Project description:Tanshinones and phenolic acids are crucial bioactive compounds biosynthesized in Salvia miltiorrhiza. Methyl jasmonate (MeJA) is an effective elicitor to enhance the production of phenolic acids and tanshinones simultaneously, while yeast extract (YE) is used as a biotic elicitor that only induce tanshinones accumulation. However, little was known about the different molecular mechanism. To identify the downstream and regulatory genes involved in tanshinone and phenolic acid biosynthesis, we conducted comparative transcriptome profiling of S. miltiorrhiza hairy roots treated with either MeJA or YE.Total 55588 unigenes were assembled from about 1.72 billion clean reads, of which 42458 unigenes (76.4%) were successfully annotated. The expression patterns of 19 selected genes in the significantly upregulated unigenes were verified by quantitative real-time PCR. The candidate downstream genes and other cytochrome P450s involved in the late steps of tanshinone and phenolic acid biosynthesis pathways were screened from the RNA-seq dataset based on co-expression pattern analysis with specific biosynthetic genes. Additionally, 375 transcription factors were identified to exhibit a significant up-regulated expression pattern in response to induction. This study can provide us a valuable gene resource for elucidating the molecular mechanism of tanshinones and phenolic acids biosynthesis in hairy roots of S.miltiorrhiza.

Project description:Tanshinones are the major bioactive compounds of Salvia miltiorrhiza Bunge (Danshen), roots, which are used in many therapeutic remedies in Chinese traditional medicine. We investigated the anticancer effects of tanshinones on the highly invasive human lung adenocarcinoma cell line, CL1-5. Tanshinone I significantly inhibited migration, invasion, and gelatinase activity in macrophage-conditioned medium (CM)-stimulated CL1-5 cells in vitro and also reduced the tumorigenesis and metastasis in CL1-5-bearing severe combined immunodeficiency mice. Unlike tanshinone IIA, which induces cell apoptosis, tanshinone I had no significant cytotoxicity. Real-time quantitative polymerase chain reaction (RTQ-PCR), luciferase reporter assay, and an electrophoretic mobility shift assay revealed that tanshinone I reduces the transcriptional activity of interleukin-8 (IL-8), the angiogenic factor involved in cancer metastasis, by attenuating the DNA-binding activity of activator protein-1 and nuclear factor kappaB in CM-stimulated CL1-5 cells. Microarray and pathway analysis of tumor-related genes identified the differentially expressed genes responding to tanshinone I, and these results were validated by RTQ-PCR. The responsive genes included human platelet-derived growth factor beta chain, Shb, H-ras, N-Ras, mitogen-activated protein kinase kinase 3, phosphoinositide-3-kinase, CD44, Rac1, and collagen type IV; these genes may be associated with the Ras MAPK and Rac1 signaling pathways. These results suggest that tanshinone I exhibits anticancer effects both in vitro and in vivo, and that these effects are mediated at least partly through the IL-8, Ras MAPK, and Rac1 signaling pathways. Five human leukemia cell lines were selected for RNA extraction and hybridization on Affymetrix microarrays.To identify genes that are related to Tan IIA sensitivities, we carried out expression profiling on five cell lines.The sample named HL60, MEG01, MOLT,THP1 and U937_control were treated with DMSO. U937 cell line was selected with Tan IIA treatment for 12 h and 24 h, respectively.

Among them, 62 miRNAs express only in root, 95 miRNAs express only in stem, 19 miRNAs express only in leaf, and 71 miRNAs express only in flower, respectively. By the degradome analysis, 69 targets potentially cleaved by 25 miRNAs were identified. Among them, Acetyl-CoA C-acetyltransferase was identified in S. miltiorrhiza, which was cleaved by miR5072 and involved in the biosynthesis of tanshinones. This study provided valuable information for understanding the tissues expression patterns of miRNAs, and offered a foundation for future studies of the miRNA-mediated tanshinones biosynthesis in S. miltiorrhiza. Project description:Trichoderma species promote growth and strengthen immunity of Arabidopsis and crop species through multiple mechanisms. However, how fungal proteins mediate growth-defense tradeoffs is unknown. We analyzed the growth, root architecture, defense and global gene expression profiles in Arabidopsis seedlings co-cultivated with T. atroviride WT, and Δnox1, Δnox2, and ΔnoxR mutants, defective on the catalytic and regulatory subunits of NADPH oxidase, respectively. The gene expression profile in the fungus was also characterized in standard growth conditions and in the presence of plants. The results revealed the critical role of Trichoderma NoxR in mediating growth-defense tradeoffs in Arabidopsis. The effects of T. atroviride WT in improving root branching and biomass production decreased in all three related NADPH defective mutants, particularly in ΔnoxR. In contrast, induction of jasmonic acid-related defense responses in roots and shoots were exacerbated in ΔnoxR compared to the WT strain. Transcriptome analyses showed a tight plant-fungus communication based on reactive oxygen species and availability of carbon resources. The ΔnoxR is unable to perceive changes in nutrient sources and activate signaling cascades, which suppresses the metabolic change from saprophyte to commensal. Thus we conclude that Trichoderma NoxR orchestrates fungal-induced development and defense tradeoffs in Arabidopsis and plays an important role in cross-kingdom plant-fungus communication.
