Project description:Seven different Solanaceae species, Potato (Solanum tuberosum), Tomato (Solanum lycopersicum), Eggplant (Solanum melongena), Pepper (Capsicum annuum), Tobacco (Nicotiana tabacum), Petunia and Nicotiana benthamiana were subjected to heat stress. Plants were grown for 4-6 weeks at 25 C after which heat stress was initiated by exposing the plants to 35 C for 6, 12, 24, 48 and 96 hours. Control samples were isolated from plants just before initiating the heat stress. RNA was isolated using Qiagen RNeasy. Keywords: Direct comparison

Tomato another proteome - Response mechanisms induced by exposure to high temperature in anthers from thermo-tolerant and thermo-sensitive tomato plants: A proteomic perspective

Project description:The study was aimed to investigate by differential proteomics heat stress response mechanisms in tomato anthers collected from flowers of Solanum lycopersicum cv Saladette (SAL), already annotated as a thermo-tolerant tomato genotype and flowers from Solanum lycopersicum cv M82, reported as a thermo-sensitive genotype. Proteins constitutively present at higher or lower level in the tolerant genotype and/or proteins whose abundance was modulated by growth under high temperature in both genotypes were identified, thus leading to define processes involved in the heat stress response and responsible for efficient reduction of the adverse effects of HS. Results showed differences in the abundance of ninety-six proteins and their functional classification highlighted that heat stress mainly affected metabolic pathways, such as energy metabolism (glycolysis and sucrose degradation), nitrogen assimilation and amino acid biosynthesis and modulated the expression of proteins involved in the folding and degradation machinery and ROS detoxification systems

Project description:The intent of the experiment was to infer, from transcriptome analysis, the occurrence of competent LTR retrotransposons in shoot apical meristems of Solanum lycopersicum. For this, we performed Illumina pair-end RNA-seq in M82 tomato samples of meristems, leaves and flowers. We also harvested meristems in plants subjected to long-term heat. In addition, we performed RNA-seq in meristems, leaves and flowers samples from Solanum pennellii, to aid phylogenetic interpretations.

Drought stress was applied by stopping watering of the plants, control plants were normally watered with nutrient solution. Samples were collected at 0, 1, 3, 5, 7 and 10 days after the first application of the drought stress. RNA was isolated using Qiagen RNeasy. Keywords: Direct comparison
