Project description:Cardiovasculopathy is the leading cause of death; patients with chronic kidney disease (CKD) are at particularly high risk. The pathophysiological role of accumulating reactive metabolites in CKD such as glucose degradation products (GDP) is uncertain. Large amounts of GDP are absorbed from conventional dialysis fluids in patients on chronic peritoneal dialysis (PD). Omental and parietal peritoneal tissues were obtained from 107 pediatric patients with CKD5 and 90 children on PD using dialysis fluids containing low and high concentrations of GDP, respectively. Fat surrounded omental arterioles, protected from local PD fluid exposure, were microdissected for transcriptome and proteome analyses.

Project description:The resident skin microbiota plays a fundamental role in the control of skin physiology and growing evidence support the idea that, at this barrier site, both immunity and inflammatory processes are controlled by skin resident microbiota. However, how defined skin microbes influence the skin immune system under both steady state conditions and inflammatory settings remains poorly understood. Obesity has been linked to increased prevalence of skin inflammatory disorders. We used microarray analysis to investigate how Corynebacterium spp., dominant members of the skin microbiota of mice and humans, influence gene expression in the skin in mice under normal diet or a high-fat diet regimen.

Myocardial adaptations in advanced kidney disease

Project description:Molecular changes underlying the failing heart in chronic kidney disease (CKD) remains largely undefined. To date, a severe paucity of translational studies investigating these changes utilizing human heart tissues from kidney failure patients exists. Moreover, no studies have examined alterations of the cardiac cytoskeleton in CKD, despite its central role in regulating organelle function, bioenergetics and myocardial remodeling. In this 3-arm cross-sectional controlled cohort study, we compared donor heart tissues from dialysis patients, to patients with hypertension with relatively preserved GFR, and healthy controls. We present evidence for transcriptomic changes in the hearts of dialysis patients, involving cytoskeletal dysregulation and dysfunction of mitochondrial bioenergetics.
