Project description:Background: Anthocyanins are the most important compounds for nutritional quality and economic values of blood orange. However, there are few reports on the pre-harvest treatment accelerate the accumulation of anthocyanins in postharvest blood orange fruit. Here, we performed a comparative Transcriptome and metabolomics analysis to elucidate the underlying mechanism involved in seasonal drought (SD) treatment during fruit expansion stage on anthocyanin accumulation in postharvest ‘Tarocco’ blood orange fruit. Results: Our results showed that SD treatment slowed down the fruit enlargement and increased the sugar accumulation during fruit development and matured period. Obviously, under SD treatment, the accumulation of anthocyanin in blood orange fruit during postharvest storage was significantly accelerated and markedly higher than that in CK. Meanwhile, the total flavonoids and phenols contents and antioxidant activity in SD treatment fruit were also sensibly increased during postharvest storage. Based on metabolome, we found that substrates required for anthocyanin biosynthesis, such as amino acids and their derivatives, and phenolic acids, have significantly accumulated and higher in SD treated mature fruit compared with that of CK. Further according to the results of transcriptome data and weighted gene coexpression correlation network analysis (WGCNA) analysis, phenylalanine ammonia-lyase (PAL3) was considered key structural gene. qRT-PCR analysis verified that the PAL3 was highly expressed in SD treated postharvest stored fruit and was significantly positively correlated with the anthocyanin content. Moreover, we found that other structural genes in anthocyanin biosynthesis pathway were also upregulated under SD treatment through transcriptome data and qRT-PCR analysis. Conclusions: The findings suggest that SD treatment promotes the accumulation of substrates necessary for anthocyanin biosynthesis during fruit ripening process, and activates the expression of anthocyanin biosynthesis pathway genes during postharvest storage period, especially PAL3, co-contributed to the rapid accumulation of anthocyanin. The present study provides a theoretical basis for postharvest quality control and water-saving utilization of blood orange fruit.

Project description:Organic acids and anthocyanins are the most important compounds for the flavor and nutritional quality of citrus fruit. In this study, a comparative transcriptome analysis was conducted to elucidate the genes and pathways involved in the accumulation of citrate and anthocyanins in postharvest citrus fruit using ‘Tarocco’ blood orange (TBO) and ‘Bingtangcheng’ sweet orange (BTSO). About 2397, 2677, 3067, 3131 and 2960 differentially expressed genes (DEGs) between two materials were identified at 0, 15, 30, 60, and 90 DAS (days after storage) respectively. A robust core set of 825 DEGs temporally associated with the citrate and anthocyanin accumulation throughout the storage period. Further function enrichment revealed the up-regulated DEGs in TBO were mainly related to UV protection, flavonoid biosynthesis process and transcription factors (TFs). And the down-regulated DEGs were mainly related to inositol biosynthesis process, carboxylic acid/organic acid transmembrane transport and transporters. Moreover, co-expression network and correlation analysis revealed that 23 differentially expressed TFs may be responsible for the simultaneously positively regulation of citrate and anthocyanin accumulation. And 19 differentially expressed transporters may be involved in citrate metabolism and negatively correlated with citrate content. These results suggesting that the high anthocyanis and citrate levels in TBO compared with BTSO were porbably attributed to the highly active of a group of core TFs and lowly active of transporters, which will accordingly provide novel insights into the molecular mechanisms underlying the difference accumulation of citrate and anthocyanin content in TBO and BTSO.

Here, we performed a comparative transcriptome analysis to elucidate the genes and pathways involved in both citrate and anthocyanin accumulation in postharvest citrus fruit with 'Tarocco' blood orange (TBO; high accumulation) and 'Bingtangcheng' sweet orange (BTSO; low accumulation).ResultsA robust core set of 825 DEGs were found to be temporally associated with citrate and anthocyanin accumulation throughout the storage period through transcriptome analysis. Further according to the results of weighted gene coexpression correlation network analysis (WGCNA), the turquoise and brown module was highly positively correlated with both of the content of citrate and anthocyanin, and p-type ATPase (PH8), phosphoenolpyruvate carboxylase kinase (PEPCK), chalcone isomerase (CHI), flavanone 3-hydroxylase (F3H), flavonoid 3'-hydroxylase (F3'H) and glutathione S transferase (GST) were considered key structural genes. Moreover, MYB family transcription factor (PH4), Zinc finger PHD-type transcription factor (CHR4, HAC12), Zinc finger SWIM-type transcription factor (FAR1) and Zinc finger C3H1-type transcription factor (ATC3H64) were considered hub genes related to these structural genes. Further qRT-PCR analysis verified that these transcription factors were highly expressed in TBO fruit and their expression profiles were significantly positively correlated with the structural genes of citrate and anthocyanin metabolism as well as the content of citrate and anthocyanin content.ConclusionsThe findings suggest that the CHR4, FAR1, ATC3H64 and HAC12 may be the new transcription regulators participate in controlling the level of citrate and anthocyanin in postharvest TBO fruit in addition to PH4. These results may providing new insight into the regulation mechanism of citrate and anthocyanin accumulation in citrus fruit.

Project description:Soft rot or Rhizopus rot, caused by the fungal pathogen Rhizopus stolonifer, is an aggressive postharvest disease that affects many fruit and vegetables. We proposed that R. stolonifer displays a necrotrophic behavior when infecting fruit, actively killing the host tissues to complete its life cycle. We tested this hypothesis by identifying R. stolonifer infection strategies when interacting with four fruit hosts (tomato, grape, strawberry, and plum). First, we generated a complete and highly contiguous genome assembly for R. stolonifer using PacBio sequencing, of 45.02 Mb in size, an N50 of 2.87Mb, and 12,644 predicted loci with protein-coding genes. We then performed a transcriptomic analysis to identify genes preferentially used by R. stolonifer when growing in fruit versus culture media, and then classified these host-related genes into clusters according to their expression patterns across four time points. Based on the expression data, we determined that R. stolonifer deploys infection mechanisms characteristic of necrotrophs, including a suite of oxidases, proteases, and cell wall degrading enzymes, when it is actively breaking down tissues of all four fruit hosts. Better understanding R. stolonifer – fruit host interactions can support better diagnostic tools and efficient management strategies in postharvest.

Project description:Background: Pear is one of the most important fruit crops worldwide. Anthocyanins and procyanidins (PAs) are important secondary metabolites that affect the appearance and nutritive quality of pear. However, few studies have focused on the molecular mechanism underlying anthocyanin and PA accumulation in pear. Results: We conducted metabolome and transcriptome analyses to identify candidate genes involved in anthocyanin and PA accumulation in young fruits of the pear cultivar ‘Clapp Favorite’ (CF) and its red mutation cultivar ‘Red Clapp Favorite’ (RCF). Gene–metabolite correlation analyses revealed a ‘core set’ of 20 genes that were strongly correlated with 10 anthocyanin and seven PA metabolites. Of these, PcGSTF12 was confirmed to be involved in anthocyanin and PA accumulation by complementation of the tt19-7 Arabidopsis mutant. Interestingly, PcGSTF12 was found to be responsible for the accumulation of procyanidin A3, but not petunidin 3, 5-diglucoside, opposite to the function of AtGSTs in Arabidopsis. Transformation with PcGSTF12 greatly promoted or repressed genes involved in anthocyanin and PA biosynthesis, regulation, and transport. Electrophoretic mobility shift and luciferase reporter assays confirmed positive regulation of PcGSTF12 by PcMYB114. Conclusion: These findings identify a core set of genes for anthocyanin and PA accumulation in pear. Of these, PcGSTF12 was confirmed to be involved in anthocyanin and PA accumulation. Our results also identified an important anthocyanin and PA regulation node comprising two core genes, PcGSTF12 and PcMYB114. These results provide novel insights into anthocyanin and PA accumulation in pear and represent a valuable data set to guide future functional studies and pear breeding.

Understanding the molecular frame underlying the cold storage process will provide a basic guidance for practical control. We used Affymetrix Citrus GeneChip to examine the transcriptional changes in cold-stored Citrus reticulata Blanco cv. âPonkanâ pulp tissue for three successive months. Ponkan postharvest fruits were under cold storage for three successive months. The pulp tissue was used for RNA extraction and hybridization on Affymetrix Citrus Genome microarrays.

Project description:Anthocyanins are specialized plant metabolites with significant dietary value due to their antioxidant and anti-inflammatory properties. Extensive research has indicated that dietary intake of these phenolic compounds contributes to preventing various chronic diseases. Consequently, incorporating anthocyanin-rich foods into one's diet, particularly from natural sources, is highly beneficial. The tomato (Solanum lycopersicum) is the most consumed vegetable worldwide, making it an excellent candidate for anthocyanin-enrichment strategies. The activation of anthocyanin biosynthesis is light-dependent in tomato, but this mechanism has not been entirely characterized. In this study, a purple tomato line in the cv. Micro-Tom (MT-Aft/atv/hp2) was utilized to investigate cyanic fruits developed under varying light conditions. This genotype is derived from natural genetic variation and exhibits anthocyanin accumulation starting early in fruit development. Transcriptional analyses of the fruit peel (exocarp or epicarp) and flesh (mesocarp) revealed that the bHLH transcription factor SlAN1 (Solyc09g065100) is the limiting factor for anthocyanin accumulation in both tissues. In this genotype, the absence of anthocyanin accumulation in the flesh results from the sun-blocking effect of the cyanic epicarp on the mesocarp, preventing light from penetrating deeper into the fruit during its development. This research enhances our comprehension of the genetic and environmental regulation of anthocyanin accumulation in fruit tissues, offering valuable insights for plant breeding and human nutrition.

Project description:We report on the kiwifruit postharvest phase through an approach consisting of 2D-DIGE/nanoLC-ESI-LIT-MS/MS-based proteomic measurements. Kiwifruit samples stored under conventional, cold-based postharvest conditions were sampled at four stages (from fruit harvest to pre-commercialization) and analyzed in comparison protein content. Proteomics showed that proteins associated with disease/defense, energy, protein destination/storage, cell structure and metabolism functions were affected at precise fruit postharvest times. By lining up kiwifruit postharvest processing to a proteomic depiction, this study integrates previous observations on protein content in postharvest pomes treated with specific chemical additives, and provides a reference framework for further studies on the optimization of fruit storage before its commercialization.
