Project description:Formation of functional pollen and successful fertilisation relies upon the spatial and temporal regulation of anther and pollen development. This process responds to environmental cues to maintain optimal fertility despite climatic changes. Arabidopsis transcription factors bHLH10,-89,-91 were previously thought to be functionally redundant in their control of male reproductive development, however here we show that they play distinct roles in the integration of light signals to maintain pollen development under different environmental conditions. Combinations of the double and triple bHLH10,-89,-91 mutants were analysed under normal (200μmol/m2/s) and low (50μmol/m2/s) light conditions to determine the impact on fertility. Transcriptomic analysis of a new conditionally sterile bhlh89,91 double mutant shows differential regulation of genes related to sexual reproduction, hormone signal transduction and lipid storage and metabolism under low-light. Here we have shown that bHLH89 and bHLH91 play a role in regulating fertility in response to light, suggesting they function in mitigating environmental variation to ensure fertility is maintained under environmental stress.

Single KO show derregulation of multiple molecular pathways and disrupted early spermatogenesys, but retain fertility. Project description:The discovery of photoperiod-sensitive male genic sterile (PGMS) lines started the development of two-line hybrid rice, which is critical to sustain production of high-yielding hybrid rice varieties. Previous reports about the PGMS lines are mainly focused on the gene cloning and their function description. The mechanisms by which PGMS lines perceive changes in photoperiod and transmit those signals to elicit downstream effects is rarely mentioned. In this work, we described the effects of photoperiod on gene expression in wild type (WT) and csa, a PGMS line, flag leaves and anthers to identify putative central factors that transduce photoperiod signals via CSA in both leaves and anthers. We also dissect the potential mechanisms that: cause csa-induced sterility under SD conditions; partially restore fertility in csa anthers under LD conditions; and finally, are responsible to restore full WT fertility under LD and SD conditions. These results provide an overview how the PGMS rice perceives and transmits photoperiod changes, and how the CSA and its homologs response to the environmental cues by regulating down-stream gene expression. That will be helpful to understand the photoperiod-sensitive mechanism and identify new photoperiod-sensitive genes.

Project description:Cannabis is commonly used amongst reproductive age males. Our group and others have shown that chronic delta-9-tetrahydrocannabinol (THC) use adversely impacts male fertility, but less is known about the potential reversibility of these changes. Our study’s objective was to determine if THC discontinuation mitigates THC-associated changes in male reproductive health using a rhesus macaque (RM) model of daily THC edible consumption over a 280-day period (4 spermatogenic cycles). Testicular volume, serum male hormones, semen parameters, sperm DNA fragmentation, seminal fluid proteomics, and whole genome bisulfite sequencing (WGBS) of sperm DNA were assessed at pre-THC, moderate-THC, and heavy-THC dosing, and at 70 and 140 days after THC discontinuation. Chronic THC use resulted in significant testicular atrophy, increased gonadotropins, decreased serum sex steroids, and increased DNA fragmentation. THC discontinuation led to partial recovery of testicular volume, sex steroids and sperm DNA integrity. Seminal fluid proteome analysis revealed differential expression of proteins enriched for processes related to cellular secretion, immune response, and fibrinolysis. WGBS identified significant differential methylation in heavy-THC versus pre-THC sperm, with partial restoration of methylation after discontinuation of THC. Genes associated with differentially methylated regions (DMRs) were enriched for pathways involved in nervous system development and function. This is the first study demonstrating that chronic THC use in RMs adversely impacts male reproductive health, methylation of genes involved in development, and expression of proteins important for male fertility. THC discontinuation improves impacts to male fertility, including partial restoration of THC-associated sperm DMRs in genes important for development.

Project description:Medicinal plants or herbs have been long used in traditional practice and considered as an alternative treatment for diseases. Gynura procumbens is one of the herbs that showed high potential for treating various diseases such as diabetes mellitus, cancer and fertility. Previous study demonstrated G. procumbens potential as anti-hyperglycaemic agent by lowering the blood glucose level of diabetic-induced male rats (Hassan et al. 2010). G. procumbens is claimed to emulate the mechanism of action in insulin, by increasing the glucose intake in the skeletal muscle (Hassan et al. 2010). Another study made by Kamaruzaman & Mat Noor (2017) showed that G. procumbens extract has the ability to improve fertility of diabetes-induced male rats. Diabetes mellitus has caused deleterious effect on male reproductive system due to disruption in spermatogenesis, testicular impairment as well as erectile and ejaculation dysfunction (Alves et al. 2013). Testicular impairment in diabetic rats leads to low sperm quality. Hence, proteomic analysis was performed to identify the differential expressions of total sperm protein after treatment of G. procumbens extract.

Project description:Male sterility is used in the production of hybrid seeds of several crops and can effectively improve the breeding efficiency of cotton hybrids. Reactive oxygen species (ROS) is closely associated with the tapetum and pollen development, but their relationship in cotton male fertility remains unclear. In this study, we comprehensively compared the cytology and proteomics of the anthers from an Upland cotton (Gossypium hirsutum) material, Shida 98 (WT), and its nearly-isogenic male sterile line Shida 98A (MS). Cytological observations indicated delayed PCD in the tapetum and defects in microspore development in MS anthers. Further results revealed that the ROS homeostasis in MS anthers was disrupted. Proteomic results showed that proteins differentially expressed between WT and MS anthers were mainly related to redox homeostasis, protein folding, and apoptotic signaling pathways. HSP70 and APX1 were identified as the two key proteins during cotton anther development. Exogenous application of HSP70 inhibitor increased the level of H2O2 and decreased the activity of APX1 and pollen viability in anthers. Altogether, the proteomics results imply HSP70 and APX1 being the key players jointly regulating tapetal PCD and pollen development by mediating ROS homeostasis. These results provide insights into the proteins associated with male fertility in cotton.
