Project description:In a concentration-response study, we have employed a human-relevant brain microphysiological system (bMPS), containing human-iPSC derived glutamatergic and GABAergic neurons co-cultured with primary human astrocytes, to monitor and detect the changes in the electrical activity of engineered human iPSC-derived neural networks to increasing concentrations of the sarin surrogate, 4-nitrophenylisopropyl methylphosphonate (NIMP). Transcriptomic analysis was conducted at 6-days post exposure to evaluate gene expression changes that could have contributed to how neurons communicated following NIMP exposure.

To develop an improved rodent system for the study of chemical warfare nerve agents, a serum carboxylesterase knock out mouse model was made (Es-1 knock out). This study utilized mRNA analysis to evaluate the effects of knocking out the serum carboxylesterase in a mouse model.

This study utilized microRNA (miRNA) analysis to evaluate potential direct cellular effects of the nerve agent VX (o-ethyl-s-[2 (diisopropylamino) ethyl]) on human (iPSC)-derived neurons and iPSC-derived cardiomyocytes. iPSC-derived cardiomyocytes were treated with VX at concentrations of 0µM (saline control), 0.1µM or 100µM for either 1 hour or 6 hours. Total RNA was then isolated and processed for miRNA microarray analysis using Affymetrix miRNA 2.0 GeneChips
