stem to F. graminearum infection was multifaceted and involved the induction of proteins from various innate immunity related pathways, which had directive significance for molecular genetic breeding of maize disease-resistant varieties.

To gain insight into these differences, we examined the transcriptome of the five isolates in infected maize roots with that of in vitro (CM agar) growing fungi by a RNA sequencing approach.

Transcription profiling by array of adenylylation of mycobacterial Glnk (PII) protein induced by nitrogen limitation

Project description:PII proteins are pivotal regulators of nitrogen metabolism in most prokaryotes, controlling the activities of many targets, including nitrogen assimilation enzymes, two component regulatory systems and ammonium transport proteins. Escherichia coli contains two PII-like proteins, PII (product of glnB) and GlnK, both of which are uridylylated under nitrogen limitation at a conserved Tyrosine-51 residue by GlnD (a uridylyl transferase). PII-uridylylation in E. coli controls glutamine synthetase (GS) adenylylation by GlnE and mediates the NtrB/C transcriptomic response. Mycobacteria contain only one PII protein (GlnK) which in environmental Actinomycetales is adenylylated by GlnD under nitrogen limitation. However in mycobacteria, neither the type of GlnK (PII) covalent modification nor its precise role under nitrogen limitation is known. In this study, we used LC-Tandem MS to analyse the modification state of mycobacterial GlnK (PII), and demonstrate that during nitrogen limitation GlnK from both non-pathogenic Mycobacterium smegmatis and pathogenic Mycobacterium tuberculosis is adenylylated at the Tyrosine-51 residue; we also show that GlnD is the adenylyl transferase enzyme responsible. Further analysis shows that in contrast to E. coli, GlnK (PII) adenylylation in M. tuberculosis does not regulate GS adenylylation, nor does it mediate the transcriptomic response to nitrogen limitation.
